Parameter estimation and model calibration are key problems in the application of biofilm models in engineering practice, where a large number of model parameters need to be determined usually based on experimental data with only limited information content. In this article, identifiability of biokinetic parameters of a biofilm model describing two-step nitrification was evaluated based solely on bulk phase measurements of ammonium, nitrite, and nitrate. In addition to evaluating the impact of experimental conditions and available measurements, the influence of mass transport limitation within the biofilm and the initial parameter values on identifiability of biokinetic parameters was evaluated. Selection of parameters for identifiability analysis was based on global mean sensitivities while parameter identifiability was analyzed using local sensitivity functions. At most, four of the six most sensitive biokinetic parameters were identifiable from results of batch experiments at bulk phase dissolved oxygen concentrations of 0.8 or 5 mg O(2)/L. High linear dependences between the parameters of the subsets (KO2,AOB,muAOB) and (KO2,NOB,muNOB) resulted in reduced identifiability. Mass transport limitation within the biofilm did not influence the number of identifiable parameters but, in fact, decreased collinearity between parameters, especially for parameters that are otherwise correlated (e.g., muAOB) and KO2,AOB, or muNOB and KO2,NOB). The choice of the initial parameter values had a significant impact on the identifiability of two parameter subsets, both including the parameters muAOB and KO2,AOB. Parameter subsets that did not include the subsets muAOB and KO2,AOB or muNOB and KO2,NOB were clearly identifiable independently of the choice of the initial parameter values. 相似文献

A system is described for the rapid generation of Bombyx mori nucleopolyhedrovirus (BmNPV)-based expression vectors. A series of novel BmNPV genomes, that include a mini-F replicon and therefore can be maintained in Escherichia coli, have been generated. These genomes lack a portion of the essential ORF1629 gene and cannot replicate independently in insect cells. However, they can be used as parental genomes for the generation of expression vectors by cotransfection with a transfer plasmid that includes an intact ORF1629. Only recombinant viruses that have acquired the ORF1629 gene from the transfer vector, and have therefore also acquired the foreign gene of interest, can replicate after cotransfection. Parental genomes with and without a polyhedrin gene are described, enabling the generation of occlusion-positive and occlusion-negative recombinant viruses. Occlusion-positive expression vectors enable the oral infection of B. mori larvae and can therefore be used for the mass production of a foreign protein in infected insects. 相似文献

The lymphatic vascular system plays an important role in tissue fluid homeostasis. Lymphedema is a chronic, progressive, and incurable condition that leads to lymphatic fluid retention; it may be primary (heritable) or secondary (acquired) in nature. Although there is a growing understanding of lymphedema, methods for the prevention and treatment of lymphedema are still limited. In this study, we investigated differential protein expressions in sham‐operated and lymphedema‐operated mice for 3 days, using two‐dimensional gel electrophoresis (2‐DE) and mass spectrometry analysis. Male improved methodology for culturing noninbred (ICR) mice developed lymphedema in the right hindlimb. Twenty functional proteins were found to be differentially expressed between lymphedema induced‐right leg tissue and normal left leg tissue. Out of these proteins, the protein levels of apolipoprotein A‐1 preprotein, alpha‐actinin‐3, mCG21744, parkinson disease, serum amyloid P‐component precursor, annexin A8, mKIAA0098 protein, and fibrinogen beta chain precursor were differentially upregulated in the lymphedema mice compared with the sham‐operated group. Western blotting analysis was used to validate the proteomics results. Our results showing differential up‐regulation of serum amyloid P‐component precursor, parkinson disease, and apolipoprotein A‐1 preprotein in lymphedema model over sham‐operated model suggest important insights into pathophysiological target for lymphedema.
