Due to their large diversity of potential blood hosts, breeding habitats, and resting sites, zoological gardens represent highly interesting places to study mosquito ecology. In order to better assess the risk of mosquito‐borne disease‐agent transmission in zoos, potential vector species must be known, as well as the communities in which they occur. For this reason, species composition and dynamics were examined in 2016 in two zoological gardens in Germany. Using different methods for mosquito sampling, a total of 2,257 specimens belonging to 20 taxa were collected. Species spectra depended on the collection method but generally differed between the two zoos, while species compositions and relative abundances varied seasonally in both of them. As both sampled zoos were located in the same climatic region and potential breeding sites within the zoos were similar, the differences in mosquito compositions are attributed to immigration of specimens from surrounding landscapes, although the different sizes of the zoos and the different blood host populations available probably also have an impact. Based on the differences in species composition and the various biological characteristics of the species, the risk of certain pathogens to be transmitted must also be expected to differ between the zoos.  相似文献

The production of recombinant proteins in the microbial host Escherichia coli often results in the formation of cytoplasmic protein inclusion bodies (IBs). Proteins forming IBs are often branded as difficult-to-express, neglecting that IBs can be an opportunity for their production. IBs are resistant to proteolytic degradation and contain up to 90% pure recombinant protein, which does not interfere with the host metabolism. This is especially advantageous for host-toxic proteins like antimicrobial peptides (AMPs). IBs can be easily isolated by cell disruption followed by filtration and/or centrifugation, but conventional techniques for the recovery of soluble proteins from IBs are laborious. New approaches therefore simplify protein recovery by optimizing the production process conditions, and often include mild resolubilization methods that either increase the yield after refolding or avoid the necessity of refolding all together. For the AMP production, the IB-based approach is ideal, because these peptides often have simple structures and are easy to refold. The intentional IB production of almost every protein can be achieved by fusing recombinant proteins to pull-down tags. This review discusses the techniques available for IB-based protein production before considering technical approaches for the isolation of IBs from E. coli lysates followed by efficient protein resolubilization which ideally omits further refolding. The techniques are evaluated in terms of their suitability for the process-scale production and downstream processing of recombinant proteins and are discussed for AMP production as an example.  相似文献

The inability of articular cartilage to heal satisfactorily is becoming, with ageing populations, an important medical problem. One question that has not been raised is whether a mechanism for the repair of cartilage evolved in animals with cartilaginous skeletons. Fin rays of dogfish were cut and the fish maintained for up to 6 months. The initial inflammatory reaction around the cut rays lasts for 2 weeks. By 4 weeks the cut ends are covered by fibrous tissue. At 12 weeks some areas of cartilage-like tissue are developing. Development of these areas continues and at 26 weeks large chondrocyte-like cells are surrounded by matrix. This tissue is in regions of poor vascularity. It does not have the typical appearance of hyaline cartilage, nor is it integrated with the cartilage of the fin rays. No changes in the cut surfaces of the fin rays are observed at any time. It is concluded that no mechanism has evolved in the elasmobranch fishes for the repair of their cartilaginous skeleton. This is discussed in relation to previous investigations of the reactions of cartilage to injury in embryonic, neonatal and adult tissues of higher vertebrates.  相似文献

It was shown that rat liver contains a series of dolichols with chain lengths of from 17 (or possibly 16) to 21 isoprene residues, the main constituent of the mixture being dolichol-18. By using double-labelled radioactive mevalonates it was demonstrated that each of these dolichols possesses three biogenetically trans-isoprene residues and that the remaining residues are biogenetically cis, suggesting that these polyprenols are biosynthesized from all-trans-farnesyl pyrophosphate by the cis additions of isoprene residues, followed by saturation of the alpha-isoprene residue. The results obtained with these radioactive mevalonates also indicated that the activity of isopentenylpyrophosphate isomerase is low relative to the activity of prenyltransferase in rat liver.  相似文献

