The metabolism of the polyamine precursors arginine and ornithine was studied in maturing and vernalised seeds of Picea abies (L.) Karst. (Norway spruce) in feeding experiments. Incorporation of radioactivity from these 14 C-labelled amino acids into liberated CO2, amino acids, polyamines, proteins and cell wall fractions, as well as polyamine levels were determined in embryos and megagametophytes. Ornithine and especially arginine decarboxylation was more active in the embryo than in the megagametophytic cells, and vernalisation increased arginine metabolism more than it increased ornithine metabolism. Both precursors were metabolised to each other, to other amino acids, and to polyamines. The only polyamine in which radioactivity incorporated was free putrescine, showing either a slow synthesis or a high degradation rate of spermidine and spermine in maturing spruce seeds. The putrescine level was approximately 10 times higher in the embryo than in the megagametophytic tissues, whereas spermidine and spermine levels were almost the same in both tissues. The label from arginine and ornithine was also incorporated into proteins as amino acids and post-translationally as polyamines. Higher radioactivity was seen in the small ≤14-kDa polypeptides. Protein hydrolysates of the embryo and the megagametophytic tissues contained spermidine and spermine and their degradation product 1,3-diaminopropane (DAP), suggesting that polyamines may play a role in the accumulation of seed storage protein and in the maturation of spruce seeds.  相似文献

Invasive bacterial pathogens intervene at various stages and by various mechanisms with the mammalian plasminogen/plasmin system. A vast number of pathogens express plasmin(ogen) receptors that immobilize plasmin(ogen) on the bacterial surface, an event that enhances activation of plasminogen by mammalian plasminogen activators. Bacteria also influence secretion of plasminogen activators and their inhibitors from mammalian cells. The prokaryotic plasminogen activators streptokinase and staphylokinase form a complex with plasmin(ogen) and thus enhance plasminogen activation. The Pla surface protease of Yersinia pestis resembles mammalian activators in function and converts plasminogen to plasmin by limited proteolysis. In essence, plasminogen receptors and activators turn bacteria into proteolytic organisms using a host-derived system. In Gram-negative bacteria, the filamentous surface appendages fimbriae and flagella form a major group of plasminogen receptors. In Gram-positive bacteria, surface-bound enzyme molecules as well as M-protein-related structures have been identified as plasminogen receptors, the former receptor type also occurs on mammalian cells. Plasmin is a broad-spectrum serine protease that degrades fibrin and noncollagenous proteins of extracellular matrices and activates latent procollagenases. Consequently, plasmin generated on or activated by Haemophilus influenzae, Salmonella typhimurium, Streptococcus pneumoniae, Y. pestis, and Borrelia burgdorferi has been shown to degrade mammalian extracellular matrices. In a few instances plasminogen activation has been shown to enhance bacterial metastasis in vitro through reconstituted basement membrane or epithelial cell monolayers. In vivo evidence for a role of plasminogen activation in pathogenesis is limited to Y. pestis, Borrelia, and group A streptococci. Bacterial proteases may also directly activate latent procollagenases or inactivate protease inhibitors of human plasma, and thus contribute to tissue damage and bacterial spread across tissue barriers.  相似文献

Owerwintering strategy of the sub-arctic evergreen Diapensia lapponica has been studied by examining storage substances in the stem cells throughout different seasons. Stem segments were taken from 5 and 20 mm behind the shoot tip, fixed in glutaraldehyde/formaldehyde and embedded in Epon. Semithin sections were stained with Toluidine blue for general observation, Sudan black for lipids, Amido black for proteins and periodic acid-Schiff s reagent (PAS) for carbohydrates. Lipids occurred abundantly throughout the year and seem to be the main storage substances. They were particularly common in mid-winter in the lower part of the shoot. The highest lipid content was found in the pith cells and parenchyma cells of the conductive tissues. Large amyloplasts appeared in mid-summer in higher amounts in the cortex than in the pith of the upper shoot, but vice versa in the lower stem. Starch was almost entirely lacking from November to April. Amido black staining did not indicate presence of protein bodies at any season.
The occurrence of lipids and starch grains in the stem of Diapensia is in general similar to the occurrence of these substances in the leaves throughout the annual cycle.  相似文献

Home ranges, relationships between individuals and dispersal among raccoon dogs ( Nyctereutes procyonoides ) were studied in southern Finland in 1989-91. The average maximum home range, calculated by the harmonic mean method. was 9·5 km2 and the core area (85% utilization) 3·4 km2. There were no statistically significant annual. seasonal or sexual differences in the size of the average core area of adults, but the home ranges of juveniles in autumn were larger than those of adults. However. the maximum home ranges of adults were larger in autumn than in summer, especially those of males, which were conspicuously small in summer and large in autumn. The core areas of adjacent pairs did not usually overlap in the pup-rearing season. but in autumn some home ranges shifted and there was much more overlap. The home ranges of the male and female of a pair overlapped almost totally, and a male and a female sharing the same home range also travelled together or close to each other, thus supporting the idea that the raccoon dog is monogamous in Finland. None of the adults left the study area, but 38% of the juveniles were recovered further than 10 km from the marking place during the first autumn.  相似文献

