Intracellular porphyrin generation following administration of 5-aminolaevulinic acid has been widely used in photodynamic therapy for a range of malignant and certain non-malignant lesions. However, cellular uptake of 5-aminolaevulinic acid is limited by its hydrophilic nature and improved means of delivery are therefore being sought. Highly branched polymeric drug carriers known as dendrimers are a promising new approach to drug delivery. The aim of this study was to investigate the efficacy of dendrimers conjugated with 5-aminolaevulinic acid for porphyrin production in the transformed PAM 212 keratinocyte cell line and skin explants. Each dendritic derivative incorporated three 5-aminolaevulinic acid residues which were conjugated as esters via methyl or propyl linkers to a central tertiary carbon whose remaining terminal bore an amino, aminobenzyloxycarbonyl or nitro group. In the cell line, all compounds were more efficient at low concentrations compared to equimolar 5-aminolaevulinic acid for porphyrin production, with the most efficient incorporating the longer propyl linker. This compound was also the most lipophilic according to partition coefficient measurements. The intracellular porphyrin fluorescence levels showed good correlation with cellular phototoxicity following light exposure for all the compounds, together with minimal dark toxicity. Our findings indicate that the key factors influencing the efficacy of the dendritic derivatives are lipophilicity and steric hindrance within the dendritic structure which could restrict access to intracellular esterases for liberation of 5-aminolaevulinic acid. These findings should be taken into account in the design of larger dendrimers of 5-aminolaevulinic acid.  相似文献

Background

Accuracy of genomic prediction depends on number of records in the training population, heritability, effective population size, genetic architecture, and relatedness of training and validation populations. Many traits have ordered categories including reproductive performance and susceptibility or resistance to disease. Categorical scores are often recorded because they are easier to obtain than continuous observations. Bayesian linear regression has been extended to the threshold model for genomic prediction. The objective of this study was to quantify reductions in accuracy for ordinal categorical traits relative to continuous traits.

Methods

Efficiency of genomic prediction was evaluated for heritabilities of 0.10, 0.25 or 0.50. Phenotypes were simulated for 2250 purebred animals using 50 QTL selected from actual 50k SNP (single nucleotide polymorphism) genotypes giving a proportion of causal to total loci of.0001. A Bayes C π threshold model simultaneously fitted all 50k markers except those that represented QTL. Estimated SNP effects were utilized to predict genomic breeding values in purebred (n = 239) or multibreed (n = 924) validation populations. Correlations between true and predicted genomic merit in validation populations were used to assess predictive ability.

Results

Accuracies of genomic estimated breeding values ranged from 0.12 to 0.66 for purebred and from 0.04 to 0.53 for multibreed validation populations based on Bayes C π linear model analysis of the simulated underlying variable. Accuracies for ordinal categorical scores analyzed by the Bayes C π threshold model were 20% to 50% lower and ranged from 0.04 to 0.55 for purebred and from 0.01 to 0.44 for multibreed validation populations. Analysis of ordinal categorical scores using a linear model resulted in further reductions in accuracy.

Conclusions

Threshold traits result in markedly lower accuracy than a linear model on the underlying variable. To achieve an accuracy equal or greater than for continuous phenotypes with a training population of 1000 animals, a 2.25 fold increase in training population size was required for categorical scores fitted with the threshold model. The threshold model resulted in higher accuracies than the linear model and its advantage was greatest when training populations were smallest.  相似文献

Isolates of a granulovirus (GV) from the Diamondback Moth, Plutella xylostella, and nucleopolyhedrovirus (NPV) isolates from Galleria mellonella and Autographa californica were characterized by restriction endonuclease analysis of viral DNA. The capacity for these viruses to infect P. xylostella larvae and some other lepidopteran pests of brassica crops (including Heliothis virescens, Crocidolomia binotalis and Mamestra brassicae) was examined in cross-transmission experiments in which the DNA isolated from purified progeny viruses, was compared by restriction endonuclease analysis with DNA from the inoculum viruses. Two P. xylostella GV isolates from Taiwan and China (Px GV-Taiwan and Px GV-China) appeared to be very closely-related on the basis of comparative restriction endonuclease analysis of viral genomic DNA. However, both virus isolates could be distinguished by 1-3 major band differences and by sub-molar band variation when their DNA was analysed following digestion with Eco RI, Bam HI and Hin dIII. Both P. xylostella GV isolates proved to be infectious for P. xylostella larvae but did not appear to infect M. brassicae, C. binotalis or H. virescens larvae. In contrast, a G. mellonella NPV (Gm NPV) isolate was infectious for P. xylostella larvae as well as for larvae of M. brassicae, C. binotalis and H. virescens. The results also confirmed that P. xylostella larvae are susceptible to infection by A. californica NPV. These studies form the basis for further evaluation of Px GV and Gm NPV as potential biological control agents for the Diamondback Moth.  相似文献

