ABSTRACTAnimal welfare concerns have plagued the professional zoo and aquarium field for decades. Societal differences remain concerning the well-being of animals, but it appears a shift is emerging. Scientific studies of animal welfare have dramatically increased, establishing that many previous concerns were not misguided public empathy or anthropomorphism. As a result, both zoo and aquarium animal welfare policy and science are now at the center of attention within the world’s professional zoos and aquariums. It is now possible to view a future that embraces the well-being of individual captive exotic animals, as well as that of their species, and one in which professional zoos and aquariums are dedicated equally to advancing both. Though the ethics of keeping exotic animals and animals from the wild in captivity are still a contentious subject both outside and even within the profession, this study argues. We argue that this path forward will substantially improve most zoo and aquarium animals' welfare and could significantly reduce societal concerns. If animal welfare science and policy are strongly rooted in compassion and embedded in robust accreditation systems, the basic zoo/aquarium paradigm will move toward a more thoughtful approach to the interface between visitors and animals. Heart rate was recorded on 210 MZ and 174 DZ same sex twin pairs participating in the MacArthur Longitudinal Twin Study (MALTS) at age 14, 20, 24, 36 months and 7 years. Heart rate was monitored in the laboratory at all ages. At ages 14 to 36 months, heart rate was monitored prior to a set of cognitive tasks. At age 7 years heart rate was recorded during a mood-eliciting videotaped presentation. At this age only heart rate monitored during neutral portions of the presentation were used. Mean heart rate declines substantially across this age range, but is similar in boys and girls and for MZ and DZ twins at each age. Heart rate is moderately correlated across all time points suggesting that individual differences in heart rate are relatively stable over this age range. Multivariate genetic and environmental models were fitted to the raw data. In general, genetic factors contribute to the stability of individual differences over time. Shared and non-shared environment factors tended to be occasion specific, with non-shared environment contributing substantially to the individual variation at each age. Shared environment and non-shared environment also contributed a modicum to the stability across time. Thus, individual differences in resting heart rate is a relatively stable, heritable trait from infancy to early childhood. 相似文献

Changes in odor quality discrimination following recovery from olfactory nerve transection 总被引:4,自引:2,他引:2
Following recovery from olfactory nerve transection, animals regain their
ability to discriminate between odors. Odor discrimination is restored
after new neurons establish connections with the olfactory bulb. However,
it is not known if the new connections alter odor quality perception. To
address this question, 20 adult hamsters were first trained to discriminate
between cinnamon and strawberry odors. After reaching criterion (> or =
90% correct response), half of the animals received a bilateral nerve
transection (BTX) and half a surgical sham procedure. Animals were not
tested again until day 40, a point in recovery when connections are
re-established with the bulb. When BTX animals were tested without food
reinforcement, they could not perform the odor discrimination task. Sham
animals, however, could discriminate, demonstrating that the behavioral
response had not been extinguished during the 40 day period. When
reinforcement was resumed, BTX animals were able to discriminate between
cinnamon and strawberry after four test sessions. In addition, their
ability to discriminate between these two familiar odors was no different
than that of BTX and sham animals tested with two novel odors, baby powder
and coffee. These findings suggest that, after recovery from nerve
transection, there are alterations in sensory perception and that
restoration of odor quality discrimination requires that the animal must
again learn to associate individual odor sensations with a behavioral
response.

The shape of the plots of initial reaction rate (ν) versus initial substrate concentration ([S]0) and versus initial concentration of allosteric effector ([F]0) for the model of allosteric enzyme of Monod, Wyman &; Changeux (1965) and for the model of dissociating regulatory enzyme has been analysed by means of the inconstant exponent (q) for substrate or effector concentration, respectively. It has been shown that allosteric interactions in above-mentioned models with “indirect” co-operativity may be manifested not only by the sigmoidal shape of the plot of ν versus [S]0 or ν versus [F]0 (with one point of inflexion) but also by the increase in the magnitude of exponent q in progress of saturation process of the enzyme by the substrate or by the effector in the absence of the sigmoidal shape of these plots. It has been shown also that the plot of ν versus [S]0 has two inflexion points when the parameters have certain definite values. One of these inflexion points (or even both at definite values of the parameters) is hardly discernible. At certain definite values of the parameters two inflexion points may be kinetically manifested by such phenomenon as “negative” co-operativity (q < 1). This is possible if one of the interconvertable enzyme forms exceeds another not only in the affinity to the substrate but also in the value of the rate constant for catalytic breakdown of the enzyme-substrate complex. 相似文献

