Japanese huchen, Hucho perryi, is an endangered fish in Japan. In order to promote a conservation program for this species, it is necessary to evaluate its genetic diversity and population genetics. For this purpose, we designed 15 primers containing dinucleotide microsatellite regions and examined their allelic variation using a total of 23 wild individuals collected from two different localities. Eight loci showed moderate allelic variation ranging from two to four alleles, with expected heterozygosities from 0.043 to 0.548. These markers will be useful for the assessment of genetic variation for this species.  相似文献

We studied isozyme variation in two annual species that produce bulbils, Sedum rosulato-bulbosum , which includes both sexually reproducing plants and obligate clonal plants that result from triploidy (fertile and sterile S. rosulato-bulbosum , respectively), and an obligate clonal plant, Sedum bulbiferum , to examine the relationship between reproductive mode and isozyme variation. The sterile S. rosulato-bulbosum population exhibited no genotypic variation, but showed high genetic variation (gene diversity, H e  = 0.60) because five of the six loci that we analyzed were heterozygous. Almost all ramets of S. bulbiferum across 20 populations shared an identical isozyme phenotype, although we could not identify the genetic basis of the phenotype. In contrast, fertile S. rosulato-bulbosum exhibited genotypic variation across the species, but comprised genotypically uniform and polymorphic populations whose genotypic variations correlated positively with the genetic variations within the populations ( H e at the genet level per population ranged from 0.08 to 0.37). Genetic drift and habitat conditions inhibiting seedling recruitment may have caused this among-population variation. The results for sterile and fertile S. rosulato-bulbosum suggest that exclusive clonal reproduction causes low genotypic variation, but maintains genetic variation within individuals. Factors that affect the maintenance of genetic variation in these plants are discussed on the basis of these findings.  相似文献

Light-sensitivity of the first and the last processes in theinductive dark period, and the effect of twilight on photoperiodicinduction were investigated with four short day plants. In Oryza sativa (Akebono), the first and the last processesare relatively light-insensitive, and the photoperiodicallyeffective day length under natural conditions is consideredto be equal to the astronomical day length. In Biloxi soybean and Perilla frutescens, the first processis re latively light-insensitive, but the last process is moresensitive. Effective dark period seems to begin when twilightis 50200 lux in the evening and continue until it is about 10lux in the morning. In Xanthium saccharatum, both the first and the last processesare relatively light-sensitive, but the former is more sensitivethan the latter. Eight polymorphic microsatellite markers were developed in Dryobalanops aromatica, an emergent tree in tropical rain forests in Southeast Asia, using an enriched library method. For the assessment of microsatellite variation, 36 individuals from a natural population were analysed. The number of alleles per locus ranged from three to 16, with observed heterozygosity of 0.056–0.833 and expected heterozygosity of 0.054–0.882. These microsatellite markers will be useful for studies of population genetics, reproductive ecology and regeneration dynamics of D. aromatica.  相似文献

The first microsatellite markers were isolated from the golden cuttlefish, Sepia esculenta Hoyle. Eleven primer sets were designed to amplify the marker sequences via polymerase chain reaction. The 45–50 individuals from one wild population in the coastal waters of Ehime Prefecture, Japan were used to screen polymorphism in the 11 microsatellite loci. All the microsatellite loci were polymorphic, with the range of alleles from seven to 27 per locus. The observed and expected heterozygosities ranged from 0.380 to 0.980 and from 0.654 to 0.940, respectively. These marker loci except for one locus showing significant deviation from Hardy–Weinberg equilibrium will be useful for the assessment of genetic variation and population structure of this species.  相似文献

The previously reported observation that micromere formation after cleavage suppression is not linked with the number of blastomeres present but rather with the time schedule of the fourth cleavage of the normal embryos has been confirmed. A hypothesis is advanced that a rhythmical fluctuation of the sulfhydryl contents of the egg proteins is the clock system, and micromere formation is connected with the fourth SH cycle after fertilization. The hypothesis was tested under 3 conditions:
  • (i) Conditions which stop the nuclear activities but preserve the SH cycle, followed by a release from these conditions.
  • (ii) Conditions which “freeze” both nuclear and cytoplasmic rhythms, and later removal of the conditions.
  • (iii) Conditions which leave nuclear activities intact but prevent the cytoplasmic rhythms, followed by normal culturing.
The results came out as anticipated by the hypothesis.  相似文献
