There is widespread interest in developing methods to investigate in situ microbial activity in subsurface environments. Novel experiments based on single borehole push–pull methods were conducted to measure in situ microbial activity at the Äspö Hard Rock Laboratory (HRL). Microbial nitrate reduction and lactate consumption were measured at in situ conditions at a depth of 450 m in the HRL tunnel. A circulation system was used to circulate ground water from the aquifer through pressure‐maintaining flow cells containing coupons for biofilm growth. The system allows microbial investigations at in situ pressure, temperature and chemistry. Four experiments were conducted in which a combination of a conservative tracer, nitrate and lactate was injected into the circulation system. Rate of nitrate utilization was 5 µm  h?1 without lactate and 13 µm  h?1 with lactate. Lactate consumption increased from 30  to 50 µm  h?1 with the addition of an exogenous electron acceptor (nitrate). Attached and unattached cells were enumerated using epifluorescence microscopy to calculate cell‐specific rates of activity. The biofilm had an average cell density of 1 × 106 cells cm?2 and there was an average of 6 × 105 unattached cells mL?1 in circulation. Cell‐specific rates of lactate consumption were higher than previously reported using radiotracer methods in similar environments. The differences highlight the importance of conducting microbial investigations at in situ conditions. The results demonstrate that an indigenous community of microbes survives at a depth of 450 m in the Fennoscandian shield aquifer with the potential to oxidize simple organic molecules such as lactate.  相似文献

1. Arbuscular mycorrhizal fungi (AMF) commonly colonise isoetid species inhabiting oxygenated sediments in oligotrophic lakes but are usually absent in other submerged plants. We hypothesised that organic enrichment of oligotrophic lake sediments reduces AMF colonisation and hyphal growth because of sediment O2 depletion and low carbon supply from stressed host plants. 2. We added organic matter to sediments inhabited by isoetids and measured pore‐water chemistry (dissolved O2, inorganic carbon, Fe2+ and ), colonisation intensity of roots and hyphal density after 135 days of exposure. 3. Addition of organic matter reduced AMF colonisation of roots of both Lobelia dortmanna and Littorella uniflora, and high additions stressed the plants. Even small additions of organic matter almost stopped AMF colonisation of initially un‐colonised L. uniflora, though without reducing plant growth. Mean hyphal density in sediments was high (6 and 15 m cm?3) and comparable with that in terrestrial soils (2–40 m cm?3). Hyphal density was low in the upper 1 cm of isoetid sediments, high in the main root zone between 1 and 8 cm and positively related to root density. Hyphal surface area exceeded root surface area by 1.7–3.2 times. 4. We conclude that AMF efficiently colonise isoetids in oligotrophic sediments and form extensive hyphal networks. Small additions of organic matter to sediments induce sediment anoxia and reduce AMF colonisation of roots but cause no apparent plant stress. High organic addition induces night‐time anoxia in both the sediment and the plant tissue. Tissue anoxia reduces root growth and AMF colonisation, probably because of restricted translocation of nutrient ions and organic solutes between roots and leaves. Isoetids should rely on AMF for P uptake on nutrient‐poor mineral sediments but are capable of growing without AMF on organic sediments.  相似文献

Understanding the consequences of inbreeding in combination with stress is important for the persistence of small endangered populations in a changing environment. Inbreeding and stress can influence the population at all stages of the life cycle, and in the last two decades a number of studies have demonstrated inbreeding depression for most life‐cycle components, both in laboratory populations and in the wild. Although male fertility is known to be sensitive to temperature extremes, few studies have focused on this life‐cycle component. We studied the effects of inbreeding on male sterility in benign and stressful environments using Drosophila melanogaster as a model organism. Male sterility was compared in 21 inbred lines and five non‐inbred control lines at 25.0 and 29.0 °C. The effect of inbreeding on sterility was significant only at 29.0 °C. This stress‐induced increase in sterility indicates an interaction between the effects of inbreeding and high‐temperature stress on male sterility. In addition, the stress‐induced temporary and permanent sterility showed significant positive correlation, as did stress‐induced sterility and the decrease in egg‐to‐adult viability. This suggests that the observed stress‐induced decline in fitness could result from conditionally expressed, recessive deleterious alleles affecting both sterility and viability simultaneously. © 2011 The Linnean Society of London, Biological Journal of the Linnean Society, 2011, 104 , 432–442.  相似文献

