Micromeres and mesomeres isolated from 16-cell embryos of the sea urchin, Strongylocentrotus intermedius , were cultured in vitro , and changes in the cells surface architecture during the differentiation of the micromere- and mesomere-derived cells were observed using scanning electron microscopy. Two types of the distribution of the surface microvilli were observed in both blastomere-derived cell masses. One type showed a uniform distribution of the microvilli and the other type showed an uneven one. Though many microvilli were observed in most of both mesomere and micromere-derived cells at the 64-cell stage and the early blastula stage (16 hr after the 16-cell stage at 6°C) respectively, the microvilli decreased in number at the later stages in both blastomere-derived cell masses as compared with the 64-cell stage and the early blastula stage respectively. Rapid disappearance of the surface microvilli was observed in the micromere-derived cells in contrast with the mesomere-derived cells which still had many microvilli even at the midmesenchyme stage.  相似文献

ABSTRACT

We found a new acoustic signal in Drosophila simulans (si) and D. melanogaster (me). It is a ‘rejection signal’ (RS) produced by adult males and young males and females in response to the courting behaviour of mature males who emit ‘pulse songs’ (i.e. love song: LS). It occurs most frequently in si, less in adults me except if the interacting males belong to different chemical morphs (i.e. temperate or equatorial population). There are no differences in the LS characteristics directed to various sexes and ages. The RSs produced by adult males or by young animals do not differ significantly either. They are emitted by neither virgin nor fecundated adult me females but a few times by virgin adult si females. The RS (like the LS) is a multipulse signal but intervals between pulses are about twice those of LS, around 90 ms for si and 80 ms for me. They are very irregular, as is the distribution of energy along the bandwidth mainly between 300 and 800 Hz for si and 200 and 600 Hz for me. The sound level of the RS is from 10 to 20 dB less than the LS. The RS seems to be linked to the ‘flicking’ behaviour produced by both wings, while the LS always corresponds to the so-called ‘wing vibration’.  相似文献

Plodia interpunctella Hübner (Lepidoptera: Pyralidae) comprises a model insect to analyse the photoperiodic time‐measuring system controlling its larval diapause. In the present study, the effective length of light pulse in night interruption experiments is determined at 25 °C. Various lengths of light pulse are tested by inserting them at the midnight of an LD 12 : 12 h photoperiod. When the light pulse is 15 or 30 min, the incidence of diapause is 86%. To inhibit the induction of diapause effectively, a light pulse of 1.75–2 h is needed. The incidence of diapause is 12% under an LD 12 : 5 : 2 : 5 h photoperiod. To determine the precise role of the light pulse, 2‐h light pulses placed at the midnight of an LD 12 : 12 h photoperiod are disrupted systematically by darkness. When a 2‐h light pulse is disrupted by 15 min of darkness, diapause is generally prevented (< 29%) regardless of the temporal position of darkness. Longer disruption by darkness induces diapause moderately (37–67%). A Bünsow experiment is also conducted at 25 and 20 °C, in which the main photophase of 12 h of light is combined with 24–72‐h scotophases scanned by a 2‐h light pulse. The photoperiodic cycle length tested, therefore, varies in the range 36–84 h. In each cycle length, the incidence of diapause fluctuates as the light pulse moves toward dawn. However, no regular and circadian changes in the percentage diapause are observed in relation to diapause determination.  相似文献

Round spermatids (steps 1–8) were isolated from rat testes and the effect of pyruvate on their intracellular ATP level was examined. Results showed that although the spermatids consumed a considerable amounts of pyruvate, this substrate alone did not maintain their ATP level. However, their ATP level was maintained in the presence of both pyruvate and α-ketovalerate or α-ketobutyrate. Maintenance of the ATP level by these substrates was associated with electron trasnport and oxidative phosphorylation. α-Ketoacid inhibited pyruvate reduction to lactate in the lactate dehydrogenase (LDH) reaction, but increased pyruvate oxidation to CO2. The NADH level in spermatids was too low to be detectable, but the NAD level remained unchanged in the presence of pyruvate and α-ketovalerate. These results suggest that pyruvate by itself is not an adequate energy-yielding substrate for spermatids and that a high NADH/NAD ratio may be essential for maintenance of their ATP level.  相似文献

Melanin precursors and free radicals, cytotoxic substances, are produced during melanin synthesis by tyrosinase. We compared these cytotoxic effects of L-dopa and oxygen on the cell cycle of melanotic retinal pigment epithelial (RPE) cells with amelanotic RPE cells because of the differences of tyrosinase activities between melanotic and amelanotic RPE cells. Flow cytometric DNA analysis of RPE cells exposed to L-dopa (100 μM and 250 μM) were conducted at several oxygen concentrations (20%, 10%, and 5%). The dose-dependent effect of L-dopa to arrest the cell cycle (the S phase) was more pronounced in melanotic than in amelanotic RPE cells, and oxygen caused arrest in the G1 phase.  相似文献

We previously observed the presence of autofluorescent lipofuscin or its like in retinal pigment epithelial (RPE) cells, which were incubated with linoleic acid hydroperoxides (LHP). We studied the effect of oxygen on the level of lipid peroxides in RPE cells in the presence of linoleic acids (LA) or LHP. The level of lipid peroxides in these cells was determined by use of the thiobarbituric acid-reactive substance (TBARS), which responded to oxygen concentrations qualitatively, and a linear regression analysis. Multiple linear regression analysis disclosed that treatment with LA for 24 hr resulted in detectable increase in the level of TBARS in the cells, whereas treatment with LA or LHP for 48 hr caused detectable decrease. Stepwise linear regression analysis showed that the level of TBARS decreased in an oxygen-tension dependent manner in the cells incubated with LA for 48 hr. Thus, it was shown that short-term incubation with LA increased the level of TBARS in the cells and that LA decreased its level in an oxygen-tension dependent manner. For these results, the postulation was made that the prolonged auto-oxidation of LA caused production of lipofuscin-like materials, a complex of lipid peroxides and proteins that were insoluble in SDS and acetic acid solution.  相似文献

Glucose utilization by spermatids was found to be 17.37±0.37 nmoles/hr/106 cells at 34°C and 28.94±1.12nmoles/hr/106 cells at 40°C. A good parallelism was observed between the increased rate of glucose utilization and lactate production at 40°C. There was no significant change in the levels of glycolytic intermediates in the cells, except for marked accumulations of fructose-1, 6-diphosphate, dihydroxyacetone phosphate and glyceraldehyde-3-phosphate in the presence of glucose (1 mM). Glucose oxidation in the citrate cycle by spermatids was higher at 40°C than at 34°C, but was never greater than 2% of the overall rate of glucose utilization. In addition, glucose did not prevent decrease of ATP at either 34 or 40°C. The effects of temperature on the activities of 11 glycolytic enzymes were examined. The activities of aldolase and phosphoglyceromutase were similar between 30 and 34°C, but increased markedly at 40°C. The higher temperature increased the Vmax values, without affecting the Kms. The activities of other glycolytic enzymes were similar at the different temperatures. These findings indicate that the increased overall rate of glucose utilization in glycolysis at higher temperature is due to increased Vmax values of aldolase and phosphoglyceromutase.  相似文献