The problem of whether phloroglucinol is a direct biosynthetic precursor of flavonoids was reinvestigated. Phloroglucinol-2,4,6-14C was found to be incorporated into rutin in Buckwheat (Fagopyrum esculentum) but most of the activity was found in the sugar moiety, the remainder being approximately equally distributed among the A- and B-rings of the aglycone, quercetin. This indicates extensive degradation of the added phloroglucinol prior to its utilization in the biosynthesis of the flavonoid. The hypothesis of a bio-Fries rearrangement of phloroglucinyl cinnamate to a chalcone, and hence to flavonoids, was also eliminated by comparing the efficiency of incorporation of 14C-labelled phloroglucinyl cinnamate and those of labelled phloroglucinol and cinnamic acid. 相似文献

Activation of peroxidase catalytic function of cytochrome c (cyt c) by anionic lipids is associated with destabilization of its tertiary structure. We studied effects of several anionic phospholipids on the protein structure by monitoring (1) Trp59 fluorescence, (2) Fe-S(Met80) absorbance at 695 nm, and (3) EPR of heme nitrosylation. Peroxidase activity was probed using several substrates and protein-derived radicals. Peroxidase activation of cyt c did not require complete protein unfolding or breakage of the Fe-S(Met80) bond. The activation energy of cyt c peroxidase changed in parallel with stability energies of structural regions of the protein probed spectroscopically. Cardiolipin (CL) and phosphatidic acid (PA) were most effective in inducing cyt c peroxidase activity. Phosphatidylserine (PS) and phosphatidylinositol bisphosphate (PIP2) displayed a significant but much weaker capacity to destabilize the protein and induce peroxidase activity. Phosphatidylinositol trisphosphate (PIP3) appeared to be a stronger inducer of cyt c structural changes than PIP2, indicating a role for the negatively charged extra phosphate group. Comparison of cyt c-deficient HeLa cells and mouse embryonic cells with those expressing a full complement of cyt c demonstrated the involvement of cyt c peroxidase activity in selective catalysis of peroxidation of CL, PS, and PI, which corresponded to the potency of these lipids in inducing cyt c's structural destabilization. 相似文献

Background
Electroporation is an established technique for enhancing plasmid delivery to many tissues in vivo, including the skin. We have previously demonstrated efficient delivery of plasmid DNA to the skin utilizing a custom-built four-plate electrode. The experiments described here further evaluate cutaneous plasmid delivery using in vivo electroporation. Plasmid expression levels are compared to those after liposome mediated delivery.Methods
Enhanced electrically-mediated delivery, and less extensively, liposome complexed delivery, of a plasmid encoding the reporter luciferase was tested in rodent skin. Expression kinetics and tissue damage were explored as well as testing in a second rodent model.Results
Experiments confirm that electroporation alone is more effective in enhancing reporter gene expression than plasmid injection alone, plasmid conjugation with liposomes followed by injection, or than the combination of liposomes and electroporation. However, with two time courses of multiple electrically-mediated plasmid deliveries, neither the levels nor duration of transgene expression are significantly increased. Tissue damage may increase following a second treatment, no further damage is observed after a third treatment. When electroporation conditions utilized in a mouse model are tested in thicker rat skin, only higher field strengths or longer pulses were as effective in plasmid delivery.Conclusion
Electroporation enhances reporter plasmid delivery to the skin to a greater extent than the liposome conjugation method tested. Multiple deliveries do not necessarily result in higher or longer term expression. In addition, some impact on tissue integrity with respect to surface damage is observed.
