Species delimitation in Cystoderma and Cystodermella was evaluated based on ITS and LSU rDNA sequences as well as morphological data. Two species of Cystoderma are synonymised with C. carcharias and three species with C. jasonis, distinguishing the synonymised taxa as varieties of these accepted species. Analyses of partial LSU rDNA sequences revealed Cystoderma and Cystodermella as distinct monophyletic genera, with Ripartitella representing a well-supported sister group of the latter. Phaeolepiota aurea represents either an unsupported sister group or member of Cystoderma in the phylogenies based on LSU and ITS sequences rDNA data, respectively. The tribe Cystodermateae sensu Singer did not appear monophyletic according to analyses of LSU sequences. On the basis of these data, the phylogenetic relationships among most of the analyzed genera could not be resolved unequivocally.  相似文献

Background

The main objective of this study was to estimate the effect of supplementation with Saccaromyces cerevisiae (SC) (Yea-Sacc® 1026) on milk production, metabolic parameters and the resumption of ovarian activity in early lactation dairy cows.

Methods

The experiment was conducted during 2005/2006 in a commercial tied-house farm with an average of 200 milking Estonian Holstein Friesian cows. The late pregnant multiparous cows (n = 46) were randomly divided into two groups; one group received 10 g yeast culture from two weeks before to 14 weeks after calving. The groups were fed a total mixed ration with silages and concentrates. Milk recording data and blood samples for plasma metabolites were taken. Resumption of luteal activity was determined using milk progesterone (P4) measurements. Uterine bacteriology and ovarian ultrasonography (US) were performed and body condition scores (BCS) and clinical disease occurrences were recorded. For analysis, the statistical software Stata 9.2 and R were used to compute Cox proportional hazard and linear mixed models.

Results

The average milk production per cow did not differ between the groups (32.7 ± 6.4 vs 30.7 ± 5.3 kg/day in the SC and control groups respectively), but the production of milk fat (P < 0.001) and milk protein (P < 0.001) were higher in the SC group. There was no effect of treatment on BCS. The analysis of energy-related metabolites in early lactation showed no significant differences between the groups. In both groups higher levels of β-hydroxybutyrate (BHB) appeared from days 14 to 28 after parturition and the concentration of non-esterfied fatty acid (NEFA) was higher from days 1–7 post partum (PP). According to US and P4 results, all cows in both groups ovulated during the experimental period. The resumption of ovarian activity (first ovulations) and time required for elimination of bacteria from the uterus did not differ between the groups.

Conclusion

Supplementation with SC had an effect on milk protein and fat production, but did not influence the milk yield. No effects on PP metabolic status, bacterial elimination from the uterus nor the resumption of ovarian activity were found.

Several behavioral assays are currently used for high-throughput neurophenotyping and screening of genetic mutations and psychotropic drugs in zebrafish (Danio rerio). In this protocol, we describe a battery of two assays to characterize anxiety-related behavioral and endocrine phenotypes in adult zebrafish. Here, we detail how to use the 'novel tank' test to assess behavioral indices of anxiety (including reduced exploration, increased freezing behavior and erratic movement), which are quantifiable using manual registration and computer-aided video-tracking analyses. In addition, we describe how to analyze whole-body zebrafish cortisol concentrations that correspond to their behavior in the novel tank test. This protocol is an easy, inexpensive and effective alternative to other methods of measuring stress responses in zebrafish, thus enabling the rapid acquisition and analysis of large amounts of data. As will be shown here, fish anxiety-like behavior can be either attenuated or exaggerated depending on stress or drug exposure, with cortisol levels generally expected to parallel anxiety behaviors. This protocol can be completed over the course of 2 d, with a variable testing duration depending on the number of fish used.  相似文献

