Plant antimicrobial peptides (AMPs) are a component of barrier defense system of plants. They have been isolated from roots, seeds, flowers, stems, and leaves of a wide variety of species and have activities towards phytopathogens, as well as against bacteria pathogenic to humans. Thus, plant AMPs are considered as promising antibiotic compounds with important biotechnological applications. Plant AMPs are grouped into several families and share general features such as positive charge, the presence of disulfide bonds (which stabilize the structure), and the mechanism of action targeting outer membrane structures.  相似文献

Dissimilatory ammonification was indicated as the common feature of ten rhizobial strains representing six species and three genera. In the absence of external electron acceptors, all investigated strains were capable of ethanolic fermentation. However, induction of anaerobic nitrite reduction was shown to be coupled with a shift of fermentation towards acetate in all the strains tested. Three metabolic groups could be distinguished with regard to nitrite regulation of ethanolic fermentation. It was shown for Bradyrhizobium sp. strain USDA 3045 that nitrite is the signal for switching between fermentative pathways although both ammonia and acetate excretion could not accelerate until nitrate had been utilized first. In the absence of N oxyanions, ethanol was indicated as the main product of mannitol fermentation, five-fold more abundant than acetate. An inverse composition was found in nitrite-amended cultures, due to a four-fold increase in acetate excretion whereas ethanol was kept at low level. Nitrite-supported fermentation towards acetate has not been previously reported for rhizobia. This benefit of this pathway was a two-fold shorter doubling time on 1% mannitol and 2.5 mM nitrite compared to no-nitrite media variants but also enabled fermentation of the more reduced carbon compound glycerol.  相似文献

Aberrant expression of the progenitor marker Neuron-glia 2 (NG2/CSPG4) or melanoma proteoglycan on cancer cells and angiogenic vasculature is associated with an aggressive disease course in several malignancies including glioblastoma multiforme (GBM) and melanoma. Thus, we investigated the mechanism of NG2 mediated malignant progression and its potential as a therapeutic target in clinically relevant GBM and melanoma animal models. Xenografting NG2 overexpressing GBM cell lines resulted in increased growth rate, angiogenesis and vascular permeability compared to control, NG2 negative tumours. The effect of abrogating NG2 function was investigated after intracerebral delivery of lentivirally encoded shRNAs targeting NG2 in patient GBM xenografts as well as in established subcutaneous A375 melanoma tumours. NG2 knockdown reduced melanoma proliferation and increased apoptosis and necrosis. Targeting NG2 in two heterogeneous GBM xenografts significantly reduced tumour growth and oedema levels, angiogenesis and normalised vascular function. Vascular normalisation resulted in increased tumour invasion and decreased apoptosis and necrosis. We conclude that NG2 promotes tumour progression by multiple mechanisms and represents an amenable target for cancer molecular therapy.  相似文献

Angelman Syndrome (AS, MIM 105830), classified among neurogenetic disorders, occurs with estimated frequency of 1:10 000 to 1:40 000. The characteristics features apart from neurodevelopmental impairment and seizures include peculiar face traits, absent speech, outburst of laughter, ataxia, stereotyped jerky (puppet-like) movements. Biochemical and genetic studies have identified peptides that play crucial roles in plant growth and development, including defence mechanisms in response to wounding by pests, the control of cell division and expansion, and pollen self-incompatibility. The first two signalling peptides to be described in plants were tomato systemin and phytosulfokine (PSK). There is also biochemical evidence that natriuretic peptide-like molecules, immunologically-related to those found in animals, may exist in plants. Another example of signalling peptide is ENOD40, a product of a gene, which became active early in the root nodulation process following Rhizobium infection of legumes. Other predicted bioactive peptides or oligopeptides have been identified by means of genetic, rather then biochemical methods. The Arabidopsis CLAVATA3 protein is required for the correct organization of the shoot apical meristem and the pollen S determinant S-locus cysteine-rich protein (SCR) also called S-locus protein 11, SP11). The plant signalling peptides discovered so far are involved in various processes and play an important role in communication between cells or organs, respectively. This review will focus on these peptides and their role in intercellular signalling.  相似文献