Floristic differentiation of the oriental beech (Fagus orientalis Lipsky) forests in Turkey and Bulgaria was investigated and the role of geographical and topographical factors in this differentiation was assessed. After geographical and ecological stratification of the available 922 relevés, 288 remained. Classification, by applying cluster analysis, resulted in seven vegetation units defined by species composition which represent the geographical and ecological variation of Fagus orientalis forests. DCA ordination was applied to these units by passively projecting their chorological structure, as supplementary variables. For more detailed interpretation of vegetation types with similar geographic distribution patterns, PCA was applied by passively projecting the chorological elements, life-forms and topographical factors as supplementary variables. Seven vegetation units representing the geographical and ecological variety of Fagus orientalis forests were described. Four vegetation units represent the core area of Fagus orientalis distribution on the western and middle coast of the Black Sea region (Euxine region); the remaining three types represent the distribution in the eastern Black Sea region (Colchic region), the distribution in western and southern Anatolia under the influence of the Mediterranean climate and the distribution in the transitional zone from the Euxine region to the continental parts of Inner Anatolia, respectively. The four vegetation types in Euxine region reflect the decreasing effect of Black Sea towards Inner Anatolia, as well as altitudinal differences, except the forest type representing forests on calcareous sites. The other three vegetation units represent ravine, lowland to montane and altimontane forests in Euxine region. Fagus orientalis forests could be distinguished by their floristic composition, their chorological elements and life-forms spectra, which reflect a geographical and ecological gradients.  相似文献

Dengue has become endemic in Pakistan with annual recurrence. A sudden increase in the dengue cases was reported from Rawalpindi in 2016, while an outbreak occurred for the first time in Peshawar in 2017. Therefore, a multi-center study was carried out to determine the circulating dengue virus (DENV) serotypes and Chikungunya virus (CHIKV) co-infection in Lahore, Rawalpindi, and Peshawar cities in 2016–18. A hospital-based cross-sectional study was carried out in Lahore and Rawalpindi in 2016–18, while a community-based study was carried out in Peshawar in 2017. The study participants were tested for dengue NS1 antigen using an immunochromatographic device while anti-dengue IgM/IgG antibodies were detected by indirect ELISA. All NS1 positive samples were used for DENV serotyping using multiplex real-time PCR assay. Additionally, dengue samples were tested for CHIKV co-infection using IgM/IgG ELISA. A total of 6291 samples were collected among which 8.11% were NS1 positive while 2.5% were PCR positive. DENV-2 was the most common serotype (75.5%) detected, followed by DENV-1 in 16.1%, DENV-3 in 3.9% and DENV-4 in 0.7% while DENV-1 and DENV-4 concurrent infections were detected in 3.9% samples. DENV-1 was the predominant serotype (62.5%) detected from Lahore and Rawalpindi, while DENV-2 was the only serotype detected from Peshawar. Comorbidities resulted in a significant increase (p-value<0.001) in the duration of hospital stay of the patients. Type 2 diabetes mellitus substantially (p-value = 0.004) contributed to the severity of the disease. Among a total of 590 dengue positive samples, 11.8% were also positive for CHIKV co-infection. Co-circulation of multiple DENV serotypes and CHIKV infection in Pakistan is a worrisome situation demanding the urgent attention of the public health experts to strengthen vector surveillance.  相似文献

Glucose‐6‐phosphate dehydrogenase (G6PD) and 6‐phosphogluconate dehydrogenase (6PGD) play an important function in various biochemical processes as they generate reducing power of the cell. Thus, metabolic reprogramming of reduced nicotinamide adenine dinucleotide phosphate (NADPH) homeostasis is reported to be a vital step in cancer progression as well as in combinational therapeutic approaches. In this study, N‐benzoylindoles 9a‐ ‐ 9d , which form the main framework of many natural indole derivatives such as indomethacin and N‐benzoylindoylbarbituric acid, were synthesized through three easy and effective steps as an in vitro inhibitor effect of G6PD and 6PGD. The N‐benzoylindoles inhibited the enzymatic activity with IC50 in the range of 3.391505 μM for G6PD and 2.19–990 μM for 6PGD.  相似文献

Background

This study was conducted in order to determine the prevalence of asthma and associated risk factors in the adult population of Karachi, Pakistan.