The introduction and spread of invasive alien species is a serious threat to ecosystem structures, functions and services worldwide. At enormous cost and effort, attempts are being made to stop and reduce the ongoing appearance of alien species in ecosystems to protect the regional and native biodiversity. Still, management of invasive alien species in particular is often not successful. One reason is a lack of robust toolkits for the management of these species. Guidance on measures which are efficient and effective and which have no negative impact on the environment and human health is urgently needed for decision-making. As a result of an expert workshop, we present a set of three evaluation criteria: effectiveness, ecological impact, and impact on human health. These criteria allow an evaluation of management measures for the prevention, eradication, control and utilisation/disposal of invasive alien species for nature conservation purposes. We applied this evaluation system for 168 invasive and potentially invasive alien species of five taxonomic groups (fungi, non-vascular plants, vascular plants, invertebrates and vertebrates) involving 3619 different management measures. The evaluation of management measures and resulting recommendations for their application are a necessary basis for appropriate, ecologically necessary and professionally sound management. This new approach provides an essential framework for future decisions on management measures, for example in accordance with the provisions of the new Regulation (EU) No 1143/2014 of the European Parliament and of the Council of 22 October 2014 on the prevention and management of the introduction and spread of invasive alien species.  相似文献

The susceptibility of larvae of the Diamondback moth (DBM), Plutella xylostella to infection by three baculoviruses was evaluated in the laboratory using a microdroplet feeding assay. The viruses tested were a granulovirus (GV), originally isolated in Taiwan from P. xylostella larvae (Px GV-Taiwan); the nucleopolyhedrovirus (NPV) from Galleria mellonella (Gm NPV), and the NPV from Autographa californica (Ac NPV). Neonate P. xylostella larvae were susceptible to infection by all three viruses. In an extensive series of bioassays carried out over a 21-month period, LD 50S for neonate DBM larvae ranged from 1.0-8.9 viral occlusion bodies (OB) for Px GV-Taiwan, and 9.5-30.2 OB for Gm NPV and Ac NPV. LT 50S for the three viruses ranged from 3.8-6.0 days at 27 C, with Gm NPV having a significantly shorter LT 50 than the other two viruses. Second and third instar larvae of P. xylostella were significantly less susceptible to infection by Px GV-Taiwan (LD 50 s ranging from 18-57 OB/larva) than were neonate larvae. Gm NPV also initiated infection in several other lepidopterous pest species that colonize brassica crops. In particular, neonate Crocidolomia binotalis larvae proved highly susceptible to Gm NPV, with mean LD 50 s ranging from 2.1 to 9.3 OB/larva and a mean LT 50 of 4.8 days at a dose of 8.08 OB. Heliothis virescens neonate larvae were also highly susceptible to Gm NPV (LD 50 , 7.1 OB), but Mamestra brassicae larvae were less so (LD 50 , 80-270 OB). The results of the bioassays suggest that Px GV-Taiwan is highly infective and could be developed as a selective microbial pesticide for DBM. While Gm NPV has a higher LD 50 in DBM larvae, its wider host range may be of considerable value in situations where DBM occurs on cruciferous crops together with a complex of other lepidopterous pests.  相似文献

The stability and recovery of recombinant proteins expressed in plants are improved by fusion to elastin-like peptides (ELPs). In order to test the suitability of ELP for the production of pharmaceutical proteins, transgenic plants were created that individually expressed the light and heavy chains of the broadly neutralizing anti-human immunodeficiency virus type 1 (anti-HIV-1) monoclonal antibody 2F5, which is being evaluated as a microbicide component. The antibody chains were expressed both with and without a C-terminal ELP fusion. Crossing these plants in all combinations resulted in transgenic lines producing the full antibody in four formats, with ELP on either the light or heavy chains, on both or on neither. Characterization of the affinity-purified antibodies by surface plasmon resonance spectroscopy showed that the kinetic binding parameters were identical to those of a Chinese hamster ovary (CHO) cell counterpart lacking ELP. N -Glycan analysis showed that all four derivatives contained predominantly oligo-mannose-type N -glycans and that the ELP fusions had no significant effect on N -glycan structure. It was concluded that ELP fusion to the light chain, heavy chain or both chains of a plant-derived antibody had no adverse affects on protein quality, but had a positive impact on the yield. ELP fusions do not interfere with folding, assembly, trafficking in the secretory pathway or post-translational modification, but enhance stability whilst at the same time simplifying recovery.  相似文献
