We developed 10 microsatellite loci from genomic DNA of a dioecious clonal plant, Dioscorea japonica. Out of 384 clones, 148 contained microsatellite repeats. Polymerase chain reaction primer pairs were designed for 95 of these clones from their sequence data, of which, 10 pairs produced successful amplification. Thirty‐eight individuals were genotyped for allelic diversity. We detected three to nine alleles per locus, and the expected heterozygosity ranged from 0.461 to 0.851.  相似文献

The effects of dietary inositol with sucrose stimulation on chewing and swallowing motor patterns in the larvae of Bombyx mori L. are investigated. Feeding activities of the larvae are significantly enhanced by a test diet containing an inositol–sucrose mixture compared with a test diet of sucrose only. Motor patterns of the mandibular closer muscle are accelerated with shorter burst durations and shorter inter‐burst intervals with the test diet of inositol–sucrose compared with sucrose. In terms of swallowing behaviours, inositol–sucrose shortens the duration of drinking. Motor patterns of the cibarial compressor muscle are accelerated with shorter burst durations and shorter inter‐burst intervals with the inositol–sucrose mixture. Peripheral interactions between inositol‐ and sucrose‐sensitive cells in the maxilla are not detected. Thus, such interactions cannot explain the positive effects of inositol on chewing and swallowing. Responses of inositol‐sensitive cells in the epipharyngeal sensillum are not affected by sucrose. These results suggest that dietary inositol can modify chewing and swallowing motor patterns when coupled with sucrose stimuli. These modifications may occur in the central neural networks involved in chewing and swallowing motor patterns but not in peripheral sensory interactions.  相似文献

Rotenone-sensitive 14CO2 formation from [14C]lactate and oxygen consumption by round spermatids were found to be greater at elevated temperatures than at 34°C. More than 96% of the total radioactivity of the metabolized [14C]lactate was recovered in the released CO2 and the acid soluble fraction of the cells. There was practically no incorporation of [14C]latctate into the lipid, nucleic acid, and protein fractions. Intracellular level of ATP in spermatids was enhanced in the presence of lactate (20 mM) at 34°C (scrotal temperature), whereas it was decrease at 37°C (body temperature). However, this was reversible when the cells were transferred from the elevated temperature to 34°C. It was also found that oxygen consumption and CO2 production were increased at 34°C by 2, 4-dinitrophenol (DNP), but decreased by oligomycin. On the other hand, oligomycin and DNP had no effect on oxygen consumption and 14CO2 formation at the elevated temperature.
These findings provide evidence that lactate utilization by spermatids is coupled with oxidative phosphorylation at scrotal temperature, but becomes uncoupled at elevated temperature, although more lactate is consumed.  相似文献

Nuclear change in relation to axis formation and condensation during spermiogenesis was investigated in the snail, Physa acuta. In the early spermatid, characteristic thick layers (termed apical and basal plates) are formed on two sides of a nuclear envelope. Soon after the formation of these plates, a developing acrosome and a flagellum attach externally to the center of the apical and basal plates, respectively. However, most (presumably all) of the chromatin filaments become attached all over the inner surface of the apical and basal plates. This means that the plates themselves are actually the specialized forms of the nuclear envelope to which chromatin filaments become connected; by means of these plates, the chromatin filaments become arranged in parallel to the antero-posterior axis as the nucleus elongates. This suggests that the formation of these two thick layers on opposing surfaces of the nucleus primarily determines the antero-posterior axis of the spermatid and the direction of the arrangement of chromatin.
The flattening of the nucleus prior to elongation is caused mainly by the enlargement of the basal plate. Subsequent nuclear shaping and condensation are discussed in relation to the change in the surface structures of the nucleus and the organization of the microtubules.  相似文献

