All isolates of tobacco rattle virus (TRV) found in naturally infected narcissus leaves produced nucleoprotein particles, mostly in large concentrations but, because of antigenic diversity, less than half of the isolates were identified by immunosorbent electron microscopy (ISEM) and still fewer by enzyme-linked immunosorbent assay. All were identified by a nucleic acid hybridisation test in which DNA complementary to RNA-1 of strain PRN of TRV was allowed to react with nucleic acid extracted from leaf tissue. Spraing-affected tubers in some potato stocks yielded only NM isolates of TRV. These isolates do not produce virus particles and they were therefore not detected by ISEM. The infectivity of nucleic acid extracts from recently harvested tubers with spraing symptoms was much greater than that of extracts prepared from tubers after 8 months' storage. In other potato stocks, some spraing-affected tubers contained NM isolates and the rest contained particle-producing isolates (M isolates) of TRV. The infectivity of sap and of nucleic acid, extracted 7 months after harvest from tubers infected with M isolates, was much greater than that of nucleic acid extracted from comparable tubers infected with NM isolates. TRV was detected by nucleic acid hybridisation in extracts of almost all tubers containing either M or NM isolates, even when the tubers were not tested until 7–8 months after harvest. The probable sequence of events occurring after tubers are infected with TRV is outlined, and it is suggested that the virus will rarely become established in fields as a result of planting infected tubers. 相似文献

SYNOPSIS. The fine structure of 2 isolates of Trypanosoma congolense maintained in laboratory rodents has been studied from thin sections of osmium- and aldehyde-fixed flagellates. The pellicular complex, nucleus, and flagellar apparatus are all similar to those of other African trypanosomes. Aberrant intracellular differentiation of the flagellum is occasionally found. As in bloodstream forms of other salivarian trypanosomes the single mitochondrion forms an irregular canal running from one end of the body to the other, with a shallow bowl-shaped expansion forming a capsule for the fibrous kinetoplast (mitochondrial DNA). A connexion between the mitochondrial envelope of the kinetoplast and the basal body of the flagellum is not evident, and sometimes the flagellum base is not even apposed to the kinetoplast but lies behind it. Tubular cristae are present in the mitochondrial canal and, by light microscopy, this structure gives a positive reaction for NAD diaphorase suggesting at least some activity in electron transport, even tho at this stage in its life cycle respiration is doubtfully sensitive to cyanide and cytochrome pigments are in all probability absent. The region of the cytoplasm between the nucleus and the flagellar pocket has all the trappings associated with secretory cells in higher animals, or with the secretion of surface structures in phytoflagellates. just behind the nucleus a limb of granular reticulum subtends a Colgi stack of flattened saccules with attendant vesicles. Close to the distal pole of the Golgi complex is a network of smooth-membraned cisternae, termed here the agranular or secretory reticulum, which undergoes localized swelling with the accumulation of a secretory product to form large spherical sacs or vacuoles. These network-linked vacuoles probably correspond to the post nuclear vacuole complex visible by light microscopy. From its apparent secretory function this complex is regarded here as being possibly an extension or derivative of the Golgi complex, the smooth-membraned tubules lying alongside the 2 structures possibly representing a link between them. By analogy with phytoflagellates and the secretory cells of higher animals, it is suggested that the secretion is transported for discharge into the flagellar pocket by way of multivesicular bodies and smooth-walled tubules or vesicles. Spiny pits in the wall of the flagellar pocket, and similar-sized vesicles in the nearby cytoplasm, could be stages in either exocytosis of secretion or endocytosis (pinocytosis). It is tentatively suggested that the secretion may be the material from which the surface coat is formed. Neither a cytostome nor a contractile vacuole has been observed in T. congolense. 相似文献

Traditionally, compared with the tropics, temperate systems are believed to: (i) have environments which are less favourable (i.e. harsher, more variable and less predictable); and (ii) support communities which are less diverse. Explanations for differences between temperate and tropical communities, including differences in diversity, generally rely on the former notion. The evidence for these ideas is, at best, equivocal. Organisms are subjected to physical stresses and disturbances on both temperate and tropical reefs. Communities on temperate reefs are not invariably less diverse than those in the tropics, at least at small spatial scales. Finally, there is as yet little evidence of genuine differences between the ecology of temperate and tropical communities. There is, however, much small-scale, spatial patchiness in the structure of reef communities and their physical environment. This patchiness in structure may result from patchiness in biological factors (e.g. recruitment) or in the physical environment. This small-scale variation in environmental factors may prove to be a more important determinant of community structure than the large-scale, latitudinal trends some ecologists have been obsessed with. 相似文献

