Removal and recovery of molybdate from aqueous solution was investigated using ZnCl2 activated carbon developed from coir pith. Studies were conducted to delineate the effects of contact time, adsorbent dose, molybdate concentration, pH and temperature. Two theoretical adsorption isotherms, namely, Langmuir and Freundlich were used to describe the experimental results. The Langmuir adsorption capacity (Q0) was found to be 18.9 mg molybdate/g of the adsorbent. Adsorption followed second order kinetics. Studies were performed at different pH values to find out the pH at which maximum adsorption occurred. The pH effect and desorption studies showed that ion exchange and chemisorption mechanism were involved in the adsorption process. Thermodynamic parameters such as DeltaG0, DeltaH0 and DeltaS0 for the adsorption were evaluated. Effect of foreign ions on adsorption of molybdate has been examined. The results showed that ZnCl2 activated coir pith carbon was effective for the removal and recovery of molybdate from water.  相似文献

Carrageenophyte farming is an expanding economical activity in North Borneo Island, Malaysia. During routine monitoring of “ice-ice” disease and epiphyte outbreak at commercial farms, it was apparent that culture lines were heavily (60–80%) infested with biofoulers, particularly Acanthophora spp. and Laurencia majuscula. However, only L. majuscula showed dominance and flourished even during “ice-ice” disease outbreak. Presence of chemical defense against seaweed pathogens was investigated in two populations of L. majuscula collected from three major carrageenophyte farms in two districts; (A) Lohok Butun, Selakan Island, and Bum-Bum Island, in Semporna district, and (B) Telutuh, Carrington Reef, and Balambangan Island, in Kudat district. The first population contained elatol (1), and iso-obtusol (2), and, second population contained (Z)-10,15-dibromo-9-hydroxy-chamigra-1,3(15),7(14)-triene (3) and (E)-10-15-dibromo-9-hydroxy-chamigra-1,3(15),7(14)-triene (4), as their antibacterial metabolites. All four metabolites showed highly selective inhibition against “ice-ice” disease bacteria compared to human pathogens at 30 μg disk−1. In addition, seasonal variation of these compounds at two representative farms (Selakan Island [P-1] and Balambangan Island [P-2]) revealed a 120–170% increase in concentration during “ice-ice” disease outbreak. Microscopy of fresh specimens showed the presence of corps en cerise, which is the synthesis and storage site of halogenated metabolites at superficial cortical cells, branch tips, and trichoblasts. This suggests the importance of these metabolites as defense chemicals against “ice-ice” disease bacteria in L. majuscula that grows on seaweed culture lines.  相似文献

Background

The present study focuses on identifying and developing an anti-diabetic molecule from plant sources that would effectively combat insulin resistance through proper channeling of glucose metabolism involving glucose transport and storage.

Methods

Insulin-stimulated glucose uptake formed the basis for isolation of a bioactive molecule through column chromatography followed by its characterization using NMR and mass spectroscopic analysis. Mechanism of glucose transport and storage was evaluated based on the expression profiling of signaling molecules involved in the process.

Results

The study reports (i) the isolation of a bioactive compound 3β-taraxerol from the ethyl acetate extract (EAE) of the leaves of Mangifera indica (ii) the bioactive compound exhibited insulin-stimulated glucose uptake through translocation and activation of the glucose transporter (GLUT4) in an IRTK and PI3K dependent fashion. (iii) the fate of glucose following insulin-stimulated glucose uptake was ascertained through glycogen synthesis assay that involved the activation of PKB and suppression of GSK3β.

General significance

This study demonstrates the dual activity of 3β-taraxerol and the ethyl acetate extract of Mangifera indica as a glucose transport activator and stimulator of glycogen synthesis. 3β-taraxerol can be validated as a potent candidate for managing the hyperglycemic state.  相似文献

Introduction

Treatment of chondral injuries remains a major issue despite the many advances made in cartilage repair techniques. Although it has been postulated that the use of marrow stimulation in combination with cell-based therapy may provide superior outcome, this has yet to be demonstrated. A pilot study was thus conducted to determine if bone marrow derived mesenchymal stromal cells (BM-MSCs) have modulatory effects on the repair outcomes of bone marrow stimulation (BMS) techniques.