  1. The capacity of green leaves for absorbing and converting substancesrelated to L-ascorbic acid was investigated, using detachedleaves of various plants, including soybean, pea, barley andspinach.
  2. The greater portion of the absorbed dehydroascorbicacid wasrecovered in the leaves as ascorbic acid, minor portionsbeingdiscovered as dehydroascorbic acid and 2,3-diketogulonicacid. The recovery was about 60% of the absorbed dehydro ascorbicacid, in the cases of detached soybean and barley leaves, forinstance, thus suggesting the instability of the substance invivo.
  3. The absorption and conversion of ascorbic acid and 2,3-di-ketogulonic acid in detached green leaves were also investigated.Most of the absorbed ascorbic acid reappeared in the leaves.On the other hand, no evidence for the conversion of 2,3-diketogulonicacid into ascorbic and dehydroascorbic acids was observed. Thegreater portion of the absorbed 2, 3-diketogulonic acid seemsto be decomposed in the leaves, under the condition of the presentstudy.

The development and the size distribution of free thick filaments which accumulate in the early stages of myofibril formation in somitic myoblasts of the ascidian tadpole were studied by electron microscopy. Such filaments appeared in the cell cortex but, rather dominantly, the aggregates of these thick filaments and filamentous structures were observed in the interior of the cell. The aggregate consisted of some of the following elements: filamentous structures (20–60 A in diameter); free thick filaments (60–220 A); dense Z-band precursor materials; bundles of thick (140–160 A) and thin (60–70 A) filaments; and ribosomal clusters. The free thick filaments were variable in diameter and showed long lateral projections (300–600 A) and tapered ends.
The variation curve in diameter of the free thick filaments indicates a continuous size distribution, suggesting the continuous growth of these filaments by polymerization of myosin molecules. Free thick filaments thicker than myosin filaments which were found within myofibrils were present; their significance is discussed in relation to myosin filament formation.  相似文献

Unfertilized eggs of the sea urchin, Hemicentrotus pulcherrimus , were separated into two fragments by centrifugal force. The enucleate fragment (merogone) was subsequently activated by treating it with butyric acid and the tension at the surface was continuously measured by a compression method. The activated merogone was found to exhibit cyclic changes in tension, with a temporal pattern very similar to that of the changes accompanying the division cycle of normally fertilized eggs. This indicates quantitatively the presence in the cytoplasm of some periodic activity which can be triggered without nuclear control. Further, a periodic thickening of the intrahyaloplasmic space of the activated merogone, as noted by K ojima (14), was confirmed on the basis of extended observation.  相似文献

Abstract. The surface electric potential of Vigna sesquipedalis stem, relative to that of the root medium, was most negative in the elongating zone where the difference in pH between parenchyma (P) and xylem sap (X) was at a maximum. The surface electrical potential was found to be determined by the distribution of radial potential difference ( V sx comprised two components; Vsx= V px− V ps.There were two electromotive forces, generating V ps at the surface of the parenchyma symplast and V px between P and X; both were partially dependent on respiration. The depolarization of V px upon removal of O2 was maximal in the elongating zone, suggesting that the activity of an H+-pump extruding protons from the parenchyma into the xylem in exchange for K+ was greatest in this zone.  相似文献

This study observed the adaptability of carp neutrophilic granulocytes possessing spontaneous cytotoxic activity to different environmental temperatures. To study the adaptability of neutrophilic granulocytes, two different temperatures (25° C and 10° C) were selected, both for rearing and forin vitroassays, in which the cytotoxicity and the adherent rate against K562 target cells were measured. The cytotoxicity and adherent rate of neutrophilic granulocytes from carp kept at 25° C for 30 days were higher when assayed at 25° C than when assayed at 10° C. On the other hand, in carp acclimated from 25° C to 10° C, the cytotoxicities and adherent rates, when assayed at 25° C, decreased with increasing acclimation times, eventually becoming smaller than the values obtained when assayed at 10° C. After the fish kept at 10° C for a long period were re-acclimated to 25° C, these activities assayed at 25° C again became higher than the activities assayed at 10° C. These results indicated that carp neutrophilic granulocytes adapt their cytotoxic activity and adherent activity to different environmental temperatures. A change in cellular composition in the head kidney was also observed in carp kept at different environmental temperatures. The percentage of neutrophilic granulocytes became higher and lymphocytes became lower in carp that were kept at 10° C for a long period compared with carp that were kept at 25° C for a long period.  相似文献
