The life cycles of many organisms are constrained by the seasonality of resources. This is particularly true for leaf-mining herbivorous insects that use deciduous leaves to fuel growth and reproduction even beyond leaf fall. Our results suggest that an intimate association with bacterial endosymbionts might be their way of coping with nutritional constraints to ensure successful development in an otherwise senescent environment. We show that the phytophagous leaf-mining moth Phyllonorycter blancardella (Lepidoptera) relies on bacterial endosymbionts, most likely Wolbachia, to manipulate the physiology of its host plant resulting in the ‘green-island’ phenotype—photosynthetically active green patches in otherwise senescent leaves—and to increase its fitness. Curing leaf-miners of their symbiotic partner resulted in the absence of green-island formation on leaves, increased compensatory larval feeding and higher insect mortality. Our results suggest that bacteria impact green-island induction through manipulation of cytokinin levels. This is the first time, to our knowledge, that insect bacterial endosymbionts have been associated with plant physiology.  相似文献

Given a choice of habitats in a laboratory experiment, whiting Merlangius merlangus preferentially spent most time over sand, then gravel and least time over a habitat with emergent structures. The introduction of a predator stimulus increased the preference for the sand habitat for large whiting, whereas small whiting had an increased preference for the habitat with emergent structures.  相似文献

Bacillus Calmette–Guérin (BCG), the only approved tuberculosis vaccine, provides only limited protection. Previously, we generated a recombinant derivative (BCG ΔureC::hly), which secretes the pore-forming toxin listeriolysin O (LLO) of Listeria monocytogenes. This vaccine shows superior protection against tuberculosis in preclinical models and is safe in humans. Here we describe two new vaccine strains which express human interleukin-7 (hIL)-7 or hIL-18 in the genetic background of BCG ΔureC::hly to modulate specific T cell immunity. Both strains exhibited an uncompromised in vitro growth pattern, while inducing a proinflammatory cytokine profile in human dendritic cells (DCs). Human DCs harbouring either strain efficiently promoted secretion of IL-2 by autologous T cells in a coculture system, suggesting superior immunogenicity. BALB/c mice vaccinated with BCG ΔureC::hly, BCG ΔureC::hly_hIL7 or BCG ΔureC::hly_hIL18 developed a more robust Th1 response than after vaccination with parental BCG. Both strains provided significantly better protection than BCG in a murine Mycobacterium tuberculosis challenge model but efficacy remained comparable to that afforded by BCG ΔureC::hly. We conclude that expression of hIL-7 or hIL-18 enhanced specific T cell responses but failed to improve protection over BCG ΔureC::hly in mice.  相似文献

Numerous studies have correlated the advancement of lay date in birds with warming climate trends, yet the fitness effects associated with this phenological response have been examined in only a small number of species. Most of these species–primarily insectivorous cavity nesters in Europe–exhibit fitness declines associated with increasing asynchrony with prey. Here, we use 25 years of demographic data, collected from 1986 to 2010, to examine the effects of spring temperature on breeding initiation date, double brooding, and annual fecundity in a Nearctic - Neotropical migratory songbird, the black-throated blue warbler (Setophaga caerulescens). Data were collected from birds breeding at the Hubbard Brook Experimental Forest, New Hampshire, USA, where long-term trends toward warmer springs have been recorded. We found that black-throated blue warblers initiated breeding earlier in warmer springs, that early breeders were more likely to attempt a second brood than those starting later in the season, and that double brooding and lay date were linked to higher annual fecundity. Accordingly, we found selection favored earlier breeding in most years. However, in contrast to studies of several other long-distance migratory species in Europe, this selection pressure was not stronger in warmer springs, indicating that these warblers were able to adjust mean lay date appropriately to substantial inter-annual variation in spring temperature. Our results suggest that this North American migratory songbird might not experience the same fecundity declines as songbirds that are unable to adjust their timing of breeding in pace with spring temperatures.  相似文献