Methods

Two full-thickness chondral 5 mm diameter defects were created in tandem on the medial condyle of left stifle joints of 18 Boer caprine (N = 18). Goats were then divided equally into three groups. Simultaneously, bone marrow aspirates were taken from the iliac crests from the goats in Group 1 and were sent for BM-MSC isolation and expansion in vitro. Six weeks later, BMS surgery, which involves subchondral drilling at the defect sites, was performed. After two weeks, the knees in Group 1 were given autologous intra-articular BM-MSCs (N = 6). In Group 2, although BMS was performed there were no supplementations provided. In Group 3, no intervention was administered. The caprines were sacrificed after six months. Repairs were evaluated using macroscopic assessment through the International Cartilage Repair Society (ICRS) scoring, histologic grading by O’Driscoll score, biochemical assays for glycosaminoglycans (GAGs) and gene expressions for aggrecan, collagen II and Sox9.

Results

Histological and immunohistochemical analyses demonstrated hyaline-like cartilage regeneration in the transplanted sites particularly in Group 1. In contrast, tissues in Groups 2 and 3 demonstrated mainly fibrocartilage. The highest ICRS and O’Driscoll scorings was also observed in Group 1, while the lowest score was seen in Group 3. Similarly, the total GAG/total protein as well as chondrogenic gene levels were expressed in the same order, that is highest in Group 1 while the lowest in Group three. Significant differences between these 3 groups were observed (P <0.05).

Conclusions

This study suggests that supplementing intra-articular injections of BM-MSCs following BMS knee surgery provides superior cartilage repair outcomes.  相似文献

Twelve isolates of Lasiodiplodia theobromae were collected from 12 different commercial banana cultivars belonging to various banana growing regions of Tamil Nadu, India. The results revealed that most of the L. theobromae isolates exhibited significant differences in morphology, colour and spore size. However, all 12 isolates took 3–4 days invariably to cover the 90 mm Petri plates. Among the 12 isolates, the isolates LT3a from Robusta and LT10c from dwarf Cavendish produced abundant pycnidia. These isolates were also analysed through Random amplified polymorphic DNA (RAPD) markers for genetic diversity. Polymerase chain reaction amplification of total genomic DNA of the 12 isolates of L. theobromae with 14 random primers generated numerous polymorphisms among the isolates, while many of the intense bands were shared among majority of the isolates. Cluster analysis also indicated a high degree of genetic variability within L. theobromae isolates from different banana cultivars. The 12 isolates were separated into three clusters namely A, B and C. The maximum similarity index of 80% was recorded between the isolates LT7d from Karpuravalli and LT10c from dwarf Cavendish. The least similarity index of 29.41% was recorded with the isolates LT1f isolated from Poovan and LT5a isolated from Virupakshi. Within the species of L. theobromae the genetic variability was high and it underlines and validates existence of significant intra-specific diversity in isolates of L. theobromae infecting different banana cultivars.  相似文献

This study was carried out to screen Phaseolus vulgaris L. germplasm accessions for anthracnose resistance genes to the fungus Colletotrichum lindemuthianum. (Sacc. and Magn.) Scrib. This fungus is made up of many pathogenic races which poses a challenge in developing resistant plant varieties; however, screening for and selection of resistant plant sources plays an important role in developing resistant plant lines. This screening work involved examining 69 accessions consisting of two resistant lines (D-line, L-line) and two susceptible varieties (Kanchana and Jwala). Fourteen SCAR primers specific to anthracnose disease resistance in the French bean were used. Of these 14 SCAR primers, 5 of them, SAS13, SF10, SC08, SZ04 and SBB14, produced amplification with good monomorphic bands.  相似文献

