Microsatellite loci were isolated from the sea hibiscus (Hibiscus tiliaceus L., Malvaceae), a pantropical plant with sea‐drifted seeds. This study describes six dinucleotide microsatellite loci for which the primers produced clear and polymorphic amplification patterns with different levels of variability (between three and nine alleles). The types of actin present during development of skeletal muscle in chick embryos were investigated by various electrophoretic methods. The proportions of the actin types in the tissue were estimated by a novel method involving separation of actin bands by electrophoresis, staining of the bands with Coomassie Brilliant Blue R–250, extraction of dye and measurement of its absorbance.
Results showed that α-, β- and γ-actins were all present in embryonic skeletal muscle and were assembled into myofibrils. However, β- and γ-actins disappeared from myofibrils as muscle development proceeded. In order to clarify the role of iron in the growth promoting effect of transferrin (Tf), the effects of the following substances were examined in cultured chick skeletal myogenic cells: transition metal ions (Fe2+ , Fe3+ , Cr3+ , Cu2+ , Mn2+ , Co2+ , Cd2+ , Zn2+ and Ni2+ ), Tf complexes with these metals and metal-free apoTf.
The cells did not grow well when incubated in a culture medium composed of Eagle's minimum essential medium and horse serum. But they grew well in the presence of Fe2+ or Fe3+ (10–100 μM) or iron-bound Tf (10–500 nM) in the medium. None of the transition metal ions other than iron was effective. Neither apoTf nor Tf complexes with these metals showed the growth promoting effect. The generality of the requirement of iron for cell growth was ascertained in the primary culture of other types of chick embryonic cells: fibroblasts, cardiac myocytes, retinal pigment cells and spinal nerve cells.
The results show that iron is one of the indispensable substances for cell growth and suggest that Tf protein plays a role in facilitating the transport of iron into the cells. 相似文献

ABSTRACT. To locate the photoreceptor involved in the photoperiodic induction of diapause in Bombyx mori L., covering of larval head with black paint or local illumination using chemiluminescent paint was carried out. A silkworm race showing a response of long-day type during the larval stage was employed. The results demonstrated that the photoreceptor is located in the head but is extraocular. The optical properties of the larval body suggest that during the first and second stadia light is admitted through the translucent clypeus of the head, but during later stadia enters over the entire larval body including the head, and that it reaches the cerebral lobe where a photoreceptor is possibly located. 相似文献

In the geographically parthenogenetic mayfly, Ephoron shigae, egg maturation and counts of chromosome number of unfertilized, parthenogenetic eggs were studied, in comparison with fertilized eggs from a bisexual population. The primary oocyte becomes mature through two successive maturation divisions. The first maturation division (meiotic division) takes place in the primary oocyte to produce a secondary oocyte and a first polar body. The second maturation division soon occurs in the secondary oocyte, in which the nucleus is divided into a mature egg nucleus (female pronucleus) and second polar body nuclei. The first polar body, in some cases, was successively divided into two polar bodies; in other instances, it was not divided. After the successive maturation division, the egg nucleus and the sister second polar body nucleus drew near to fuse into the zygote nucleus. The chromosome number was doubled in the zygote, and this conjugation initiates subsequent embryonic development.
