Sargassum hemiphyllum is commonly found in Japan and Korea, with a variety, var. chinense, that is found distributed in the southern Chinese coast. We previously reported distinct genetic differentiation between the two taxa based on the PCR‐RFLP data of plastid RubiscoL‐S spacer. The present study aims at elucidating the phylogeographic pattern of S. hemiphyllum based on more markers in the nuclear and extranuclear genomes, with a view to reveal the occurrence of hybridization. The two allopatrically distributed taxa were found to be genetically distinct in nuclear ITS2, plastidial Rubisco (Rbc) and mitochondrial TrnW_I (Trn) spacers. Their divergence was postulated to be attributable to the vicariant event which resulted from the isolation of the Sea of Japan during the late Miocene (6.58–11.25 Mya). Divergence within both S. hemiphyllum and the chinense variety was observed based on Trn spacer, while the divergence in S. hemiphyllum was further confirmed in Rbc spacer. This divergence appears to correspond to the separation of the Japanese populations between the Sea of Japan and the Pacific that occurred around 0.92–2.88 Mya (the early Pleistocene). The presence of an ITS2 clone resembling var. chinense sequences in a Japanese population of S. hemiphyllum (JpNS) raises the possibility of the introgression of var. chinense individuals into S. hemiphyllum population. Compared to that between S. hemiphyllum and the chinense variety, hybridization among the Japanese and Korean populations of S. hemiphyllum is highly probable as all these individuals share a pool of nuclear ITS2 sequences, possibly attributable to incomplete concerted evolution of ITS2.  相似文献

Natural hybridization is common in the genus Lespedeza. No hybrids between Lespedeza leptostachya Englem. and Lespedeza capitata Michx. are formally recognized in any of the current floras, however observations in the field suggest that hybridization might occur in many of their shared habitats. Putative hybrids were compared to L. leptostachya and L. capitata using morphological measurements and screened for the presence of species-specific trnL-F gene region (cpDNA) and the ITS gene region (nrDNA). A discriminate analysis of 10 morphological measurements identified the hybrids as intermediate to both parents with two PCA axes explaining 99% of the variation between taxa. The presence of hybrids was confirmed by genetic markers with individuals morphologically identified as hybrids having cpDNA trnL-F genotypes identical to L. leptostachya and the ITS (nrDNA) phenotypes in most cases contain the ITS genotype of both parents, however, some putative hybrid individuals contained the ITS genotype of only one parents. Those individuals with L. leptostachya ITS and trnL-F could be a case of misclassification, but the presence of both L. capitata ITS genotypes and L. leptostachya trnL-F genotypes suggest segregation has occurred, which may result from either selfing or backcrossing.  相似文献

An in situ mesocosm experiment was performed at neutral pH Lake O'Woods, West Virginia, to assess the impacts of acidification on the common planktonic rotifer Keratella cochlearis. This rotifer is typically replaced by K. taurocephala during the acidification of North American lakes. Despite a rapid pH reduction in this experiment (from 7.0 to 4.8 in 14 days), the abundance, mean body length and egg ratio of K. cochlearis did not decline in the acid treatment as compared to the untreated control. These results support the hypothesis that K. cochlearis is acid-tolerant, and suggest that its disappearance from acid lakes is the result of biotic interactions within the plankton, rather than the result of toxic effects of altered water chemistry.  相似文献

