Analysis of phytoplankton data from about 1,500 lakes in 20 European countries has revealed that two-thirds of the species that dominate lakes during the summer are dominant right across Europe. Using Canonical Correspondence Analyses, we have examined how both habitat conditions within lakes and environmental factors over broad geographical scales explained the distribution of the 151 most common summer dominant species. The distributions of these species were best explained by water colour and latitude, although alkalinity and total phosphorus also appeared to be important explanatory factors. Contrary to our original hypothesis, summer water temperatures had a negligible impact on the distribution of dominants, although, due to the restricted summer season we examined, only a limited temperature gradient was present in the dataset. Cryptophytes occurred more frequently among dominants in Northern Europe whereas cyanobacteria and dinophytes dominated more in Central and Southern Europe. Our analyses suggest that besides nutrient concentrations, other water chemistry variables, such as alkalinity and the content of humic substances, have at least as important a role in determining the distribution of the dominant phytoplankton species in European lakes.  相似文献

Background

Although the relationship between allergic inflammation and lung carcinogenesis is not clearly defined, several reports suggest an increased incidence of lung cancer in patients with asthma. We aimed at determining the functional impact of allergic inflammation on chemical carcinogenesis in the lungs of mice.

Methods

Balb/c mice received single-dose urethane (1 g/kg at day 0) and two-stage ovalbumin during tumor initiation (sensitization: days -14 and 0; challenge: daily at days 6-12), tumor progression (sensitization: days 70 and 84; challenge: daily at days 90-96), or chronically (sensitization: days -14 and 0; challenge: daily at days 6-12 and thrice weekly thereafter). In addition, interleukin (IL)-5 deficient and wild-type C57BL/6 mice received ten weekly urethane injections. All mice were sacrificed after four months. Primary end-points were number, size, and histology of lung tumors. Secondary end-points were inflammatory cells and mediators in the airspace compartment.

Results

Ovalbumin provoked acute allergic inflammation and chronic remodeling of murine airways, evident by airspace eosinophilia, IL-5 up-regulation, and airspace enlargement. Urethane resulted in formation of atypical alveolar hyperplasias, adenomas, and adenocarcinomas in mouse lungs. Ovalbumin-induced allergic inflammation during tumor initiation, progression, or continuously did not impact the number, size, or histologic distribution of urethane-induced pulmonary neoplastic lesions. In addition, genetic deficiency in IL-5 had no effect on urethane-induced lung tumorigenesis.

Conclusions

Allergic inflammation does not impact chemical-induced carcinogenesis of the airways. These findings suggest that not all types of airway inflammation influence lung carcinogenesis and cast doubt on the idea of a mechanistic link between asthma and lung cancer.  相似文献

Steel factor (SLF, also called KIT-ligand, mast cell growth factor, or stem cell factor) acting through the tyrosine kinase receptor KIT is thought to be indispensable for the early phase of melanocyte development both in vivo and in vitro . In the present study, Kit-independent precursor cells were generated in mice expressing exogenous SLF in their skin keratinocytes and were detected as pigmented spots after administration of Kit function-blocking antibody. We successfully purified these precursor or stem cells as Kit+CD45 cells by flow cytometry. The purified cells showed normal but delayed differentiation into mature melanocytes, indicating the immature nature of Kit-independent precursors. The Kit-independent interfollicular population generated in SLF transgenic mice was suggested to be the counterpart of the follicular melanocyte stem cell based on the Kit-independent nature for their survival.  相似文献

We assessed the impacts of damming on the biological quality of Estonian streams. A total of 24 dammed sites on 22 streams were sampled. Standard samples were taken during two consecutive springs, 2009 and 2010 in three habitat locations: above dam (reservoir), immediately below dam, and at an undisturbed (reference) site nearby. To estimate biological quality, the national multimetric index (based on five pollution‐sensitive and/or general quality metrics) was used. To estimate direct hydromorphological effects on macroinvertebrates, the locally adapted index, Macroinvertebrates in Estonia: Score of Hydromorphology (MESH) was employed. We found a significant decrease in the biological quality of the above‐dam sites compared to the undisturbed sites (probably due to accumulation of fine sediments and lower flow velocity). Of the five ordinary quality indicators tested, only one (ASPT index) was significantly different between the below‐dam sites and the undisturbed sites. However, the MESH indicated significant stress of macroinvertebrates both at the above‐dam and below‐dam sites, even when flow velocity and bottom type at the below‐dam sites was similar to that at the undisturbed sites. A bacterial culture of Nitrosomonas sp. was isolated from a nitrifying biofilm to construct a biosensor for ammonium nitrogen (NH 4 + ?N) measurements in high ammonia wastewaters. The pure culture of microorganisms was immobilized into agarose gel matrix to attain a stable biosensor with a long service life. Biosensors were calibrated using (NH4)2SO4 solution and a steady-state method. Subsequently, several experiments with synthetic and industrial wastewaters were conducted. A linear range up to 20 mg/L of NH 4 + ?N, and sensitivities between 0.030 and 0.036 were gained with biosensors. During 14 days of stable service life of the Nitrosomonas sp. biosensors, variation of the signal was less than 7%. Response times of biosensors were 15 ~ 25 min, while recovery times were up to 25 min. Measurements with high ammonia content synthetic and industrial wastewaters were conducted, and 8.3 and 5.6% over estimation of NH 4 + ?N was gained, respectively, compared with results of Nessler method. In spite of the small overestimation, the biosensor based on a pure culture of Nitrosomonas sp. and calibrated with (NH4)2SO4 is suitable for the analysis of NH 4 + ?N in high ammonia content wastewaters.  相似文献

Aeromonas hydrophila P69.1 (A. hydrophila) was used to construct a semi-specific biosensor to estimate biochemical oxygen demand (BOD) in high fat and grease content wastewaters. A. hydrophila cells were grown in fat containing medium to induce necessary enzymes for transport and degradation of fatty substances. Universal biosensor based on non-specific Pseudomonas fluorescens P75 (P. fluorescens) was used to conduct comparison experiments. Biosensors were calibrated using OECD synthetic wastewater and steady-state method, subsequently several experiments with synthetic and industrial wastewaters were conducted. A linear range up to 45 mg l(-1) BOD(7) was gained using A. hydrophila biosensor, in comparison to 40 mg l(-1) BOD(7) obtained using P. fluorescens biosensors. The lower limit of detection was 5 mg l(-1) BOD(7). Service life of A. hydrophila and P. fluorescens biosensors were 110 and 115 days, respectively. The response time of the biosensors depended on the BOD(7) of measuring solution and was up to 20 min when analyzing different wastewaters. Both biosensors underestimated BOD in meat industry wastewater from 43% up to 71%, but more accurate results could be obtained with A. hydrophila biosensor. Semi-specific A. hydrophila biosensor was able to measure proportion of fat found in wastewater sample, while other refractory compounds remained undetectable to both biosensors.  相似文献