Studies of bird phenology can help elucidate the effects of climate change on wildlife species but observations over broad spatial scales are difficult without a network of observers. Recently, networks of citizen volunteers have begun to report first arrival dates for many migratory species. Potential benefits are substantial (e.g., understanding ecological processes at broad spatial and temporal scales) if known biases of citizen data reporting are identified and addressed. One potential source of bias in bird phenology studies is the tendency for more “first” migratory arrivals to be reported on weekends than on weekdays. We investigated weekend bias in data reporting for five common bird species in North America (Baltimore Oriole, Icterus galbula; Barn Swallow, Hirundo rustica; Chimney Swift, Chaetura pelagica; Purple Martin, Progne subis; and Ruby-throated Hummingbird, Archilochus colubris), and assessed whether this bias affected mean arrival dates reported using data from historical (1880–1969; N?=?25,555) and recent (1997–2010; N?=?63,149) Citizen Science databases. We found a greater percentage of first arrivals reported on weekends and small but significant differences in mean arrival dates (approximately 0.5 days) for four of five species. Comparing time periods, this weekend bias decreased from 33.7 % and five species in the historical time period to 32 % and three species in the recent, perhaps related to changes in human activity patterns. Our results indicate that weekend bias in citizen data reporting is decreasing over time in North America and including a ‘day of week’ term in models examining changes in phenology could help make conclusions more robust.  相似文献

Toll-like receptor (TLR) signaling is triggered by pathogen-associated molecular patterns that mediate well established cytokine-driven pathways, activating NF-κB together with IRF3/IRF7. In addition, TLR3 drives caspase 8-regulated programmed cell death pathways reminiscent of TNF family death receptor signaling. We find that inhibition or elimination of caspase 8 during stimulation of TLR2, TLR3, TLR4, TLR5, or TLR9 results in receptor interacting protein (RIP) 3 kinase-dependent programmed necrosis that occurs through either TIR domain-containing adapter-inducing interferon-β (TRIF) or MyD88 signal transduction. TLR3 or TLR4 directly activates programmed necrosis through a RIP homotypic interaction motif-dependent association of TRIF with RIP3 kinase (also called RIPK3). In fibroblasts, this pathway proceeds independent of RIP1 or its kinase activity, but it remains dependent on mixed lineage kinase domain-like protein (MLKL) downstream of RIP3 kinase. Here, we describe two small molecule RIP3 kinase inhibitors and employ them to demonstrate the common requirement for RIP3 kinase in programmed necrosis induced by RIP1-RIP3, DAI-RIP3, and TRIF-RIP3 complexes. Cell fate decisions following TLR signaling parallel death receptor signaling and rely on caspase 8 to suppress RIP3-dependent programmed necrosis whether initiated directly by a TRIF-RIP3-MLKL pathway or indirectly via TNF activation and the RIP1-RIP3-MLKL necroptosis pathway.  相似文献

Aims and background

Release of ‘non-exchangeable’ potassium (K) from interlayers of illite is diffusion-controlled and has been shown to depend on the solution concentration of K and other cations (Ca2+, Mg2+, NH4 +).

Methods

We analysed changes in soil solution concentrations of K and competing cations in situ at different distances from the root surface over time and related them to the transformation of illite, as revealed by X-ray diffraction, and chemical measures of differently bound K.

Results and Conclusions

Within 49 and 98 days, respectively, 6.4 and 14.4 % of the illite’s total K was released upon contact with the root system. Mixed layered minerals increased from 33 (0 d) to 35 (49 d) to 40 % (98 d). Release of K from interlayers and the transformation of illite occurred at soil solution K concentrations close to the threshold of 80 μM suggested earlier. Concentrations of Ca and Mg increased with decreasing distance from the root surface, promoting the release of K. The NaBPh4 method supposed to determine ‘non-exchangeable’ K extracted only 1/3 of the total K from illite.  相似文献

Background and aims

We studied the response of lignin oxidation in soils of a beech/oak forest to changes in litter fall. Additionally we considered possible factors in lignin oxidation, including altered (i) input of fresh organic matter and (ii) fungi-to-bacteria ratios.