If sufficient seedling establishment can be achieved, seed-based restoration provides an affordable, active restoration approach that can be implemented quickly at scale. However, establishment has served as a major restoration bottleneck, highlighting the need for improved understanding of seed germination niche and interactions with site conditions. Germination niche breadth (NB) is expected to increase with gene flow, resulting in broader environmental tolerance range, reduced sensitivity to site conditions, and less variation among seed sources. To investigate how germination NB relates to inter- and intraspecific variation in establishment from seed, we compared field recruitment for two milkweeds (Asclepias), the larval host plant of the monarch butterfly and thus a high priority group for habitat creation. Consistent with species-level NB derived from laboratory trials, there was strong evidence that early life stages of the habitat specialist (Asclepias incarnata) varied among seed collection regions (separated by 423–572 km) but no evidence that the generalist (A. syriaca) varied among seed sources collected across an approximately 750-km transect. Regeneration trends demonstrate that A. incarnata is significantly more sensitive to seed source and therefore requires more restricted seed zones. However, climate change may necessitate that we separate seed collection zones from seed application zones, upending the traditional framework of seed transfer zones. Until taxon-specific studies have identified the scale of adaptive, phenotypic variance, restoration practitioners should continue to adjust the scale of seed collection zones for milkweeds and other taxa based on species traits known to influence gene flow, such as abundance and habitat specificity.  相似文献

Many animal lineages exhibit allometry in sexual size dimorphism (SSD), known as ‘Rensch’s rule’. When applied to the interspecific level, this rule states that males are more evolutionary plastic in body size than females and that male‐biased SSD increases with body size. One of the explanations for the occurrence of Rensch’s rule is the differential‐plasticity hypothesis assuming that higher evolutionary plasticity in males is a consequence of larger sensitivity of male growth to environmental cues. We have confirmed the pattern consistent with Rensch’s rule among species of the gecko genus Paroedura and followed the ontogeny of SSD at three constant temperatures in a male‐larger species (Paroedura picta). In this species, males exhibited larger temperature‐induced phenotypic plasticity in final body size than females, and body size and SSD correlated across temperatures. This result supports the differential‐plasticity hypothesis and points to the role phenotypic plasticity plays in generating of evolutionary novelties.  相似文献

Entomopathogenic fungi infect insects via penetration through the cuticle, which varies remarkably in chemical composition across species and life stages. Fungal infection involves the production of enzymes that hydrolyse cuticular proteins, chitin and lipids. Host specificity is associated with fungus–cuticle interactions related to substrate utilization and resistance to host‐specific inhibitors. The soil fungus Conidiobolus coronatus (Constantin) (Entomophthorales: Ancylistaceae) shows virulence against susceptible species. The larvae and pupae of Calliphora vicina (Robineau‐Desvoidy) (Diptera: Calliphoridae), Calliphora vomitoria (Linnaeus), Lucilia sericata (Meigen) (Diptera: Calliphoridae) and Musca domestica (Linnaeus) (Diptera: Muscidae) are resistant, but adults exposed to C. coronatus quickly perish. Fungus was cultivated for 3 weeks in a minimal medium. Cell‐free filtrate, for which activity of elastase, N‐acetylglucosaminidase, chitobiosidase and lipase was determined, was used for in vitro hydrolysis of the cuticle from larvae, puparia and adults. Amounts of amino acids, N‐glucosamine and fatty acids released were measured after 8 h of incubation. The effectiveness of fungal enzymes was correlated with concentrations of compounds detected in the cuticles of tested insects. Positive correlations suggest compounds used by the fungus as nutrients, whereas negative correlations may indicate compounds responsible for insect resistance. Adult deaths result from the ingestion of conidia or fungal excretions.  相似文献