The barnacle exhibits a high degree of control over its attachment onto different types of solid surface. The structure and composition of barnacle cement have been reported previously, but mostly for barnacles growing on low surface energy materials. This article focuses on the strategies used by barnacles when they attach to engineering materials such as polymethylmethacrylate (PMMA), titanium (Ti) and stainless steel 316L (SS316L). Adhesion to these substrata is compared in terms of morphological structure, thickness and functional groups of the primary cement, the molting cycle and the nanomechanical properties of the cement. Structural characterization studies using scanning electron microscopy (SEM) and transmission electron microscopy (TEM) in conjunction with nanomechanical characterization and infrared spectroscopy (FTIR) are used to understand the differences in the adhesion of primary barnacle cement to the different substrata. The results provide new insights into understanding the mechanisms at work across the barnacle–substratum interface.  相似文献

The widespread use of antimicrobials has increased the occurrence of multidrug resistant microbes. The commonest mechanism of antimicrobial resistance in Enterobacteriaceae is production of β‐lactamases such as metallo‐β‐lactamases (MBL) and extended spectrum β‐lactamases (ESBL). Few studies have used a molecular approach to characterize the prevalence of β‐lactamases. Here, the prevalence of different β‐lactamases was characterized by performing three multiplex PCRs targeting genes similar to those described in earlier publications. Antimicrobial susceptibility tests for all isolates were performed using the agar dilution method. β‐lactamase was detected in 72% of the isolates, the detection rate being 64% in 2011 and 75% in 2012. The isolates were highly resistant to carbapenems such as meropenem and imipenem and susceptible to colistin and tigecycline. In this study, 22% of isolates contained both MBL and ESBL. ESBL was detected more frequently in Escherichia coli isolates, whereas carbapenemase was detected more frequently in Klebsiella pneumoniae isolates. These findings suggest the spread of multi‐resistant ESBL and MBL producers in the community. Our results have implications for patient treatment and also indicate the need for increased surveillance and molecular characterization of isolates.  相似文献

Abstract

The PfHslUV, a Plasmodium falciparum homolog of prokaryotic HslUV systems, is a newly identified drug target. The HslUV complex is an assembly of Heat Shock Locus gene products U and V. The formation of complete complex is essential for the proteasome to carry out its biochemical and physiological role in the parasite, namely to degrade specific target proteins in an ATP-dependent chaperone assisted manner. PfHslV subunit, a protease, exhibits increased proteolytic activity in the presence of PfHslU, the subunit believed to be responsible for allosteric activation of PfHslV. In the present work, we have employed computational methods to simulate the interaction of PfHslU and PfHslV subunits. We have used three methods—namely homology modeling, molecular docking and computational alanine scanning to model the complex, to predict the binding mode of PfHslU-V interaction and to predict the binding-energy hot-spots in protein-protein interface, respectively. The three dimensional models of PfHslV and PfHslU have been generated using MODELLER, based on the crystal structures of prokaryotic HslUV complex as templates. The modeled structures were docked using PatchDock, a geometry-based molecular docking algorithm. Finally, a three-dimensional PfHslUV complex model was generated that helped in comparing protein-protein interface characteristics with that of crystal structures of prokaryotic HslUV. Further, computational alanine scanning analysis of the generated complex was performed to calculate the binding free energy changes (ΔΔGbind), which helped in identifying residues crucial for PfHslU and PfHslV interactions.  相似文献

We have identified a novel 7-azaindole series of anaplastic lymphoma kinase (ALK) inhibitors. Compounds 7b, 7m and 7n demonstrate excellent potencies in biochemical and cellular assays. X-ray crystal structure of one of the compounds (7k) revealed a unique binding mode with the benzyl group occupying the back pocket, explaining its potency towards ALK and selectivity over tested kinases particularly Aurora-A. This binding mode is in contrast to that of known ALK inhibitors such as Crizotinib and NVP-TAE684 which occupy the ribose binding pocket, close to DFG motif.  相似文献
