SUMMARY.
  • 1 The microbial heterotrophic utilization of dissolved organic matter (DOM) was determined experimentally in microcosms using stream water and stream-bed sediments from a third order reach of White Clay Creek (Pennsylvania, U.S.A.).
  • 2 Sources of DOM for the experiments included White Clay Creek water at baseflow and stormflow and cold water extracts of jewel weed (Impatiens capensis L.) and spicebush (Linderu henzoin (L.) Blume).
  • 3 The heterotrophic activity of the sediments was measured as uptake of the following: dissolved organic carbon (DOC), molecular weight fractions within the DOC pool, carbohydrates, amino acids and peptides, phenolics, and dissolved oxygen (DO), all in the overlying water.
  • 4 Concentrations of adenosine triphosphate (ATP), and direct microscopic counts of bacteria were used to estimate bacterial biomass in the surface sediments.
  • 5 The microcosm experiments showed that specific DOC molecular size classes and DOM functional groups were selectively removed from solution, exposure to one DOM source affected responses to a different DOM source and certain DOM sources were more readily utilized than others.
  • 6 Continued exposure to a DOM source increased microbial heterotrophic activity (a condition which persisted even after removal of the DOM source for several days).
  • 7 Rates of biotic DOC uptake ranged from 3.6 to 242.8 mg Cm-2h-1.
  • 8 Indirect estimates of biosynthesis calculated from DOC and DO data ranged from 1.6 at baseflow and 2.6–61.2 at stormflow to as high as 192.6 mg C m-2 h-1 when the community was repeatedly exposed to enriched DOM sources.

The permeability of the plasmalemma of Chlamydomonas reinhardtiicells was increased by treatment with poly-L-lysine or dimethylsulphoxideas indicated by 3-phosphoglyceric acid dependent O2 evolution.These treatments decreased the ability of the cells to accumulateinorganic carbon internally and hence their photosynthetic affinityfor inorganic carbon in the medium. With saturating light andinorganic carbon, the photosynthetic rate was less affectedby the poly-L-lysine and dimethylsulphoxide treatments. Thusthe poly-L-lysine and dimethylsulphoxide did not alter the activityof the chloroplasts but rather made the intracellular inorganiccarbon pool more freely exchangeable with the medium. It isconcluded that the transporting system for inorganic carbonis located at the plasmalemma. Treatment with Diamox, an inhibitor of carbonic anhydrase, didnot affect photosynthetic rate and accumulation of inorganiccarbon when CO2 was supplied but strongly inhibited both parameterswhen HCO3 was supplied. In a mutant of Chlamydomonasreinhardtii lacking a cell wall, carbonic anhydrase leaks tothe medium and uptake of inorganic carbon is much faster whenCO2 is supplied than when HCO3 is supplied. Abstract 1. As herbivory often elicits systemic changes in plant traits, indirect interactions via induced plant responses may be a pervasive feature structuring herbivore communities. Although the importance of this phenomenon has been emphasised for herbivorous insects, it is unknown if and how induced responses contribute to the organisation of other major phytoparasitic taxa. 2. Survey and experimental field studies were used to investigate the role of plants in linking the dynamics of foliar‐feeding insects and root‐feeding nematodes on tobacco, Nicotiana tabacum. 3. Plant‐mediated interactions between insects and nematodes could largely be differentiated by insect feeding guild, with positive insect–nematode interactions predominating with leaf‐chewing insects (caterpillars) and negative interactions occurring with sap‐feeding insects (aphids). For example, insect defoliation was positively correlated with the abundance of root‐feeding nematodes, but aphids and nematodes were negatively correlated. Experimental field manipulations of foliar insect and nematode root herbivory also tended to support this outcome. 4. Overall, these results suggest that plants indirectly link the dynamics of divergent consumer taxa in spatially distinct ecosystems. This lends support to the growing perception that plants play a critical role in propagating indirect effects among a diverse assemblage of consumers.  相似文献