1. An in situ experiment was performed in the littoral zone of a large, subtropical lake to quantify effects of phosphorus (P) and nitrogen (N) on algal biomass, productivity, nutrient content and phosphate uptake kinetics. 2. We hypothesized that resident periphyton rapidly sequester added nutrients from the water column, but once a certain threshold is reached, nutrients remain in the water and permit a shift to a phytoplankton-dominated community. 3. Three duplicate sets of 1.2-m diameter mesocosms were treated with 10, 20 or 50 μg P L??1 in combination with 100, 200 or 500 μg N L??1, respectively. The nutrients were added thrice weekly for 14 days, after which the treatment doses were doubled for an additional 9 days. The cumulative amounts of P and N added over the course of the study were 700 and 7000 μg L??1, respectively. Two untreated mesocosms and two open reference sites were used as controls. 4. The total P concentration in the water column of nutrient-treated mesocosms remained low, even after prolonged high dosing. However, there was a two-fold increase in the P content of surface algal mats and epiphyton. This indicates that some of the added P was sequestered by those components of the community. In contrast, metaphyton and epipelon displayed little or no increase in their P content. Large quantities of added P could not be accounted for in the periphyton community, and may reflect unmeasured losses to the sediments or other pools. 5. Nitrogen also was depleted from the water column, but there were no significant increases in periphyton N content. Much of the added N could not be accounted for in mass balances, and may have been lost from the mesocosms through volatilization or other biochemical processes. 6. Chlorophyll-a in epiphyton increased significantly after 14 days in the highest nutrient treatment, where there also was a proliferation of Spirogyra on day 28. 7. On day 28, water column samples from the highest nutrient treatment also displayed a significantly higher rate of carbon uptake, and a significantly higher concentration of midday dissolved oxygen. 8. The hypothesis that phytoplankton become dominant at high nutrient loading rates was not supported. However, there were dramatic changes in community structure (increased dominance by epiphytic Spirogyra) and function (increased productivity and dissolved oxygen) in response to nutrient additions.  相似文献

During summer, Chaoborus punctipennis larval densities in the water column of fishless, eutrophic Triangle Lake become very high, and coincidently, the spined loricate rotifer Kelfcottia bostoniensis becomes the dominant zooplankter. Research was done to test the hypothesis that selective predation by Chaoborus on soft-bodied rotifers controls species dominance in the mid-summer zooplankton of this lake. In situ predation experiments showed positive selection by Chaoborus for the soft-bodied Synchaeta oblonga, negative selection for K. bostoniensis, and intermediate selection for Polyarthra vulgaris, a species with rapid escape tactics. However, during a 21 day in situ mesocosm experiment, zooplankton dominance and succession in Chaoborus-free enclosures was identical to that in enclosures with Chaoborus at lake density. Despite the selective predation, Chaoborus larvae may not exert significant top-down control on rotifers, whose intense reproductive output during mid-summer in temperate eutrophic lakes results in new individuals at rates that exceed predatory losses.  相似文献

The relationships between genome size and the systematic and evolutionary patterns in vascular plants are equivocal, although a close relationship between genome size and evolutionary patterns has been previously reported. However, several studies have also revealed the dynamic nature of genome size evolution and its considerable ‘ups’ and ‘downs’. Thus, in this study, the phylogenetic relationships among three previously revealed genome size groups and among species of the highly polyploid genus Curcuma were evaluated using AFLP. Our results suggest two main lineages within Indian Curcuma reflecting evolution of genome size. The first one includes hexaploids and higher polyploids of the previously recognized genome size group I, and the second one includes mainly hexaploids of genome size groups II and III. Within genome size group I, relationships among species seem to be influenced by reticulate evolution and higher polyploids are likely to be of allopolyploid origin. Reproductive systems in Indian Curcuma vary considerably among ploidy levels and these differences considerably affect morphological and genetic variation. In general, clonally reproducing species are expected to exhibit low genotypic diversity, but, at the same time, species of allopolyploid origin are expected to maintain higher levels of heterozygosity compared with their progenitors. We investigated intra‐populational genetic variability in Curcuma spp. to evaluate whether mode of reproduction or ploidy represent the main factor influencing the degree of genetic diversity. We found that hexaploid species exhibited significantly higher genetic diversity than higher polyploids (9x, 15x). Our results suggest that this genetic diversity pattern is largely influenced by the mode of reproduction, as higher polyploids reproduce exclusively vegetatively, whereas hexaploids reproduce mainly sexually. © 2011 The Linnean Society of London, Botanical Journal of the Linnean Society, 2011, 165 , 388–401.  相似文献
