The transient receptor potential vanilloid 1 (TRPV1) is primarily localized to sensory nerve fibers and is associated with the stimulation of pain and inflammation. TRPV1 knockout (TRPV1KO) mice show enhanced LPS-induced sepsis compared with wild type (WT). This implies that TRPV1 may have a key modulatory role in increasing the beneficial and reducing the harmful components in sepsis. We investigated immune and inflammatory mechanisms in a cecal ligation and puncture (CLP) model of sepsis over 24 h. CLP TRPV1KO mice exhibited significant hypothermia, hypotension, and organ dysfunction compared with CLP WT mice. Analysis of the inflammatory responses at the site of initial infection (peritoneal cavity) revealed that CLP TRPV1KO mice exhibited: 1) decreased mononuclear cell integrity associated with apoptosis, 2) decreased macrophage tachykinin NK(1)-dependent phagocytosis, 3) substantially decreased levels of nitrite (indicative of NO) and reactive oxygen species, 4) increased cytokine levels, and 5) decreased bacteria clearance when compared with CLP WT mice. Therefore, TRPV1 deletion is associated with impaired macrophage-associated defense mechanisms. Thus, TRPV1 acts to protect against the damaging impact of sepsis and may influence the transition from local to a systemic inflammatory state.  相似文献

The formation of spore-filled fruiting bodies by myxobacteria is a fascinating case of multicellular self-organization by bacteria. The organization of Myxococcus xanthus into fruiting bodies has long been studied not only as an important example of collective motion of bacteria, but also as a simplified model for developmental morphogenesis. Sporulation within the nascent fruiting body requires signaling between moving cells in order that the rod-shaped self-propelled cells differentiate into spores at the appropriate time. Probing the three-dimensional structure of myxobacteria fruiting bodies has previously presented a challenge due to limitations of different imaging methods. A new technique using Infrared Optical Coherence Tomography (OCT) revealed previously unknown details of the internal structure of M. xanthus fruiting bodies consisting of interconnected pockets of relative high and low spore density regions. To make sense of the experimentally observed structure, modeling and computer simulations were used to test a hypothesized mechanism that could produce high-density pockets of spores. The mechanism consists of self-propelled cells aligning with each other and signaling by end-to-end contact to coordinate the process of differentiation resulting in a pattern of clusters observed in the experiment. The integration of novel OCT experimental techniques with computational simulations can provide new insight into the mechanisms that can give rise to the pattern formation seen in other biological systems such as dictyostelids, social amoeba known to form multicellular aggregates observed as slugs under starvation conditions.  相似文献

Spectacular long-distance migration has evolved repeatedly in animals enabling exploration of resources separated in time and space. In birds, these patterns are largely driven by seasonality, cost of migration, and asymmetries in competition leading most often to leapfrog migration, where northern breeding populations winter furthest to the south. Here, we show that the highly aerial common swift Apus apus, spending the nonbreeding period on the wing, instead exhibits a rarely found chain migration pattern, where the most southern breeding populations in Europe migrate to wintering areas furthest to the south in Africa, whereas the northern populations winter to the north. The swifts concentrated in three major areas in sub-Saharan Africa during the nonbreeding period, with substantial overlap of nearby breeding populations. We found that the southern breeding swifts were larger, raised more young, and arrived to the wintering areas with higher seasonal variation in greenness (Normalized Difference Vegetation Index) earlier than the northern breeding swifts. This unusual chain migration pattern in common swifts is largely driven by differential annual timing and we suggest it evolves by prior occupancy and dominance by size in the breeding quarters and by prior occupancy combined with diffuse competition in the winter.  相似文献

The non-rewarding flowers of Disa spathulata (L.f) Sw. have a highly elongated scent-producing lip that varies in size and form between the two subspecies of this orchid. We investigated whether this unusual morphology is associated with the evolution of a specialized scent-based pollination system. Field observations carried out in South Africa showed that flowers of D. spathulata are pollinated solely by bees belonging to the genus Tetraloniella (Anthophoridae). Flowers of D. spathulata subsp. spathulata were visited by Tetraloniella brevikeraia, while flowers of D. spathulata subsp. tripartita (Lindl.). H. P. Linder were visited by Tetraloniella junodi. The floral fragrance of D. spathulata is emitted by the lip blade and differs markedly between the two subspecies (only 24 [42.1] of the 57 compounds identified were shared). Fragrance of D. spathulata subsp spathulata is dominated by the fatty acid derivatives decyl actetate, octanol, and decanol, while that of D. spathulata subsp. tripartita is dominated by the terpenoids (E) - ocimene, caryophyllene, and (E) nerolidol. Both male and female Tetraloniella bees were strongly attracted to cut flowers of D. spathulata subsp spathulata, even at sites where the orchid does not naturally occur. Bees visit the orchids repeatedly despite the lack of rewards in their flowers. Sexual deception in D. spathulata can be excluded by the attraction of both male and female bees, and yet the manner in which bees are attracted seems too intense and species-specific to be characterized as food-deception. This implies the existence in D. spathulata of a system of scent-based deception of insects that has not been described previously in plants.  相似文献

In combination with the 7th International Aquarium Congress (7th IAC), which was held in 2008 in Shanghai, the authors visited in Japan the aquariums in Tokio, Fukushima, Enoshima, Osaka and Okinawa as well as the National Museum of Marine Biology & Aquarium in Pintung/Taiwan and in China the Beijing Aquarium and the Shanghai Ocean Aquarium, the host of the IAC. The different themes and the specialities of these aquariums are described and compared with the European situation. All the visited aquariums are not older than 20 years. Therefore their aquarium technology as well as their husbandry and presentation of animals are rather modern. Some of the differences in the exhibition are maybe based on the different cultural background or they are a result of the high visitor numbers, because almost all the aquariums are visited by more than one million visitors per year.  相似文献

To study the nature of antibody-antigen interactions, we have determined the variable gene sequences of the anti-cytochrome c immunoglobulin G1 (IgG1) monoclonal antibody E8, and obtained diffraction-quality crystals of the E8 antigen-binding fragment (Fab), both free and bound to its antigen, horse cytochrome c. The FabE8 crystals belong to space group P21 with unit cell dimensions of a = 45.0 A, b = 85.1 A, c = 63.3 A and beta = 105.5 degrees, have one FabE8 molecule per asymmetric unit and diffract to at least 2.1 A resolution. Crystals of the FabE8-cytochrome c complex belong to space group P212121 with unit cell dimensions of a = 84.3 A, b = 73.3 A and c = 94.9 A, accommodate one complex per asymmetric unit and diffract to 2.4 A resolution. In the nucleotide-derived amino acid sequences, the light-chain variable domain (VL) but not the heavy-chain variable domain (VH) of E8 is nearly identical to that of the anti-lysozyme antibody D1.3, differing by only five amino acid residues. Only one of these interacts with lysozyme in the D1.3-lysozyme crystal structure. Six negative and four positive charges in the VH complementarity determining regions of E8 complement four positive and three negative charges in the E8 epitope on cytochrome c. These data suggest that only a subset of the residues in an antibody-protein interface may be critical for binding and that the VH may play a dominant role in antigenic recognition.  相似文献
