The Effects of Priming and Ageing on Seed Vigour in Tomato   总被引:1,自引:0,他引:1  
A comparison was made of the effects of seed priming or ageingtreatments on the performance of tomato (Lycopersicon esculentumMill. cv. UC204C) seeds according to a number of indices ofseed vigour. A single lot of tomato seeds was primed in 120mol m–3 K2HPO4 + 150 mol m–3 KNO3 for 5 d at 20?C, or aged at 13% moisture content (dry weight basis) and 50?C for 6 d. Germination percentage (>98%) was unaffectedby priming and reduced to 85% by ageing. X-ray photographs andlongitudinal sections revealed the formation of free space surroundingthe embryo in dry primed seeds, which was not evident in controlor aged seeds. Priming increased the rate of germination atall temperatures above the base temperature (Tb), while ageingdecreased it. Tb was unaffected by priming and only slightlyincreased by ageing. The variation in individual times to germinationwas approximately doubled in both primed and aged seed comparedto the control, based upon the slopes of probit germinationpercentage versus log thermal time curves. Root growth aftergermination tests and seedling growth in both greenhouse andfield tests were not influenced by either priming or ageing.The conductivity test was found to be unreliable as a vigourtest for tomato seeds. The results identify several indiceswhich can be used to quantify seed vigour in tomato. They alsoillustrate that seed priming can enhance seed performance accordingto some criteria, while having no effect or decreasing qualityaccording to other criteria. Seed vigour can apparently be separatedinto various components which can be independently influencedby seed enhancement treatments. Two new species of Myxozoa from the brain of the green knife fish Eigemannia virescens are described: Myxobolus inaequus sp. n. has an unusually large spore body and extremely unequal polar capsules, and Henneguya theca sp. n. has an attenuated spore encased in a sheath not previously described in other Myxozoa. Only spores of the two species were observed, and infections caused no obvious pathological changes in the brain.  相似文献

Incubation of excised cucumber cotyledons (Cucumis sativus L.) with NH4Cl solutions exceeding 0.001 M inhibited their greening, fresh weight increases, and incorporation of 14C-leucine into insoluble N compounds. The absorption of 14C-leucine during incubation and retention of moisture by the excised cotyledons after incubation were greatly diminished by the NH4Cl treatments. Treatment with KCl solutions of the same concentrations as the NH4Cl stimulated the greening, fresh weight increases, and the absorption and incorporation of 14C-leucine. Desiccation of cotyledons stored at 5°C for 10 days was inhibited by dilute KCl solutions. The toxicity of NH4Cl was alleviated by KCl solutions at low concentrations.  相似文献

The germinability, vigour, and desiccation tolerance of muskmelon(Cucumis melo L. cv. Top Mark) seeds was studied in relationto changes in seed water content during development within thefleshy fruit. Seed water content (fresh weight basis) declinedfrom 91% to 42% between 10 d and 35 d after anthesis (DAA) (whenmaximum dry weight was attained), then declined more slowlyto a minimum of 35% at 50 DAA before increasing again to 43%at 65 DAA. Fresh intact seeds were first germinable at 25 to30 DAA and attained maximum germination percentages at 45 DAA.Between 15 and 35 DAA, cotyledons, hypocotyls, radicles andepicotyls of isolated embryos (testa and perisperm enveloperemoved) sequentially developed the ability to grow when incubatedon water. Dehydration to water contents less that those attainedwithin the fleshy fruit is not a requirement for developmentof germination capacity of muskmelon seeds. Seeds became tolerantof rapid desiccation after 25 DAA, and drying of immature seeds(25 to 40 DAA) increased their germination percentages uponsubsequent imbibition. Washing, drying, or washing followedby drying increased seedling vigour (root length) as comparedto fresh seeds, which had very poor vigour. Water absorptionisotherms were constructed to test whether changes in water-bindingcomponents were correlated with the development of desiccationtolerance. Isotherms for seeds older than 25 DAA fit well tothe D'Arcy/Watt model, which postulates the existence of high-affinity,low-affinity and multi-molecular water-binding sites. Desiccation-intolerantseeds younger than 25 DAA lacked the component of the absorptionisotherm characteristic of the high-affinity water-binding siteswhich have been hypothesized to confer desiccation tolerance.However, we were unable to determine whether the absence ofhigh-affinity binding characteristics was specifically relatedto desiccation intolerance or was artifactual due to the lossof volatiles when immature seed samples were dried at high temperatures.