Methods

The field-based experiment included (i) doubling and (ii) exclusion of litter fall and (iii) controls with ambient litter fall. Soil (0–20 cm depth) was sampled after 8 years. We analyzed (i) lignin using the CuO oxidation method, (ii) stocks of free and mineral-bound organic carbon (OC), (iii) the response of soil organic matter (SOM) decomposition to addition of labile organic compounds in laboratory incubations, and (iv) ratios of fungal- vs. bacterial-derived amino sugars (F/B ratios).

Results

Litter exclusion increased stocks of free-light fraction OC, F/B ratios, the ability of the microbial community to use labile compounds for SOM decomposition, as well as acid-to-aldehyde ratios of vanillyl-type lignin phenols in A horizons. Litter addition had no such effects. We assume that litter exclusion caused enhanced transport of organic debris from lower forest floor horizons with rainwater into the A horizon. Enhanced input of organic debris might have increased (i) the availability of labile compounds and (ii) F/B ratios. Consequently, lignin oxidation increased.

Conclusions

Enhanced input of organic debris from forest floors can increase lignin oxidation in mineral topsoils of the studied forest. The expected gradual changes in litter fall due to climate change likely will cause no such effects.  相似文献

To test the hypothesis that competitor density and time spent at a food resource influences aggressive behaviour in male swordtail, Xiphophorus sp., fish were kept at two densities (low: 18 fish m?3; high: 54 fish m?3) and aggressive behaviour was recorded with the time at the food resource used as a covariate. Competitor density is likely to affect the cost:benefit ratio of food defence. When density is low, there should be sufficient food for all individuals and intra‐specific interactions are expected to be rare, while at high densities, increased intra‐specific encounter rates mean that individuals may spend more time defending a resource than utilising it. Food resource defence should occur at intermediate densities. The frequency of aggression, i.e. bites, chases, and display behaviour was significantly positively influenced by time spent at the food resource (Bites: F1,31 = 8.186, P = 0.007; Chases: F1,31 = 6.439, P = 0.016; Displays: F1,31 = 4.435, P = 0.043) suggesting that food resource defence occurred. Competitor density had no effect on food resource defence and minimal effect on the frequency of aggression with only one type of aggressive behaviour, male–male displays, showing a difference between densities (F1,31 = 6.975, P = 0.013). This finding is suggested to be a result of the formation of dominance hierarchies in this species. Aggression from the dominant individual may be directed at only subordinates of the next dominance rank and subordinate behaviour may be restricted by status rather than immediate threat. In such a situation, aggression may be independent of competitor density.  相似文献

The New Zealand International Polar Year–Census of Antarctic Marine Life (NZ IPY-CAML) project added to previous benthic studies in the Ross Sea by extending sampling north from the continental shelf to previously unsampled areas of the shelf break, slope, abyssal plain and seamounts in the region. The aim of the current study is to give first insights into the deep-sea community structure of the Ross Sea focussing on a component of the benthic boundary layer that is macrofaunal crustaceans collected one metre above the seafloor. We assess changes in Ross Sea crustacean community composition from the shelf break (474 m) to the abyss (3,490 m) and compare the Ross Sea crustacean fauna to areas elsewhere in the Southern Ocean. Analyses reveal high relative abundances, suggesting an important role in the food web. Among the peracarid crustaceans, there was a decline in the proportion of amphipods with increasing depth. Three of 15 isopod families (Acanthaspidiidae, Nannoniscidae and Desmosomatidae) were identified to species level and about 72 % of the species were new to science. Isopod diversity in the Ross Sea abyss appears to be comparable to that in the highly speciose Weddell deep sea. Standardised sampling of these crustacean communities allows setting the biodiversity of the Ross Sea into a global context.  相似文献
