SYNOPSIS. Autoclavable, natural particulate media simplify axenic cultivation of tetrahymenid ciliates and presumably favor selection for phagotrophy. Viability is at least 2 months at room temperature (24–26 C) for the lipid-sensitive tetrahymenids Tetrahymena setosa, T. corlissi, T. paravorax, T. limacis, and T. patula, also for T. rostrata and (at 12 C), for strains of the T. pyriformis complex and Glaucoma chattoni. A typical medium consists of crude soy “lecithin”+ skim milk powder +Saccharomyces cerevisiae cells. Other useful particules readily available commercially are: whole liver powder, cells of Micrococcus lysodeikticus and Escherichia coli, and powdered residue of liver which had been extracted with 70% ethanol (liver #2). Preliminary experiments indicate that some of these media are suitable for the maintenance of Paramecium octaurelia stock 299S and Colpidium campylum. Such mixtures may serve as points of departure for devising media for more fastidious phagotrophs.  相似文献

Potential vectors of Leishmania braziliensis Vianna were assessed at four study sites in the mountainous Valle del Cauca, western Colombia, from March to June 1989. In an active focus of transmission at 1450 m altitude, a coffee plantation at Versalles, there were high densities of antropophilic phlebotomines: Lutzomyia columbiana (Ritorcelli & Van Ty) and Lu.townsendi (Ortiz), both in the verrucarum species group, and of Lu.pia (Fairchild & Hertig). At a comparable altitude in a forest reserve at Yotoco where leishmaniasis is unknown, Lu.pia was the prevalent species and Lu.townsendi was absent. In two localities at 1150 m altitude, there were plentiful Lu.lichyi (Floch & Abonnenc) plus both species in the verrucarum group, but Lu.pia was absent. One of these localities, a coffee plantation at Villa Hermosa where a leishmaniasis outbreak occurred in 1986, was compared with a leishmaniasis-free, partly wooded nature reserve at Mateguadua. No natural infections of Leishmania were found in a total of 1896 wild-caught female phlebotomines belonging to at least seven species. It remains unclear why Leishmaniasis transmission is associated with coffee plantations in this part of Colombia. Laboratory-bred Lu.lichyi females were invariably autogenous, and blood-seeking females of this species were always parous. Parity rates in wild-caught females of other species were 55% Lu.pia, 24% Lu.columbiana and 14% Lu.townsendi. Female Lutzomyia infected artificially with Le.braziliensis promastigotes developed peripylarian infections. Higher proportions of Lu.townsendi (96%) and Lu.columbiana (78%) became infected but these species developed lower rates of stomodaeal infections (P less than 0.1) than Lu.lichyi (37%) or Lu.pia (44%).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

1. Phyllosphere interactions are known to influence a variety of tree canopy community members, but less frequently have they been shown to affect processes across ecosystem boundaries. Here, we show that a fungal endophyte (Rhytisma punctatum) slows leaf litter decomposition of a dominant riparian tree species (Acer macrophyllum) in an adjacent stream ecosystem. 2. Patches of leaf tissue infected by R. punctatum show significantly slower decomposition compared to both nearby uninfected tissue from the same leaf, and completely uninfected leaves. These reduced rates of decomposition existed despite 50% greater nitrogen in infected tissues and may be driven by slower rates of decomposition for fungal tissues themselves or by endophyte–hyphomycete interactions. 3. Across a temperate forest in the Pacific Northwest, approximately 72% of all A. macrophyllum leaves were infected by R. punctatum. Since R. punctatum infection can influence leaf tissue on entire trees and large quantities of leaf litter at the landscape scale, this infection could potentially result in a mosaic of ‘cold spots’ of litter decomposition and altered nutrient cycling in riparian zones where this infection is prevalent.  相似文献

