Reference charts are valuable tools for clinicians in their daily work on pediatric clinics. Reference charts are often constructed by smoothing techniques, and in this paper we present a newly developed non-parametric test for comparing these charts. We illustrate the method by an example. The worked example compares cross-sectional data on height in children from two Danish studies from 1970 and 1990, respectively. The test compares the average height over the range of ages for the two groups.  相似文献

Fluorescence spectroscopy Excitation Emission Matrix (EEM) measurements were applied on human blood plasma samples from a case control study on colorectal cancer. Samples were collected before large bowel endoscopy and included patients with colorectal cancer or with adenomas, and from individuals with other non malignant findings or no findings (N = 308). The objective of the study was to explore the possibilities for applying fluorescence spectroscopy as a tool for detection of colorectal cancer. Parallel Factor Analysis (PARAFAC) was applied to decompose the fluorescence EEMs into estimates of the underlying fluorophores in the sample. Both the pooled score matrix from PARAFAC, holding the relative concentrations of the derived components, and the raw unfolded spectra were used as basis for discrimination models between cancer and the various controls. Both methods gave test set validated sensitivity and specificity values around 0.75 between cancer and controls, and poor discriminations between the various controls. The PARAFAC solution gave better options for analyzing the chemical mechanisms behind the discrimination, and revealed a blue shift in tryptophan emission in the cancer patients, a result that supports previous findings. The present findings show how fluorescence spectroscopy and chemometrics can help in cancer diagnostics, and with PARAFAC fluorescence spectroscopy can be a potential metabonomic tool.

Bile acid sequestrants have been shown to reduce glucose levels in patients with type 2 diabetes. We previously reported that the bile acid sequestrant colesevelam HCl (Welchol) (COL) induced the release of glucagon-like peptide (GLP)-1 and improved glycemic control in insulin-resistant rats. In the present study, we tested whether adding sitagliptin (Januvia) (SIT), which prolongs bioactive GLP-1 half life, to COL would further enhance glycemic control. Male Zucker diabetic fatty (ZDF) rats were assigned to four groups: diabetic model without treatment (the model), the model treated with 2% COL or 0.4% (120 mg/day) SIT alone, or with the combination (COL+SIT). After 4 wk of treatment, the glucose area under the curve (AUC) was reduced more in the COL+SIT than the COL although both groups showed decreased glucose AUC with increased AUC of bioactive GLP-1 (GLP-1A) compared with the model group. The above changes were not observed after 8 wk. Increasing the SIT dose by 50% (180 mg SIT/day) in the diet reduced the glucose AUC in the COL+SIT group even after 8 wk but still not in the SIT alone group compared with the model. It was noteworthy that, after 8 wk, insulin levels in the SIT group declined to levels similar to the model. Histological examination of the pancreatic β-cell islets showed that islet sizes were larger, proliferation enhanced, and cell apoptosis reduced in the COL+SIT but not the SIT alone group compared with the model. We hypothesize that the combination of COL with SIT extends the half life of COL-induced GLP-1A and benefits preservation of the islets that delay the development of diabetes and improve glycemic control. This study suggests that the combined therapy (COL+SIT) is more effective than either drug alone for reducing glucose levels in diabetes.  相似文献

The objective of this study was to investigate piglet use of the creep area, comparing litters of sows with a high vs. low breeding value for piglet survival in the first 5 days postpartum, that were either housed in crates or individual pens during farrowing and lactation. Seventy-five Yorkshire × Danish Landrace sows were video recorded for 4 days after farrowing, and the analysis was conducted using instantaneous sampling every 10 min commencing 24 h after the birth of the first piglet for a period of 72 h. Breeding value for piglet survival had no effect on piglet use of the creep area or time spent in any location of the farrowing environment. Farrowing environment had significant effects on piglet location; during all days there were significantly more piglets in the creep area in the crates compared to the pens (P < 0.01), and this difference was larger at 24–48 h than at 49–72 h and at 73–96 h after birth (P < 0.05). Piglets in pens spent significantly more time resting near the sow, excluded nursing (P < 0.001), and this percentage decreased over time after farrowing (P < 0.001) in both the crates and the pens. In conclusion, piglet use of the creep area was higher in the crate compared to the pen particularly during the second day of life. This may partly be due to a much larger proportion of uncomfortable, slatted floor in the crates, and the shorter distance from the sow to the creep area in the crate.  相似文献

Peptides 18(9), 1511–1516, 1997.—Tachykinins are thought to be involved in extrinsic control of motility in the gastrointestinal tract. Using the isolated perfused porcine antrum with intact vagal innervation, we studied the effects of substance P, neurokinin A and capsaicin infusion, and electrical stimulation of the vagus nerves on antral motility without or with infusion of non-peptide antagonists for NK-1 receptors (CP96345) and NK-2 receptors (SR48968). Substance P and neurokinin A stimulated antral motility in a dose-dependent manner. The effect could be inhibited by atropine or a combination of the NK-1 and NK-2 receptor antagonists. Electrical stimulation of the vagus nerves and infusion of capsaicin (10−5 M) stimulated antral motility. Vagally induced motility was not influenced by infusion of CP96345 and SR48968, whereas the effect of capsaicin was blocked. We conclude that tachykinins may be involved in regulation of antral motility through sensory nerves in the porcine antrum, but they do not seem to be involved in vagal regulation of antral motility.  相似文献

