The subfamily Horelophopsinae was originally proposed as one of the earliest diverging clades of Hydrophilidae (s.s.), but its phylogenetic placement has never been tested. We describe the larva of Horelophopsis hanseni Satô et Yoshitomi, 2004 of the Horelophopsinae. Larval data are based on larval specimens collected together with adults, and unambiguously associated with them by means of DNA barcoding. We perform an analysis testing the phylogenetic position of H. hanseni based on larval and adult morphological characters. Horelophopsis hanseni is unambiguously placed within the hydrophilid subfamily Hydrophilinae and its close relationships to the genus Agraphydrus Régimbart, 1903 (Hydrophilinae, Acidocerini) is recognized. The results suggest that the subfamily Horelophopsinae is unlikely to be a basal taxon of Hydrophilidae, as originally suggested.  相似文献

对产自乳酸菌Enterococcuze fecalis TN-9的蛋白酶,进行了硫酸铵沉淀,DEAE—Sephadex A-25以及DEAE Cellulofine A-500离子交换层析的3步纯化和特性研究。纯化酶Native PAGE显示1条蛋白带。SDSPAGE和凝胶层析分子量分别为30ku及69ku。纯化酶最适作用温度为30℃,最适作用PH为7.5~8.0,在pH6.0~9.5和45℃以下条件下稳定,在0℃下显示了6.1%的相对活性,60℃以上热处理完全失去酶活。该酶被EDTA-2Na,Hg^2+、Cu^2+、Ni^2+、Ag^2+、Co^2+及Pepstatin A不完全抑制。Zn^2+对蛋白酶具有明显的激活作用。纯化酶作用于偶氮酪蛋白的Km和Vmax分别为0.098%和72mg/(h·mg)。该酶为N末端VGSEVTLKNS的明胶酶(Gelatinase)的一种,性质属于低温蛋白酶。  相似文献

Female damselflies in the family Calopterygidae have two sperm storage organs: a spherical bursa copulatrix and a tubular spermatheca. Male flies have a peculiar aedeagus with a recurved head with which to remove bursal sperm, and lateral spiny processes to remove spermathecal sperm. The lateral processes differ among species and populations in terms of their width relative to the spermathecal duct: the narrower processes are physically able to access spermathecal sperm, while the wider ones are not. In the present study, sperm storage patterns and aedeagal structures were compared between two calopterygid species with different spermathecal structures –Calopteryx cornelia and Mnais pruinosa– with respect to not only sperm quantity (number) but also sperm quality (viability), by using a recently developed method based on live/dead dual fluorescence. Calopteryx cornelia is a typical spermathecal sperm remover. In this species, viability was similar between bursal and spermathecal sperm. In contrast, in M. pruinosa, the spermatheca was much smaller than the bursa and often contained no sperm. Even when the spermatheca of this species did contain sperm, a high percentage of it was dead. Although the spermatheca of M. pruinosa has such atrophic tendencies, males have nevertheless developed long and spiny lateral processes similar to those of C. cornelia, suggesting the processes have functions other than spermathecal sperm removal. They possibly function as stoppers or guides for manipulating the aedeagal head to remove the sperm mass from the bursa.  相似文献

Recent studies show advancing onset of plant growing season in many regions for the last several decades. With the well‐established dependence of plant phenology on temperature, these trends are interpreted as an indication of global warming. For several decades, however, other determinants of plant phenology, e.g. varieties and trends in managed systems, may have changed and confounded the phenological trends. In this study, we tested if long‐term changes in phenology of apple (Malus pumila var. domestica) are attributable to long‐term changes in temperature by comparing the phenological response to long‐term trend in air temperature, which is of our interest, with that to year‐to‐year fluctuation in air temperature, which should represent the real effect of temperature on phenology. We collected records of air temperature and phenological events (budding and flowering) in apple from 1977 to 2004 at six locations in Japan. Linear trends in flowering showed advancing rate in the range from 0.21 to 0.35 day yr?1, statistically significant at three locations (P<0.05). We also found a warming trend in mean air temperature throughout March and April, with which flowering was closely correlated, in the range from 0.047 to 0.077 °C yr?1, statistically significant at five locations (P<0.05). We separated the temperature time‐series into two components: a long‐term trend and a year‐to‐year fluctuation, by fitting smoothing spline to the trend and taking the residuals as the anomaly. We then fit a multiple regression model of phenological response to air temperature with separate coefficients for long‐term trend and anomaly. Flowering date responded to the long‐term trend at ?3.8 day °C?1 and to the anomaly at ?4.6 day °C?1. The temperature coefficients were not statistically different from each other or among locations, suggesting that the advance of apple phenology has predominantly been caused by the temperature increase across the locations studied. The early processes of proventricular gland formation in the chick embryo were investigated. The glands appeared as intra-epithelial invaginations of the proventricular endoderm on day 6 of incubation. By day 6.5 they began to protrude into the mesenchyme and elongated without branching until day 9. Before elongation of the glands, the immunofluorescence of laminin and the ultrastructure of the basal lamina were consistently observed in the intra-epithelial invaginations as well as in other regions, and the mitotic activity in the gland rudiments was not different from that in other regions. However, at the tips of the elongating glands, little laminin was detected and the basal lamina were thin and discontinuous. The mitotic activity at the tip of the glands was higher than that in non-glandular epithelium or in the stalk of the glands. These results suggest causal relationships between thinning of the basement membrane and localized epithelial cell proliferation at the tip of the elongating glands.  相似文献