SYNOPSIS. Cryptobia vaginalis (Hesse 1910) occurs as long thin and short broad forms in the vagina of the gnathobdelliform leeches Haemopis sanguisuga (Linnaeus) and Hirudo medicinalis Linnaeus. Cytochemical staining for DNA and transmission electron microscopy of sectioned material indicate that in the thin forms the kinetoplast DNA (kDNA) is dispersed irregularly through the mitochondrial network ( pankinetoplastic condition) rather than concentrated in the adbasal region of the mitochondrion ( eukinetoplastic condition) as in trypanosomatids and most other kinetoplastid flagellates. Light-microscopic studies on the rare broad forms, however, suggest that these have conventional adbasal location of the kinetoplast. Binary fission appears to occur in the thin forms, suggesting that the dispersed kinetoplast is either highly polyenergid or lacks a genetic function. In other features of its microanatomy, C. vaginalis is a conventional kinetoplastid. The flagellate has an incomplete corset of pellicular microtubules which may have a role in the cortical contractility characteristic of the genus Cryptobia . Feeding is by pinocytosis of vaginal colloids through a microtubule-lined cytopharynx, possibly after binding to a prominent filament-coated preoral ridge. A pulsatile (contractile) vacuole is present and appears to be responsible for defecation as well as osmoregulation. Some individuals have elongate bacterial epibionts attached to the body in parallel with the cortical microtubules. All individuals have 2–8 spheroplast-like endobiotic bacteria in the prenuclear cytoplasm. 相似文献

- 1 Eggs of the British woodland mosquito, Aedes punctor (Kirby), were subjected to a variety of media in an attempt to determine the hatching stimulus.
- 2 It was found that continuous immersion in distilled water evoked almost no hatching (0.2%); intermittent removal from the water gave low hatching (8.9%).
- 3 Slow deoxygenation of the medium by either bacterial growth or the introduction of nitrogen produced the highest level of hatching (26.0–90.0%). Eggs of A.punctor were shown to hatch within a 4 h period of the oxygen concentration in the medium reaching zero.

(1) A series of factors controls sexual morphogenesis in the Ascomycotina, a process involving the formation of novel structures such as ascocarps (fruit bodies) and asci (sacs containing spores) during sexual reproduction. (2) Environmental and genetic factors must be correct before Ascomycetes may sexually reproduce. Compatibility in many heterothallic species is under polygenic control, with the mating type loci and also other genetic factors determining the productivity of sexual crosses. (3) Classical genetic studies have shown that sexual morphogenesis involves the expression of a series of developmentally regulated genes, and this has been confirmed by recent molecular studies which have demonstrated changes in patterns of mRNA and protein synthesis during ascocarp formation. (4) Hyphal differentiation leading to the formation of mature fruit bodies occurs in response to a series of signals, which include various physical and chemical factors. (5) Chemical sex factors have been identified which are believed to have important regulatory or nutritional roles in sexual morphogenesis. These include the following. (a) Diffusible sex hormones which may regulate developmental switching between asexual and sexual modes of reproduction, including (i) pheromones involved with the induction of gametangia and gamete attraction, and (ii) sex morphogens involved with triggering particular stages of fruit body formation. (b) Sexual growth substances which are required as nutrients, and may be precursors for the production of sex hormones, or metabolites used in the synthesis of novel sexual structures. Most of these sex factors are lipids. (6) Certain sex morphogens and sexual growth substances have been shown to exhibit activity in a variety of fungal species, suggesting that fungi of related phylogenetic descent may utilize similar metabolites or signalling factors during sexual reproduction. (7) Phenoloxidase enzymes may catalyse hyphal aggregation in developing fruit bodies. (8) Initial stages of ascocarp development may occur independently of the events of the sexual cycle. However, a link(s) with the functional ascogenous hyphae is needed for the formation of morphologically mature ascocarps. (9) Suitable environmental conditions are sufficient to trigger sexual morphogenesis in homothallic Ascomycetes. However, an extra level of control is present in heterothallic species, with a compatible partner required to complete sexual reproduction. This may be partly because novel regulatory products, formed by the combined action of the mating type loci of different partners, are required for further ascocarp development. (10) Further research is required to identify more fungal chemical sex factors and to determine the role of environmental stress in controlling sexual morphogenesis, and how this may be related to temporal patterns in the expression of mating type genes. 相似文献

Abstract A clear correlation between the presence of stomata on both surfaces and factors such as habitat, growth form, and physiology has yet to emerge in the literature. However, certain loose trends with these factors are evident, and these are reviewed along with evidence for hypostomaty as the primitive form. It is proposed that the effect of developing stomata on the upper surface as well as the lower is to increase maximum leaf conductance to CO2. Plants with a high photosynthetic capacity, living in full-sun environments, and experiencing rapidly fluctuating or continuously available soil water (as opposed to seasonal or long-term soil water depletion), are identified as deriving an adaptive advantage from a high maximum leaf conductance. The correlation between groups of plants fitting the above conditions and those noted to be amphistomatic is remarkable. Plants not fitting the conditions are found to be largely hypostomatic. 相似文献