We have investigated the relationship between the inflow of air masses and the ragweed pollen concentration in SW Poland (Wroc?aw) for a 10-year period of 2005–2014. The HYSPLIT trajectory model was used to verify whether episodes of high concentrations can be related to regions outside of the main known ragweed centres in Europe, like Pannonian Plain, northern Italy and Ukraine. Furthermore, we used two different meteorological data sets (the global GDAS data set and from the WRF mesoscale model; the meteorological parameters were: U and V wind components, temperature and relative humidity) into HYSPLIT to evaluate the influence of meteorological input on calculated trajectories for high concentration ragweed episodes. The results show that the episodes of high pollen concentration (above 20 pm?3) represent a great part of total recorded ragweed pollen in Wroc?aw, but occur rarely and not in all years. High pollen episodes are connected with air masses coming from south and south-west Europe, which confirms the existence of expected ragweed centres but showed that other centres near Wroc?aw are not present. The HYSPLIT simulations with two different meteorological inputs indicated that footprint studies on ragweed benefit from a higher resolution meteorological data sets.  相似文献

Excised, carbohydrate-starved root meristems of Vicia faba subsp. minor have been used to investigate the impact of the auxin indole-3-acetic acid (IAA) and the cytokinin benzyl-6-aminopurine (BAP) on (1) the expression of Principal Control Points (PCPs) during the G1- and G2-phases of the cell cycle, and (2) the dynamics of sucrose-mediated resumption of DNA replication and mitosis (G1-to-S and G2-to-M transitions). Compared with the excised root tips starved in mineral medium without hormones, stationary phase meristems induced during continuous treatment with BAP, IAA, or a mixture of BAP+IAA, increased the number of G2 cells, producing characteristic profiles of nuclear DNA content. In medium containing 2% sucrose, BAP accelerated PCP1-->S and PCP2-->M, whereas continuous treatment with IAA resulted in marked prolongation of both transitions. Since the PCPs regulate progression through the key events of interphase and mitosis by interacting with cyclin dependent kinases (CDKs), these results seem to correspond with current data indicating functional connections between phytohormones, nutritional signals, gene expression and the cell division cycles in plants.  相似文献

The serious and growing impact of the neurodegenerative disorder Alzheimer''s disease (AD) as an individual and societal burden raises a number of key questions: Can a blanket test for Alzheimer''s disease be devised forecasting long-term risk for acquiring this disorder? Can a unified therapy be devised to forestall the development of AD as well as improve the lot of present sufferers? Inflammatory and oxidative stresses are associated with enhanced risk for AD. Can an AD molecular signature be identified in signaling pathways for communication within and among cells during inflammatory and oxidative stress, suggesting possible biomarkers and therapeutic avenues? We postulated a unique molecular signature of dysfunctional activity profiles in AD-relevant signaling pathways in peripheral tissues, based on a gain of function in G-protein-coupled bradykinin B2 receptor (BKB2R) inflammatory stress signaling in skin fibroblasts from AD patients that results in tau protein Ser hyperphosphorylation. Such a signaling profile, routed through both phosphorylation and proteolytic cascades activated by inflammatory and oxidative stresses in highly penetrant familial monogenic forms of AD, could be informative for pathogenesis of the complex multigenic sporadic form of AD. Comparing stimulus-specific cascades of signal transduction revealed a striking diversity of molecular signaling profiles in AD human skin fibroblasts that express endogenous levels of mutant presenilins PS-1 or PS-2 or the Trisomy 21 proteome. AD fibroblasts bearing the PS-1 M146L mutation associated with highly aggressive AD displayed persistent BKB2R signaling plus decreased ERK activation by BK, correctible by gamma-secretase inhibitor Compound E. Lack of these effects in the homologous PS-2 mutant cells indicates specificity of presenilin gamma-secretase catalytic components in BK signaling biology directed toward MAPK activation. Oxidative stress revealed a JNK-dependent survival pathway in normal fibroblasts lost in PS-1 M146L fibroblasts. Complex molecular profiles of signaling dysfunction in the most putatively straightforward human cellular models of AD suggest that risk ascertainment and therapeutic interventions in AD as a whole will likely demand complex solutions.  相似文献
