To study the role of type III-secreted effectors in the host adaptation of the tobacco ( Nicotiana sp.) pathogen Pseudomonas syringae pv. tabaci , a selection of seven strains was first characterized by multilocus sequence typing (MLST) to determine their phylogenetic affinity. MLST revealed that all strains represented a tight phylogenetic group and that the most closely related strain with a completely sequenced genome was the bean ( Phaseolus vulgaris ) pathogen P. syringae pv. phaseolicola 1448A. Using primers designed to 21 P. syringae pv. phaseolicola 1448A effector genes, it was determined that P. syringae pv. phaseolicola 1448A shared at least 10 effectors with all tested P. syringae pv. tabaci strains. Six of the 11 effectors that failed to amplify from P. syringae pv. tabaci strains were individually expressed in one P. syringae pv. tabaci strain. Although five effectors had no effect on phenotype, growth in planta and disease severity of the transgenic P. syringae pv. tabaci expressing hopQ1-1 Pph1448A were significantly increased in bean, but reduced in tobacco. We conclude that hopQ1-1 has been retained in P. syringae pv. phaseolicola 1448A, as this effector suppresses immunity in bean, whereas hopQ1-1 is missing from P. syringae pv. tabaci strains because it triggers defences in Nicotiana spp. This provides evidence that fine-tuning effector repertoires during host adaptation lead to a concomitant reduction in virulence in non-host species.  相似文献

The possible involvement of gibberellins and amylase in heterosisof maize seedlings was investigated in two parental inbreds,CM7 and CM49, and their single cross F1 hybrid, CM7xCM49. Germinationof all three genotypes was complete within 36 h after the onsetof imbibition. By 48 h, heterosis (hybrid vigour) for increasedshoot and root length was consistently observed. The endogenousconcentration of gibberellin A1 (GA1) was measured in 48 h seedlingsby gas chromatography-mass spectrometry with selected ion monitoring(GC-SIM) using [2H2]-GA1 as an internal standard. The GA1 concentrationwas highest in the hybrid (59 ng g–1 dry wt.), intermediatein CM49 (9.0 ng g–1), and lowest in CM7 (<5.0 ng g–1).Amylase activities in all three genotypes were very low at 24h, but increased during the next 24 h, after which time amylaseactivity in the hybrid was significantly higher than that ofeither parental inbred. Inhibitors of gibberellin (GA) biosynthesis,AMO-1618 or CCC, inhibited germination, shoot and root growth,and amylase activity in all three genotypes. Conversely, exogenousgibberellic acid (GA3) increased amylase activity, particularlyin the inbred CM7. Amylase isozymes were separated through polyacrylamidegel electrophoresis and generally similar profiles of starchdegrading enzymes were observed in the three genotypes. SinceGA is known to control a-amylase biosynthesis in some cereals,these results are consistent with the hypothesis that GAs areinvolved in the regulation of heterosis in maize. A higher endogenousGA1 concentration in the hybrid could result in increased amylaseactivity in the hybrid seedlings and consequently, more rapidstarch hydrolysis which fuels heterosis for early growth. The distinction of hepatocellular carcinoma (HCC) from benign lesions of the liver in fine needle aspiration (FNA) specimens can be problematic. In an attempt to separate well-differentiated HCC from benign hepatocellular lesions, the presence of tissue fragments displaying peripheral endothelial cells (PE) has been proposed in a previous study as a useful feature in favour of malignancy. In this study, we evaluated slides from 59 cases of liver masses undergoing FNA (19 HCC, 40 benign) and evaluated them for the presence of tissue fragments containing PE. We found that 90% of cases of HCC contained tissue fragments in which PE were either focally present or abundant. However, 68% of cases containing only benign hepatocytes also contained tissue fragments in which PE were at least focally present. In addition, it appears that within the group of benign lesions, the presence of PE was related to the overall cellularity of the specimen rather than the specific nature of the lesion. Thus, the presence of PE in tissue fragments does not, in isolation, appear to be a useful morphological feature for the separation of benign and malignant hepatocellular lesions in FNA material.  相似文献

The concept of 'metabolic cold adaptation', namely that polar marine ectotherms are adapted in having an elevated basal metabolic rate, has been examined in the light of recent biochemical, physiological and ecological data for Antarctic marine organisms. It is now clear that marine invertebrates from Antarctic waters are characterized by slow growth rates, low basal metabolism and reduced annual reproductive effort, and there is thus no clear evidence of the traditional view of an elevated metabolic rate. By analogy with fish, protein synthesis rates are probably also low. This suggests that the major feature of cold adaptation is a reduction in the individual total annual energy intake in comparison with ecologically similar organisms from warm water. This allows a high standing crop of suspension feeders to develop, and low temperature is thus a significant factor in the successful widespread adoption of typical K-strategies in Antarctic marine invertebrates.  相似文献

The genus Cicer has traditionally been placed in the Vicieae, although it has a number of morphological characters which suggest that it is out of place in that tribe and that its affinities lie elsewhere. A survey of pollen morphology in Cicer , the other genera of the Vicieae, the Trifolieae and the Ononideae has been carried out to help determine the relationships of Cicer. The tribe Vicieae (without Cicer) is very homogeneous and is characterized by its long, rectangular pollen grains with small, heavily thickened endoapertures. The Trifolieae is more variable in its pollen morphology but, in general, has rather similar grains to the Vicieae although the structure of the endoaperture is different. The Ononideae and Cicer have nearly spheroidal pollen with very large, unthickened endoapertures. It therefore appears that as far as pollen characters are concerned, Cicer does not belong in the Vicieae but has much more in common with the Ononideae.  相似文献