Aphanizomenon ovalisporum is reported for the first time inGreece, in two warm, monomictic lakes. Aphanizomenon ovalisporumwas dominant constituting 99 and 58% of the total cyanobacterialbiomass in lakes Lysimachia and Trichonis, respectively. Trichomeswere solitary (length 60–700 µm), were narrowedslightly at the ends, had a few terminal hyaline cells and hadcells containing gas vesicles (length 2.5–6.9, width 2.4–5.1µm). Heterocytes, spherical or ellipsoidal (length 4.4–10.5,width 2.41–5.1 µm) and akinetes (length 16.0–27.8,width 6.0–15.9 µm) were located in the middle ofthe trichome. High performance liquid chromatography (HPLC)analysis detected microcystin–LR (MC–LR) and a putativeanabaenopeptin in the L. Lysimachia sample. The sestonic MC–LRconcentration was 0.9 µg L–1. The origin of MC–LRin L. Lysimachia is discussed. The other cyanobacteria presentwere Pseudanabaena sp. and Planktothrix mougeotii (1% of thetotal cyanobacterial biomass).  相似文献

Microcystins are small cyclic heptapeptide toxins produced by a range of distantly related cyanobacteria. Microcystins are synthesized on large NRPS-PKS enzyme complexes. Many structural variants of microcystins are produced simulatenously. FXYD5 (related to ion channel, dysadherin) is a member of the FXYD family of single span type I membrane proteins. Five members of this group have been shown to interact with the Na,K-ATPase and to modulate its properties. However, FXYD5 is structurally different from other family members and has been suggested to play a role in regulating E-cadherin and promoting metastasis (Ino, Y., Gotoh, M., Sakamoto, M., Tsukagoshi, K., and Hirohashi, S. (2002) Proc. Natl. Acad. Sci. U. S. A. 99, 365-370). The goal of this study was to determine whether FXYD5 can modulate the Na,K-ATPase activity, establish its cellular and tissue distribution, and characterize its biochemical properties. Anti-FXYD5 antibodies detected a 24-kDa polypeptide that was preferentially expressed in kidney, intestine, spleen, and lung. In kidney, FXYD5 resides in the basolateral membrane of the connecting tubule, the collecting tubule, and the intercalated cells of the collecting duct. However, there is also labeling of the apical membrane in long thin limb of Henle's loop. FXYD5 was effectively immunoprecipitated by antibodies to the alpha subunit of Na,K-ATPase and the anti-FXYD5 antibody immunoprecipitates alpha. Co-expressing FXYD5 with the alpha1 and beta1 subunits of the Na,K-ATPase in Xenopus oocytes elicited a more than 2-fold increase in pump activity, measured either as ouabain-blockable outward current or as ouabain-sensitive (86)Rb(+) uptake. Thus, as found with other FXYD proteins, FXYD5 interacts with the Na,K-ATPase and modulates its properties.  相似文献

To investigate the responses of Baltic Sea wintertime bacterial communities to changing salinity (5 to 26 practical salinity units), an experimental study was conducted. Bacterial communities of Baltic seawater and sea ice from a coastal site in southwest Finland were used in two batch culture experiments run for 17 or 18 days at 0 degrees C. Bacterial abundance, cell volume, and leucine and thymidine incorporation were measured during the experiments. The bacterial community structure was assessed using denaturing gradient gel electrophoresis (DGGE) of PCR-amplified partial 16S rRNA genes with sequencing of DGGE bands from initial communities and communities of day 10 or 13 of the experiment. The sea ice-derived bacterial community was metabolically more active than the open-water community at the start of the experiment. Ice-derived bacterial communities were able to adapt to salinity change with smaller effects on physiology and community structure, whereas in the open-water bacterial communities, the bacterial cell volume evolution, bacterial abundance, and community structure responses indicated the presence of salinity stress. The closest relatives for all eight partial 16S rRNA gene sequences obtained were either organisms found in polar sea ice and other cold habitats or those found in summertime Baltic seawater. All sequences except one were associated with the alpha- and gamma-proteobacteria or the Cytophaga-Flavobacterium-Bacteroides group. The overall physiological and community structure responses were parallel in ice-derived and open-water bacterial assemblages, which points to a linkage between community structure and physiology. These results support previous assumptions of the role of salinity fluctuation as a major selective factor shaping the sea ice bacterial community structure.  相似文献

Effects of nutrient enrichment on the biomass and communitycomposition of heterotrophic bacteria and picocyanobacteriawere studied in large (42 m3) mesocosms in the brackish-waterArchipelago Sea (Baltic Sea) in late summer 2000 using cellcounts and denaturing gradient gel electrophoresis (DGGE) ofpolymerase chain reaction (PCR)-amplified 16S rRNA gene fragments.The identity of the major DNA bands was determined by sequencing.The obtained sequences were related to - and -proteobacteria,actinobacteria, verrucomicrobia and cyanobacteria. Nitrogenand phosphorus additions increased the biomasses of heterotrophicbacteria and picocyanobacteria and caused significant changesin their community composition judging from the DGGE bandingpatterns. Most verrucomicrobial bands had their highest relativeintensity in the control treatment and their lowest in the highernutrient addition treatment, whereas most Synechococcus-relatedbands had their lowest relative intensity in the lower nutrientaddition treatment. The responses of proteobacteria and actinobacteriawere more variable. The presence of both freshwater and marinesequences among the closest relatives to our sequences highlightsthe intermediate character of the Archipelago Sea between afreshwater and truly marine environment.  相似文献

The impact of predator removal or protection on the reproductive success and density of grouse populations was studied experimentally in southern and northern Finland. In predator removal areas small and medium-sized carnivores wore efficiently hunted, while in predator protection areas hunting was prohibited. Both predator (red fox, raccoon dog, pine marten and stoat) and grouse populations were monitored. The breeding success of grouse, indicated by the young/adults ratio, decreased in the predator protection area in northern Finland during a vole low. Despite a decline in the vole population, the breeding success of grouse did not decline in the predator removal area. The mean brood size during the experiment was significantly higher in the removal areas than in the protection areas both in southern and northern Finland. Predator removal/protection thus affected the reproductive success of grouse, but the impact of control on adult grouse populations was not as evident.  相似文献