Ostracod faunas from 18 gravity offshore cores taken from S-SW of Mersin–Ta?ucu harbour in Turkey (Eastern Mediterranean) at water depths comprised between 285 and 665 m were studied. Thirty-two (32) species have been identified. Argilloecia acuminata s.l. and Polycope cf. tholiformis are the dominant species in the studied area. The fauna corresponds very well to the “Argilloecia acuminata community, C11” from the Pleistocene to Holocene established by Sissingh (Sissingh, W., 1982. Ecostratigraphical outline history of the Late Cenozoic ostracode fauna of the Central and Eastern Mediterranean Basin. Proceedings of the Koninklijke Nederlandse Akademie van Wettenschappen B 85, 299–322), and indicates circalittoral to upper bathyal environment in the Mediterranean. Plasmid transformation of three thermotolerant strains of methylotrophic bacterium Methylophilus spp. was investigated using three different methods. The plasmids could be transformed by electroporation and were stabily maintained in these methylotrophic bacteria. The calcium chloride (competent cell) method was successful only in the case of one of the three strains studied. Protoplast transformation of methylotrophic bacteria was not successful. The antibiotic resistance markers conferred resistance on the transformants, whereas the wild-type bacteria were sensitive to the antibiotics.  相似文献

Daily feeding patterns of tank-reared Atlantic salmon Salmo salar parr under ambient conditions and their relationship with patterns of aggression were investigated during spring and autumn. A single, early afternoon feeding peak was observed during spring but no consistent pattern emerged from the autumn trial (fish did tend to feed in synchrony, even though the population's feeding pattern varied between days). Aggression was highest at first light during both spring and autumn, reducing thereafter throughout the day. Feeding and aggression were out of phase during spring, with the level of feeding increasing as aggression decreased. It is suggested that the high levels of early morning aggression are associated with re-establishment of the social order that breaks down after a period of darkness; in the spring this may have inhibited feeding.  相似文献

We showed that erythropoietin induced rapid glycosylphosphatidylinositol (GPI) hydrolysis and tyrosine phosphorylation of phospholipase C (PLC)-gamma(2) in FDC-P1 cells transfected with the wild-type erythropoietin-receptor. Erythropoietin-induced tyrosine phosphorylation of PLC-gamma(2) was time- and dose-dependent. By using FDC-P1 cells transfected with an erythropoietin receptor devoid of tyrosine residues, we showed that both effects required the tyrosine residues of intracellular domain on the erythropoietin receptor. Erythropoietin-activated PLC-gamma(2) hydrolyzed purified [(3)H]GPI indicating that GPI hydrolysis and PLC-gamma(2) activation under erythropoietin stimulation were correlated. Results obtained on FDC-P1 cells transfected with erythropoietin receptor mutated on tyrosine residues suggest that tyrosines 343, 401, 464, and/or 479 are involved in erythropoietin-induced GPI hydrolysis and tyrosine phosphorylation of PLC-gamma(2), whereas tyrosines 429 and/or 431 seem to be involved in an inhibition of both effects. Thus, our results suggest that erythropoietin regulates GPI hydrolysis via tyrosine phosphorylation of its receptor and PLC-gamma(2) activation.  相似文献

Territorial ecological networks (in US and some other countries known as greenways) are coherent assemblages of areas representing the natural and semi-natural landscape elements that need to be conserved, managed or, where appropriate, enriched or restored in order to ensure the favourable conservation status of the ecosystems, habitats, species and landscapes across their traditional range. An ecologically compensating areas network is a hierarchical system with the following levels: (1) core areas, (2) buffer zones of core areas, (3) corridors and stepping stones, and (4) nature development and/or restoration areas that support resources, habitats and species. Rivers form natural ecological networks and riparian buffer zones of rivers are typical elements of ecological networks. We studied the distribution of Clouded Apollo (Parnassius mnemosyne) and its habitat requirements in Estonia. Seventy-eight percent of all Clouded Apollo observations were recorded in riparian meadows along the banks of rivers with riparian strips consisting of bushes and trees. Detailed study showed that the butterfly is in most cases associated with meadows with a riparian strip of alder. This is the habitat of the food plant (fumeworth—Corydalis solida) of the larvae, the feeding and mating place of adults, and the migration and hiding site for the Clouded Apollo. The population area and number of individuals have been increasing during the last years, and a new growing South-Estonian sub-population of Clouded Apollo has also been discovered in Estonia.  相似文献