1. Ophrydium versatile is a symbiotic ciliate which forms gelatinous colonies up to several centimetres in diameter in transparent temperate lakes. The ciliates are evenly spaced at the colony surface and constitute a small proportion of the surface area (7%) and volume (3.1%) of the colony, but a large proportion of organic carbon (74%) and nitrogen content (82%) (exemplified for 1 cm3 colonies). The majority of the colony volume is formed by the jelly. The biomass proportion of ciliates scales inversely with colony size, following the decline of surface area to colony volume. The largest colonies found in Danish lakes in early summer contain almost 1 million ciliates, and assuming they derive from a single ciliate undergoing exponential division, they need twenty generations and, presumably, almost a year to reach maximum size. 2. The ciliates contain numerous symbiotic zoochlorellae that constitute about 10% of ciliate volume and more than half of the carbon content. Zoochlorellae dominate oxygen metabolism of the assemblage, resulting in low light compensation points, a large diel photosynthetic surplus, and a marked dependence on light for sustained growth and ciliate metabolism. Estimated gross photosynthesis (7ng C ciliate?1 day?1) of Ophnrydium from shallow, clear waters in June greatly exceeded the estimated carbon contained in filtered bacteria and small algae (1.9ng C cilicate?1 day?1). Nitrogen and phosphorus content of the prey, however, may provide the main nutrient source consistent with the correspondence between mass-specific rates of nutrient uptake and measured relative growth rates (average 0.067 day?1, generation time 10 days). 3. The large Ophrydium colonies require increased allocation of photosynthetic carbohydrates with increasing colony size to maintain the jelly. The large colonies tend to become gas-filled, floating, mechanically destroyed and their ciliate inhabitants abandon them as swarmers. Colony formation, however, should offer protection against predators which may be more important for the natural abundance than the costs of growing in a colony.  相似文献

In a population of Lapwings Vanellus vanellus studied for 5 years near Bergen, southwester Norway, 23–41% of the males were polygynous, having two and sometimes three mates, whereas 50–77% were monogamous and 0–21% remained unmated. Bachelors held territories in or immediately adjacent to the study area each of the years. As predicted from the Polygyny Threshold model, primary females generally laid eggs earlier than monogamous females, but the overlap in laying dates was substantial between these two classes. The number of females and start of breeding on the territories were analysed in relation to nine variables pertaining to safety from predation and to food. Of these, territory size correlated most consistently with number of females (positively) and the laying of first egg (negatively). Numbers of females observed were not significantly different from numbers expected from territory size in 4 out of 5 years; however, earlier start of breeding on large territories indicated that females did not settle according to the Neutral Mate Choice model. The Female-biased Sex Ratio hypothesis was refuted, unmated territorial males being available throughout the breeding seasons.  相似文献

