A comparison of the benthic fauna found in two shallow lakes in the New Jersey Pinelands (USA) illustrated the impact of elevated pH and nutrients caused by residential and agricultural disturbance on a naturally acidic, poorly buffered aquatic environment. Detailed community analysis suggested that change in community composition was a better indicator of response to disturbance than biological diversity indices. Chironomidae (insecta) were the predominant components of the benthic macroinvertebrate assemblage of both undisturbed Oswego Lake (low pH, low nutrients) and Nescochague Lake (fluctuating pH, elevated nutrients). The genera Procladius, Tribelos, and Pagastiella dominated Oswego Lake, where as Zalutschia zalutschicola, Procladius, Dicrotendipes, and Tanytarsus dominated Nescochague Lake. Glyptotendipes was a common and unique member of the Nescochague Lake assemblage. Cluster analysis indicated that the chemical differences between lakes were the most important community determinants, although, within each lake, depth and substrate affected the local communities. Oswego Lake exhibited a depauperate nonchironomid benthic fauna typical of low nutrient, acid lakes. In turn, Nescochague Lake exhibited an enriched nonchironomid fauna including mollusks and planaria which were not found in Oswego Lake. 相似文献

Testes of the palaemonid shrimp Macrobrachium rosenbergii were prepared for study in the light, scanning and transmission electron microscopes and shown to be composed of solid, convoluted cords of tissue composed of two major sets of cells, spermatogenic and sustentacular cells. Among the spermatogenic cells, preleptotene spermatocytes and encysted spermatozoa were of most frequent occurrence. The sustentacular cells sent long, cytoplasmic extensions ramifying between and around tightly packed spermatocytes of the seminiferous cords and separated the spermatocytes from the basal lamina which surrounded the cords. Spermatocytes formed desmosome-like and short gap junctions with one another, while sustentacular cells formed intermediate-like junctions and gap junctions with spermatocytes. No special junctions between one sustentacular cell and another were encountered in the present study. 相似文献

We describe a novel method for attaching any DNA molecule to submicron latex beads and characterize the hybridization kinetic properties of these bead-DNA conjugates. The conjugates hybridize to DNA in solution with rates comparable to homogeneous hybridization reactions, are compatible with common hybridization conditions and are conveniently manipulated. They should thus serve as useful reagents for the fractionation and characterization of DNA and RNA. 相似文献

The RNA genome of tobacco etch virus (TEV) is organized as a single translational unit coding for a 346,000 (346 kd) mol. wt (Mr) polyprotein. The 346 kd Mr polyprotein is cleaved by a 49 kd Mr virus-encoded proteinase at five different sites between the dipeptides Gln-Ser or Gln-Gly. These cleavage sites or gene product boundaries are defined by the heptapeptide sequence...Glu-Xaa-Xaa-Tyr-Xaa-Gln-Ser or Gly.... We have used the 54 kd Mr nuclear inclusion protein/30 kd Mr capsid protein junction as a model to examine the role of these conserved amino acids in defining a cleavage site. The 54 kd/30 kd Mr protein cleavage site sequence of 10 TEV isolates from geographically distinct locations has been deduced. The conserved amino acids are present in all isolates. To determine if these four amino acids are an absolute requirement for polyprotein substrate activity, a site-directed mutational analysis has been performed. A recombinant cDNA molecule encoding the TEV 54 kd/30 kd Mr gene product cleavage site was mutated and polyprotein substrates were synthesized and processed in a cell-free system. The dorsal anterior cingulate cortex (dACC) has previously been implicated in processes that influence action initiation. In humans however, there has been little direct evidence connecting dACC to the temporal onset of actions. We studied reactive behavior in patients undergoing therapeutic bilateral cingulotomy to determine the immediate effects of dACC ablation on action initiation. In a simple reaction task, three patients were instructed to respond to a specific visual cue with the movement of a joystick. Within minutes of dACC ablation, the frequency of false starts increased, where movements occurred prior to presentation of the visual cue. In a decision making task with three separate patients, the ablation effect on action initiation persisted even when action selection was intact. These findings suggest that human dACC influences action initiation, apart from its role in action selection. 相似文献

Treatment of penicillin-sensitive and intrinsically resistant Neisseria gonorrhoeae strains with their respective inhibitory concentrations of penicillin caused rapid cell death. When the peptidoglycan syntheses of these two strains were examined in the presence of penicillin, the sensitive strain continued to make this cell wall polymer for an extended time, whereas the resistant strain underwent a rapid and marked depression in synthesis. Examination of the labeled sodium dodecyl sulfate-insoluble peptidoglycan made in the presence of inhibitory concentrations of penicillin revealed further differences. The primary effect on the penicillin-sensitive gonococcus was a slight change in peptide cross-linking and a sharp decline in the degree of O-acetylation. In contrast, the resistant strain exhibited a substantial decline in cross-linking, with a very moderate change in O-acetylation. The degree of saturation of the individual penicillin-binding proteins (PBPs) was assessed under these conditions. PBP 2, which exhibits a reduced affinity for penicillin in the resistant strain, appeared to be related to O-acetylation, whereas PBP 1 was implicated in the transpeptidation reaction. 相似文献

The murB gene, which complemented the UDP-N-acetylenolpyruvoylglucosamine reductase (EC 1.1.1.158) mutation in Escherichia coli ST5, was cloned from an E. coli chromosomal library. murB was subcloned on a 2.8-kb PvuII fragment into pUC19 and sequenced. A 1,029-bp open reading frame encoded a 342-amino-acid polypeptide of 37,859 Da. A DNA sequence homology search revealed that murB had almost 100% homology with a previously reported unidentified open reading frame, ORFII, at 89.9 min. Physical and genetic mapping results were consistent with this map position, and minicell analyses of murB subclones showed a plasmid-encoded protein of approximately 37,000 Da, which closely matched the calculated size of the murB protein. 相似文献