1. Female feeding regime exhibited a cascade of effects on reproductive biology and behaviour in the mantid Stagmomantis limbata (Hahn). 2. Well‐fed females (High diet) consistently attained greater body mass, thicker abdomens, and higher fecundity than food‐limited females (Low diet) in four annual generations. Dorso‐ventral abdominal thickness strongly correlated with fecundity. 3. In an experiment in captivity, Low diet females were more likely to cannibalize males than High diet females. 4. High diet females attracted more males than Low diet females in several contexts. In terms of long‐range attraction, caged High diet females attracted more males in the field. In terms of short‐range attraction, males preferentially mounted and copulated with High diet females in captive paired choice trials. In naturally‐occurring pairs in the field, a preference for females with thick abdomens was evident. These results point to two possible explanations: male choice for well‐fed females and state‐dependent female pheromone emission. 5. Two years of field data indicate considerable variation in female feeding success, as measured by abdominal thickness. Average feeding success in nature appears to be relatively high, when comparing the rate of abdominal expansion in the field to captive females maintained on High diets. 6. In the field, nearly all mounts and copulations occurred in September in both years, when female feeding success is expected to be high. If mating activity occurs during a window of high female feeding success, so as to reduce the risk of cannibalism for males, then the female feeding regime may be implicated in the phenology of mating activity in cannibalistic species.  相似文献

A complex signal transduction network involving salicylic acid, jasmonic acid and ethylene underlies disease resistance in Arabidopsis. To understand this defence signalling network further, we identified mutants that expressed the marker gene PR-1::luciferase in the absence of pathogen infection. These cir mutants all display constitutive expression of a suite of defence-related genes but exhibit different disease resistance profiles to two biotrophic pathogens, Pseudomonas syringae pv. tomato and Peronospora parasitica NOCO2, and the necrotrophic pathogen Botrytis cinerea. We further characterized cir3, which displays enhanced resistance only to the necrotrophic pathogen. Cir3-mediated resistance to B. cinerea is dependent on accumulated salicylic acid and a functional EIN2 protein.  相似文献

This review covers four areas of clinical importance to practicing psychiatrists: a) symptoms and course of uncomplicated (normal) grief; b) differential diagnosis, clinical characteristics and treatment of complicated grief; c) differential diagnosis, clinical characteristics and treatment of grief-related major depression; and d) psychiatrists’ reactions to patient suicides. Psychiatrists often are ill prepared to identify complicated grief and grief-related major depression, and may not always be trained to identify or provide the most appropriate course of treatment. Both conditions overlap with symptoms found in ordinary, uncomplicated grief, and often are written off as “normal” with the faulty assumption that time, strength of character and the natural support system will heal. While uncomplicated grief may be extremely painful, disruptive and consuming, it is usually tolerable and self-limited and does not require formal treatment. However, both complicated grief and grief-related major depression can be persistent and gravely disabling, can dramatically interfere with function and quality of life, and may even be life threatening in the absence of treatment; and both usually respond to targeted psychiatric interventions. In addition, patient suicide has been reported as one of the most frequent and stressful crises experienced by health providers, and psychiatrists are not immune to complicated grief or grief-related depression when they, themselves, become survivors. Thus, it is essential for psychiatrists to recognize their own vulnerabilities to the personal assaults that often accompany such losses, not only for their own mental health and well-being, but also to provide the most sensitive and enlightened care to their patients.  相似文献

The first change in the sequence of morphological events occurring as fully developed Ichthyophthirius multifiliis trophonts spontaneously left gill epithelium or as younger trophonts departed, following experimentally induced death of the fish, was the separation of parasites from overlying host cells. Discharge of contractile vacuoles may have played a role in this process. Spaces then appeared between host cells, and individual epithelial cells became vacuolated. Finally, the epithelium ruptured and the parasites swam free. In induced exit after three days residence in the host, departure of the trophont was evident only after autolysis of epithelium had occurred. Induced departure of trophonts after four days residence was more rapid, suggesting an active role for the parasite in exit. Changes in parasite and epithelium observed in induced exit were similar to those in spontaneous departure after five days residence.  相似文献

The anomalous secondary growth was studied in roots and stemsof two species of Bougainvillea. The anomalous cambia arisesuccessively in centrifugal order, each originating among thederivatives of the preceding cambium. Each cambium layer functionsbidirectionally producing xylem towards the inside of the axisand phloem towards the outside. The sequence of production ofvascular cells is the following: (1) conjunctive tissue andxylem fibres towards the inside; (2) phloem towards the outside;(3) additional xylem with vessels towards the inside and additionalphloem towards the outside. The new cambia arise outside theoldest phloem cells of a given increment. This phloem may benonfunctional and crushed at that time. The phloem and the xylemdifferentiate from radially seriated derivatives produced sequentiallyby tangential divisions in the cambium. Divisions among thephloem initials and growth readjustments in the differentiatingxylem obscure the radial seriation to a moderate extent.  相似文献