Nitrogen (N) deposition may affect litter decomposition and may thus have an impact on the rate of carbon (C) sequestration in Sphagnum peatlands. We present results from four separate experiments aimed at delineating the effects of litter N-enrichment, Sphagnum species, stem part of Sphagnum , and place of incubation on decomposition rate and N release. We measured mass loss and N loss from litterbags incubated at 10-15 cm in the field for one year.
Mass loss was positively related to the N/C quotient of the litter, but depended strongly on the range in N/C quotients observed; only a distinct difference in N/C quotients affected mass loss. Although hummock species decayed at a slower rate than hollow species, the differences between the species became less pronounced for old stem parts and for N-enriched litter. Old stem parts decayed at a slower rate than young stem parts, except for S. papillosum . Neither position of incubation (low hummock or hollow), nor the inorganic N concentration of the incubation environment affected mass loss. N loss was mainly determined by, and positively related to, the N/C quotient of the litter; species and stem part had minor effects. Above a N/C quotient of about 0.015, net N loss was observed for all species.
We conclude that decomposition of Sphagnum is stimulated by N deposition. As the latter also affects litter N concentration and thus N release, we think that positive feedbacks through changing litter quality should be taken into account when modelling the effects of N deposition on Sphagnum peatlands and C sequestration in these systems.  相似文献

Objective To determine whether screening Danish men aged 65 or more for abdominal aortic aneurysms reduces mortality.Design Single centre randomised controlled trial.Setting All five hospitals in Viborg County, Denmark.Participants All 12 639 men born during 1921-33 and living in Viborg County. In 1994 we included men born 1921-9 (64-73 years). We also included men who became 65 during 1995-8.Interventions Men were randomised to the intervention group (screening by abdominal ultrasonography) or control group. Participants with an abdominal aortic aneurysm > 5 cm were referred for surgical evaluation, and those with smaller aneurysms were offered annual scans.Outcome measures Specific mortality due to abdominal aortic aneurysm, overall mortality, and number of planned and emergency operations for abdominal aortic aneurysms.Results 4860 of 6333 men were screened (attendance rate 76.6%). 191 (4.0% of those screened) had abdominal aortic aneurysms. The mean follow-up time was 52 months. The screened group underwent 75% (95% confidence interval 51% to 91%) fewer emergency operations than the control group. Deaths due to abdominal aortic aneurysms occurred in nine patients in the screened group and 27 in the control group. The number needed to screen to save one life was 352. Specific mortality was significantly reduced by 67% (29% to 84%). Mortality due to non-abdominal aortic aneurysms was non-significantly reduced by 8%. The benefits of screening may increase with time.Conclusion Mass screening for abdominal aortic aneurysms in Danish men aged 65 or more reduces mortality.  相似文献

BackgroundGlucagon-like peptide-2 (GLP-2) is known to increase mesenteric blood flow. The aim of the study was to evaluate the effect of GLP-2 on blood flow in different vascular sites, and dynamic changes in cardiac parameters.Methods10 healthy volunteers were given 450 nmol subcutaneous (SC) GLP-2 or isotonic saline (5 subjects) in a single blinded manner. During the following 90 min, blood flow in the superior mesenteric artery (SMA), celiac artery (CA), renal artery (RA), common carotid artery (CCA) was measured using Doppler ultrasound (US), and cardiovascular variables were measured by impedance cardiography and finger plethysmography. Plasma GLP-2 was measured at times 0, 30 and 60 min.ResultsCompared to the placebo group, GLP-2 elicited a 27% decrease in the resistance index (RI) and a 269.4% increase in Time Averaged Maximal Velocity (TAMV) in the SMA (P < 0.01). CA, RA and CCA: There were no significant changes in RI or TAMV in the GLP-2 or placebo group, and no change in CA diameter.Cardiac parameters: GLP-2 increased cardiac output (CO), stroke volume (SV) and heart rate (HR) compared to baseline (respectively: 15.3, 4.81 and 8.2% (P < 0.001, P < 0.01 and P < 0.01)). The CO, SV and HR changes were not significantly different from the placebo group.Mean plasma GLP-2 serum levels in the placebo group at times 0, 30 and 60 min were 22.8, 23.4 and 23.2 pmol/l. In the GLP-2 group 20.3, 1273 and 1725 pmol/l.ConclusionSC GLP-2 increased SMA blood flow, as previously shown, but elicited no changes in other vascular sites. CO and HR increased significantly, presumably due to the increased mesenteric blood flow.  相似文献