Methods

This multi-stage, cross-sectional survey was conducted from May 2014–August 2015; comprising 1629 adults in 75 randomly selected clusters in Karachi, Pakistan. Definitions included: ‘self-reported asthma’, ‘reversibility in FEV1 and ‘respiratory symptoms and reversibility in FEV1’.

Results

Prevalence of asthma was 1.8% (self-reported) (95% CI: 1.0–2.6), 11.3% (reversibility in FEV1) (95% CI: 9.4–13.3) and 6.6% (symptoms and reversibility in FEV1) (95% CI: 5.1–8.1). Asthmatics were more likely to belong to the age group ≥38?years according to ‘reversibility in FEV1’ and ‘respiratory symptoms and reversibility in FEV1’ (AOR: 1.9, 95% CI: 1.2–3.3) and (AOR: 2.1, 95% CI: 1.1–4.2), respectively. Asthmatics were more likely to report history of allergies (AOR: 1.9, 95% CI: 1.2–2.9) and (AOR: 2.8, 95% CI: 1.7–4.8); and were exposed to environmental tobacco smoke (AOR: 1.6, 95% CI: 1.1–2.5) and (AOR: 1.9, 95% CI: 1.1–3.3) according to ‘reversibility in FEV1’ and ‘respiratory symptoms and reversibility in FEV1’, respectively. Asthmatics were more likely to report pack years of smoking ≥5 (AOR: 2.3, 95% CI: 1.1–4.7) according to ‘respiratory symptoms and reversibility in FEV1’.

Conclusion

This study reports a high prevalence of asthma among Pakistani adults and calls for developing appropriate public health policies for prevention and control of asthma in the country. Further studies should be conducted to determine the national prevalence as well as follow-up studies to identify preventable causes for adult asthma.

Coiled coils are a fundamental emergent motif in proteins found in structural biomaterials, consisting of α-helical secondary structures wrapped in a supercoil. A fundamental question regarding the thermal and mechanical stability of coiled coils in extreme environments is the sequence of events leading to the disassembly of individual oligomers from the universal coiled-coil motifs. To shed light on this phenomenon, here we report atomistic simulations of a trimeric coiled coil in an explicit water solvent and investigate the mechanisms underlying helix unfolding and coil unzipping in the assembly. We employ advanced sampling techniques involving steered molecular dynamics and metadynamics simulations to obtain the free-energy landscapes of single-strand unfolding and unzipping in a three-stranded assembly. Our comparative analysis of the free-energy landscapes of instability pathways shows that coil unzipping is a sequential process involving multiple intermediates. At each intermediate state, one heptad repeat of the coiled coil first unfolds and then unzips due to the loss of contacts with the hydrophobic core. This observation suggests that helix unfolding facilitates the initiation of coiled-coil disassembly, which is confirmed by our 2D metadynamics simulations showing that unzipping of one strand requires less energy in the unfolded state compared with the folded state. Our results explain recent experimental findings and lay the groundwork for studying the hierarchical molecular mechanisms that underpin the thermomechanical stability/instability of coiled coils and similar protein assemblies.  相似文献

Changes in the antioxidant defense system and proline accumulation were examined at different growth stages (vegetative, boot and reproductive) in plants of two hexaploid spring wheat cultivars (S-24, salt tolerant; MH-97, salt sensitive), grown in hydroponics and salinity-affected with 0, 50, 100 and 150 mM of NaCl. Salt stress provoked a marked decline in plant dry mass and ascorbic acid contents, and increased proline, total soluble proteins and H2O2 contents in both wheat cultivars at all growth stages. However, higher accumulation of proline and H2O2 contents was recorded at the vegetative and boot stages, respectively, in both wheat cultivars. Salt stress caused a consistent rise in the activities of some key antioxidant enzymes (CAT, SOD, POD, and APX) at all growth stages only in the salt tolerant cultivar S-24, whereas such pattern of enhanced activities of enzymatic antioxidants in cv. MH-97 was found only at the vegetative stage under saline regimes. Maximum activities of various enzymatic antioxidants were observed at the vegetative stage in both wheat cultivars under varying external salt treatments. The results showed that high salinity tolerance of cv. S-24, as manifested by lower decrease in its dry matter under salt stress, was associated with higher activities of antioxidant enzymes, increased accumulation of proline, and lower levels of H2O2, as compared with cv. MH-97 at all growth stages under saline regimes.  相似文献