Using the free‐air CO2 enrichment (FACE) techniques, we carried out a 3‐year mono‐factorial experiment in temperate paddy rice fields of Japan (1998–2000) and a 3‐year multifactorial experiment in subtropical paddy rice fields in the Yangtze River delta in China (2001–2003), to investigate the methane (CH4) emissions in response to an elevated atmospheric CO2 concentration (200±40 mmol mol?1 higher than that in the ambient atmosphere). No significant effect of the elevated CO2 upon seasonal accumulative CH4 emissions was observed in the first rice season, but significant stimulatory effects (CH4 increase ranging from 38% to 188%, with a mean of 88%) were observed in the second and third rice seasons in the fields with or without organic matter addition. The stimulatory effects of the elevated CO2 upon seasonal accumulative CH4 emissions were negatively correlated with the addition rates of decomposable organic carbon (P<0.05), but positively with the rates of nitrogen fertilizers applied in either the current rice season (P<0.05) or the whole year (P<0.01). Six mechanisms were proposed to explain collectively the observations. Soil nitrogen availability was identified as an important regulator. The effect of soil nitrogen availability on the observed relation between elevated CO2 and CH4 emission can be explained by (a) modifying the C/N ratio of the plant residues formed in the previous growing season(s); (b) changing the inhibitory effect of high C/N ratio on plant residue decomposition in the current growing season; and (c) altering the stimulatory effects of CO2 enrichment upon plant growth, as well as nitrogen uptake in the current growing season. This study implies that the concurrent enrichment of reactive nitrogen in the global ecosystems may accelerate the increase of atmospheric methane by initiating a stimulatory effect of the ongoing dramatic atmospheric CO2 enrichment upon methane emissions from nitrogen‐poor paddy rice ecosystems and further amplifying the existing stimulatory effect in nitrogen‐rich paddy rice ecosystems.  相似文献

The ground beetle Leptocarabus procerulus (Chaudoir) possesses seminal substances that have a physical function to form mating plugs and a physiological function to induce female refractory behaviour, which act together to hinder female remating. Little is known about the physiological properties of the substances inducing female refractory behaviour, especially with respect to their secretory organ, dose‐dependency, molecular characteristics and the effect of female maturity. By injecting male‐derived substances into females, substances that induce female refractory behaviour are shown to be produced in the male accessory gland but not in the testis. Injection of extracts from the accessory gland increases the female refractory period at moderate doses but not at lower or higher doses. By contrast, injection of extracts from the testis reduces the female refractory period at high doses. The lack of an effect of accessory gland substances at higher doses could be the result of an anomalous effect of unnaturally large doses of seminal products by direct injection, the toxicity of seminal substances that deter female responses, or counteraction by injected substances that promote female receptivity. The accessory gland substances lose their activity when heated, although the testis substances do not. Females without mature eggs tend to reject mating entirely, although variation in the number of mature eggs (one or more) is not associated with the female refractory period, indicating the limited effect of female reproductive maturity. These findings may help to clarify the physiological basis of the evolution of the elaborated male mating behaviour in L. procerulus.  相似文献

  • 1 The female weevil of Apoderus balteatus Roelofs cuts the leaf near its base in order to make a leaf-roll‘cradle', in which one, two or three eggs are laid. Cuts are always located about 5 mm from the leaf base, independent of leaf size.
  • 2 Larger leaves are made into larger cradles.
  • 3 Weevils tend to lay more eggs in large cradles.
  • 4 Eggs are laid through different holes and separated by leaf layers in the cradle. Cannibalism by larvae will be prevented by this isolated oviposition.
  • 5 Multiple oviposition of this weevil allows larger leaves to be used by multiple larvae. It facilitates more efficient use of larger leaves and economizes on female effort in cradle formation.