1. The European Water Framework Directive provides a framework for improving the ecological quality of stream ecosystems, with deviation from reference used as a measure of ecological status.
2. Here we examine the possibility of using less impacted stream sites from Latvia, Lithuania and Poland to establish a Danish reference network for macrophyte assemblages, and as a guiding image for identification of possible references sites within Denmark. Both approaches were evaluated using historical Danish records.
3. Four different macrophyte assemblages were identified for mid-sized streams in the Central and Eastern Lowland ecoregions. Macrophyte assemblages could not be delineated using physical stream site characteristics; however a gradual change in assemblage composition was attributed to differences in alkalinity and human impact.
4. Assemblages of contemporary vegetation in Denmark were quite similar to those found in Polish, Latvian and Lithuanian streams (26–35%). However, more importantly, from species-based predictions we noted higher similarity, particularly with Latvian and Lithuanian streams, before intensive land use commenced in Denmark ( c. 1900). These results show that stream sites from these three countries can be considered in a Danish reference network.
5. Two of the four macrophyte assemblages comprised species such as Fontinalis antipyretica, Myriophyllum spicatum , Nuphar lutea , Potamogeton alpinus and P. perfoliatus that have a very scattered occurrence in the contemporary vegetation in Denmark. These groups were closely associated with the predictions from historic records, thereby lending support the conjecture that these assemblages could be part of the guiding image for the identification of potential reference sites within Denmark.  相似文献

The aim of the present study was to study the sensibility in the area of saline-induced muscle pain. In three experiments, ten subjects were exposed to computer-controlled infusion of 0.5 ml isotonic (0.9%) or hypertonic (9%) saline into the anterior tibial muscle. The pain intensity was assessed on a visual analogue scale (VAS). The pain threshold (PT) to pressure and electrical stimulation in muscle and subcutaneous tissues was determined. Three experiments were performed in which infusion of hypertonic saline produced significantly higher VAS scores than isotonic saline. In all three experiments, there was no significant difference in PT obtained after infusion of isotonic saline compared with infusion of hypertonic saline. In experiment 1, the PT was determined at the infusion site and 4 cm from the infusion site. At the infusion site, the pressure PT decreased (- 19 2%) 1, 3, 5, 7 and 9 min after infusion of isotonic and hypertonic saline, but remained unchanged 4 cm from the infusion site. The intramuscular electrical PT at the infusion site and 4 cm from the infusion site increased significantly (29 6%) 5, 7 and 9 min after saline infusion. In experiment 2, the pressure PT and the intramuscular electrical PT were recorded after two infusions of saline separated by 1 day. The day after the first infusion, the pressure PT was decreased compared with the PT before the first infusion, but the electrical PT was not affected. Moreover, the hypertonic saline infusion given on the second day produced significantly higher (130 50%) VAS scores than the infusion given on the first day. In experiment 3, the PT was determined in the subcutaneous tissue, but no significant effects of saline infusion were found. The present placebo-controlled experiments failed to show muscular or subcutaneous hyperalgesia after saline-induced muscle pain per se.  相似文献

  • 1 Invertebrate herbivory on the submerged macrophyte Potamogeton perfoliatus was studied in a Danish lowland stream during the main growth seasons of summer 1991 and 1992. Young apical leaves escaped consumption probably because of their location and dense packing, but herbivory loss increased linearly with age and exposure time of older leaves. Nitrogen content was relatively high in both young (4.61% DW) and old leaves (3.65% DW) but a food preference experiment showed that young leaves were preferred by the main herbivore, the trichopteran Anabolia nervosa.
  • 2 The percentage of standing plant biomass lost to herbivory (the apparent loss) between sampling periods increased from early May to a mid-June maximum (24.8% in 1991, 4.2% in 1992), and subsequently declined to zero within a month. The averages for the growth seasons were 10.5% in 1991 and 2.0% in 1992. Yet, the proportions of annual plant production harvested by herbivores were low and almost the same (1.3 and 1.8%) because consumption was low when plant production peaked in late summer. Consumption was almost the same early in the two years, but plant growth dynamics differed markedly and was the main factor determining apparent herbivory loss.
  • 3 Despite heavy damage early in the summer, P. Perfoliatus contributed a minor fraction (1–5%) of the trichopteran diet. This fraction appeared to be restricted by the low macrophyte biomass during early summer.
  • 4 The results emphasize that apparent herbivory loss does not estimate the harvested proportion of plant production, and that plant growth dynamics should be analysed to attain precise estimates of herbivory rates.