86Rb uptake was examined in two species of unicellular greenalgae, Chlamydomonas nivalis isolated from snow, and a cellwall-less mutant of the temperate freshwater Chlamydomonas reinhardii.In C. reinhardii cells grown at 20°C and cooled rapidlyto 0°C, 86Rb uptake was abolished. Cells cooled rapidlyto –5°C in the absence of ice accumulated 86Rb veryrapidly but the time course of this uptake suggested non-selectiveaccumulation through a damaged plasmalemma. Cells grown at 8°Cwere viable, able to divide and motile; they showed no signsof cold-shock and 86Rb uptake, albeit slow, was measurable at–5°C in the absence of extracellular ice. Cells ofC. nivalis grown at 20°C were damaged at sub-zero temperaturesalthough they did show an enhanced 86Rb uptake at 0°C. Cellsgrown at 5°C were able to accumulate 86Rb from media undercooledto -5°C in the absence of extracellular ice, and again showedenhanced uptake at 0°C. After nest predation, breeding dispersal can be an effective strategy to avoid local nest predators. Furthermore, encounters with predators at a nest during the pre-laying stage may be used by parents to judge future risk, such that they may abandon a nest when a nest predator has been encountered. We studied whether the between- and within-year breeding dispersal of Northern Flickers Colaptes auratus was dependent upon the outcome of the previous nesting attempt. We also tested whether pairs presented with a model predator prior to egg-laying were more likely to abandon their nests than were pairs presented with a control model. Between years, males moved significantly further after having their nest depredated than did successful males, and females showed the same trend. However, these movements did not result in greater reproductive success. More pairs switched sites within years after having their nest depredated, but those that remained and those that moved had equal subsequent nest success. Stressful encounters with predators involving nest defence may trigger dispersal both between and within years, although reproductive benefits are unclear. The proportion of pairs abandoning nests did not differ between parents presented with control or predator models, suggesting that a single encounter with a predator is not a sufficient deterrent against continued use of a particular nest.  相似文献

The Light mutation (Blt) is a dominant allele of the b-locus on mouse chromosome 4 which causes progressive dilution of coat colour. Melanocytes within the hair follicles of mutant mice develop normally but later degenerate, due to the accumulation of a toxic product, so that the hair becomes lighter with age. Previous studies on W-locus spotting mutants, from which melanocytes are absent, have shown that melanocytes in the stria vascularis of the inner ear are essential for the development and/or maintenance of the endocochlear potential (EP) which is normally around 100 mV. In this study, physiological recordings from the ears of Light mutants were correlated with strial ultrastructure. EPs recorded from all b/b controls and young homozygous and heterozygous mutants (20–22 days old) were normal (77 to 113 mV), but were reduced (19 to 59 mV) in about 30% of ears from older mutants (Blt/Blt and Blt/b). Strial function therefore appears to develop normally but later degenerates in some mutants. This suggests that strial melanocytes are affected by the Light allele and that the continued presence of melanocytes is necessary for strial function. There was no obvious association between the recorded EP value and the ultrastructural appearance of the stria. No structural abnormalities of the stria were noted in control or mutant mice aged 20 days to 4 months including those which had a reduced EP. Strial atrophy was common in old controls and mutants (1–2 years), and appeared to be an age-related process rather than an effect of the Light mutation. Similarly, pigment build-up was common in all strial cells of old mice. However, the accumulations of lipofuscin-like pigment were much larger and more abundant in aged brown non-agouti mice than those observed in old agouti mice, which suggests that this age-related process also has a genetic component.  相似文献

1. Organisms can respond to changing climatic conditions in multiple ways including changes in phenology, body size or morphology, and range shifts. Understanding how developmental temperatures affect insect life‐history timing and morphology is crucial because body size and morphology affect multiple aspects of life history, including dispersal ability, whereas phenology can shape population performance and community interactions. 2. It was experimentally assessed how developmental temperatures experienced by aquatic larvae affected survival, phenology, and adult morphology of dragonflies [Pachydiplax longipennis (Burmeister)]. Larvae were reared under three environmental temperatures: ambient, +2.5, and +5 °C, corresponding to temperature projections for our study area 50 and 100 years in the future, respectively. Experimental temperature treatments tracked naturally‐occurring variation. 3. Clear effects of temperature were found in the rearing environment on survival and phenology: dragonflies reared at the highest temperatures had the lowest survival rates and emerged from the larval stage approximately 3 weeks earlier than animals reared at ambient temperatures. There was no effect of rearing temperature on overall body size. Although neither the relative wing nor thorax size was affected by warming, a non‐significant trend towards an interaction between sex and warming in relative thorax size suggests that males may be more sensitive to warming than females, a pattern that should be investigated further. 4. Warming strongly affected survival in the larval stage and the phenology of adult emergence. Understanding how warming in the developmental environment affects later life‐history stages is critical to interpreting the consequences of warming for organismal performance.  相似文献
