Boraginaceae seeds are particularly rich in Γ -linolenic acid (6,9,12-octadecatrienoic acid, Γ -18:3). In microsomes, the analysis of phosphatidylcholine (PC) molecular species by HPLC led to identification of 15 different molecular species; among them 4 contained Γ -18:3, mostly at position 2 of sn -glycerol. Time courses of acylation and desaturation in PC molecular species were examined when [14C]oleoyl-CoA or [14C]linoleoyl-CoA was provided as substrates to isolated microsomes. With [14C]oleoyl-CoA or [14C]linoleoyl-CoA and in the absence of NADH, 3 main labelled PC molecular species were found: 18:2/[14C]18:1, 16:0/[14C]18:1 and 18:1/[14C]18:1. When NADH was present in the incubation medium, the fatty acids were progressively desaturated by the Δ12- and Δ6-desaturases successively (with [14C]oleoyl-CoA as precursor) or by the Δ6-desaturase alone (with [14C]linoleoyl-CoA as precursor). In both types of experiments, 7 final desaturation products in microsomes were evidenced; among them, 3 contained radioactive Γ -18:3, i.e . 18:2/[14C] Γ -18:3, 18:1/[14C] Γ -18:3 and 16:0/[14C] Γ -18:3. While the Δ12-desaturase had no specificity for position on the glycerol backbone, labelled Γ -linolenic acid was recovered exclusively in the sn -2 position.  相似文献

Mature intact `Bartlett' pear fruit (Pyrus communis L.) were stored under a continuous flow of air or air + 10% CO2 for 4 days at 20°C. Fruit kept under elevated CO2 concentrations exhibited reduced respiration (O2 consumption) and ethylene evolution rates, and remained firmer and greener than fruit stored in air. Protein content, fructose 1,6-bisphosphate levels, and ATP:phosphofructokinase and PPi:phosphofructokinase activities declined, while levels of fructose 6-phosphate and fructose 2,6-bisphosphate increased in fruit exposed to air + 10% CO2. These results are discussed in light of a possible inhibitory effect of CO2 at the site of action of both phosphofructokinases in the glycolytic pathway, which could account, at least in part, for the observed reduction in respiration.  相似文献

A phospholipid transfer protein has been purified 195-fold from an extract of spinach leaves. This protein is capable of transferring phosphatidylcholine, phosphatidylinositol, phosphatidylglycerol and phosphatidylethanolamine from liposomes to mitochondria. In addition to this protein, a minor part of the total activity was associated with a less purified fraction. The pure protein has an isoelectric point of 9.0 +/- 0.2 determined by a chromatofocusing technique. Electrophoresis on sodium dodecyl sulfate/polyacrylamide gel showed that the protein is homogeneous and has an apparent molecular weight of 9000 +/- 1000, in agreement with the value (8832) calculated from the amino acid composition. This composition is characterized by a high amount of alanine and glycine and by the absence of phenylalanine, whereas arginine, glutamine, histidine and methionine are minor components. The spinach protein is also able to transfer phosphatidylcholine and phosphatidylglycerol from liposomes to intact chloroplasts. This observation reinforces the hypothesis that plastid phospholipids are partly imported from outside the organelle by a transfer process.  相似文献

Cytosol proteins prepared from castor bean endosperm (4-day-old) seedlings stimulate the exchange of [3H]phosphatidylethanolamine between liposomes and mitochondria. The acceleration of the exchange depends on the quantity of cytosol proteins, the time of incubation, and the respective amounts of liposomes and mitochondria. On a per seedling basis, the active proteins are essentially located in the endosperm, whereas the roots and the cotyledons are less rich in these proteins.  相似文献

We have isolated a novel cDNA coding for maize phospholipid-transfer protein. The cDNA sequence is similar to the first one obtained by Tchang et al. [J. Biol. Chem. 263 (1988) 16849-16855] differing only by a mslal number of nucleotide substitutions and insertions. One of these insertions is 74 bp long and is flanked by consensus intron splicing sequences. The protein coded by the two cDNA has identical amino acids except in the C terminus. This difference derived from the presence of the 74-bp insert. The possible existence of an alternative splicing mechanism that could introduce heterogeneity in the sequence of these proteins is proposed.  相似文献

14C-labeled microsomes were prepared by feeding [1-14 C]acetateto endosperm tissues from 4-day-old seedlings of castor beanseeds and incubated with unlabeled mitochondria from the sametissues. The loss of 14C-lipids from the microsomes was accompaniedby an increase of 14C-lipids in the mitochondria. The additionof 105,000?g supernatant and also pH 5.1-treated supernatant,both of which had been prepared from castor bean endospermsat the same stage, markedly enhanced the lipid transfer frommicrosomes to mitochondria. The activity in this fraction wasprecipitated by ammonium sulfate and lost with trypsin or heattreatment. The transfer of lipids was limited to phospholipids.Thus, it is concluded that in castor bean endosperms, phospholipidsare transferred from the endoplasmic reticulum to the mitochondriaby a phospholipid-exchange protein contained in the cytosol. The substrate stereospecificity of NADH-ferricyanide reductase activities in the inner mitochondrial membrane and peroxisomal membrane of potato (Solanum tuberosum L.) tubers, spinach (Spinacea oleracea L.) leaf plasma membrane, and red beetroot (Beta vulgaris L.) tonoplast were all specific for the [beta]-hydrogen of NADH, whereas the reductases in wheat root (Triticum aestivum L.) endoplasmic reticulum and potato tuber outer mitochondrial membrane were both [alpha]-hydrogen specific. In all isolated membrane fractions one or several polypeptides with an apparent size of 45 to 55 kD cross-reacted with antibodies raised against a microsomal NADH-ferricyanide reductase on western blots.  相似文献

Previous studies of the rate constants for the elementary steps of ATP hydrolysis by the soluble and membrane-bound forms of beef heart mitochondrial F1 supported the proposal that ATP is formed in high-affinity catalytic sites of the enzyme with little or no change in free energy and that the major requirement for energy in oxidative phosphorylation is for the release of product ATP.The affinity of the membrane-bound enzyme for ATP during NADH oxidation was calculated from the ratio of the rate constants for the forward binding step (k +1) and the reverse dissociation step (k –1).k –1 was accelerated several orders of magnitude by NADH oxidation. In the presence of NADH and ADP an additional enhancement ofk –1 was observed. These energy-dependent dissociations of ATP were sensitive to the uncoupler FCCP.k +1 was affected little by NADH oxidation. The dissociation constant (K d ATP) increased many orders of magnitude during the transition from nonenergized to energized states.  相似文献
