Ultrastructure and peculiarities of interneuronal connections in various zones of neuropil of procerebral olfactory centers of the brain in snails and slugs: in the outer and inner neuropil, zone of input of afferent fibers of labial nerves, as well as zones of running of afferent and efferent fibers of tentacular nerves, were studied. A pronounced spatial morpho-functional differentiation and a complex zonal synaptoarchitectonics of procerebrums is revealed. It has been shown that the procerebrum neural elements, both intrinsic and numerous ones coming from other brain regions and chemosensory systems, contain an enormous variety of vesicles. These vesicles provide connections between neural elements in various synapses and synapse-like junctions and in the composite divergent and convergent complexes formed by them. A positive polychemical nature of granular cells, the main neural elements of procerebrums, and functional significance of symmetric junctions predominant in procerebrums is discussed.  相似文献

pH-Induced intermediates of Omp F-like porin from the outer membrane of Yersinia pseudotuberculosis (yersinin) were characterized by fluorescence and fluorescent probe spectroscopy and circular dichroism. The most dramatic changes in the intrinsic fluorescence of the protein induced by pH titration correlated with different conformational states of the porin molecule. pH-induced conformational transitions of yersinin can be described in terms of a three-state model: (1) disordering of porin associates and formation of porin trimers structurally similar to the native protein; (2) unfolding of individual porin domains followed by cooperative dissociation of trimers into monomers; (3) formation of two loosely structured forms of monomer intermediates. It is assumed that one of these monomeric forms (at pH 3.0) corresponds to the molten-globule state of porin with native secondary structure, while the other one (at 2.0) represents a partly denatured (misfolded) monomer, which retains no more than 50% of the regular secondary structure. The putative mechanism of low pH-induced β-barrel unfolding is discussed in terms of a theoretical model of yersinin spatial structure.  相似文献

Acrylamide is a neurotoxin inhibiting neurotransmission in peripheral nerves. Less is known about acrylamide influence on the central nervous system. Here we measured acrylamide influence on the acetylcholinesterase activity in brain stem, hemispheres, and cerebellum of mice (males, Swiss strain) in relation to the thiol groups and malondialdehyde concentration. Acrylamide was injected intraperitoneally (20 and 40 mg/kg, i.e. 0.52 and 1.04 mg per animal). The brain structures were taken 24, 48, and 192 h after the injection. Acetylcholinesterase activity was significantly lower (p < 0.001 to p < 0.05) in all the structures. It was accompanied by the statistically significant (p < 0.001 to p < 0.05) increase in malondialdehyde concentrations in most of the studied structures time periods and ACR doses. –SH groups concentrations were significantly depleted in the right hemisphere (p < 0.01) after 24 h and in brain stem (p < 0.05) after 48 h. We suggest that neurotoxicity of acrylamide in brain is related to acetylcholinesterase inhibition and redox imbalance.  相似文献

The presented paper is focused on the calculation of hyperfine coupling constants (HFCC) of Cu 2+ ion in water environment. To simulate the conditions of the electron paramagnetic resonance (EPR) experiment in aqueous phase, molecular dynamics using the density functional theory (DFT) was employed. In total three different functionals (BLYP, B3LYP, M06) were employed for studying their suitability in describing coordination of Cu 2+ by water molecules. The system of our interest was composed of one Cu 2+ cation surrounded by a selected number (between thirty and fifty) of water molecules. Besides the non-relativistic HFCCs (Fermi contact terms) of Cu 2+ also the four-component relativistic HFCC calculations are presented. The importance of the proper evaluation of HFCCs, the inclusion of spin-orbit term, for Cu 2+ containing systems (Neese, J. Chem. Phys. 118, 3939 2003; Almeida et al., Chem. Phys. 332, 176 2007) is confirmed at the relativistic four-component level of theory.

Phytoplankton of eutrophic shallow lakes are frequently dominated by one species or species of the same functional group, resulting in species-pure algal assemblages. Knowledge of the structure of these assemblages is essential to understand their functioning; therefore, species and functional diversity were investigated in five sub-types of eutrophic shallow lake. Among the sub-types, astatic saline lakes and hypertrophic ponds had type-specific assemblages dominated by SN and W0, W1 codons. The diversity of the phytoplankton in the sub-types was quite similar, except for the astatic saline lakes, which were characterised by lower values of both functional and species diversity. We found that both functional and species diversity were low when bloom-forming cyanobacteria (H1, SN functional groups) became dominant. Dominance of other groups (J, Y, LO and W1) did not coincide with decrease in species diversity. Analysis of the biovolume versus diversity relationships revealed that decrease in diversity might be expected at biovolume >20 mm3 l−1 for shallow lakes.  相似文献

A series of 14 new compounds related to pyrazinamide were synthesized, characterized with analytical data and screened for in vitro antimycobacterial activity against Mycobacterium tuberculosis, Mycobacterium kansasii and two types of Mycobacterium avium. The series comprised of N-substituted 3-aminopyrazine-2,5-dicarbonitriles derived from 3-chloropyrazine-2,5-dicarbonitrile by nucleophilic substitution of chlorine by various non-aromatic amines (alkylamines, cycloalkylamines and heterocyclic amines). Noteworthy antimycobacterial activity against M. tuberculosis was found among the alkylamino derivatives, for example, 3-(heptylamino)pyrazine-2,5-dicarbonitrile inhibited M. tuberculosis at MIC=51 μmol/L. 3-(Hexylamino)pyrazine-2,5-dicarbonitrile inhibited M. kansasii at MIC=218 μmol/L. Basic structure-activity relationships are discussed. A comparison between calculated and experimentally determined lipophilicity parameters within the series is included.  相似文献

An unstable expansion of the polyglutamine repeat within exon 1 of the protein Htt (huntingtin) causes HD (Huntington's disease). Mounting evidence shows that accumulation of N-terminal mutant Htt fragments is the source of disruption of normal cellular processes which ultimately leads to neuronal cell death. Understanding the degradation mechanism of mutant Htt and improving its clearance has emerged as a new direction in developing therapeutic approaches to treat HD. In the present study we show that the brain-enriched adaptor protein FE65 is a novel interacting partner of Htt. The binding is mediated through WW-polyproline interaction and is dependent on the length of the polyglutamine tract. Interestingly, a reduction in mutant Htt protein level was observed in FE65-knockdown cells, and the process requires the UPS (ubiquitin/proteasome system). Moreover, the ubiquitination level of mutant Htt was found to be enhanced when FE65 is knocked down. Immunofluroescence staining revealed that FE65 associates with mutant Htt aggregates. Additionally, we demonstrated that overexpression of FE65 increases mutant Htt-induced cell death both in vitro and in vivo. These results suggest that FE65 facilitates the accumulation of mutant Htt in cells by preventing its degradation via the UPS, and thereby enhances the toxicity of mutant Htt.  相似文献
