Lepidopterists have long acknowledged that many uncommon butterfly species can be extremely abundant in suitable locations. If this is generally true, it contradicts the general macroecological pattern of the positive interspecific relationship between abundance and distribution, i.e. locally abundant species are often geographically more widespread than locally rare species. Indeed, a negative abundance–distribution relationship has been documented for butterflies in Finland. Here we show, using the Finnish butterflies as an example, that a positive abundance–distribution relationship results if the geographically restricted species are missed, as may be the case in studies based on random or restricted sampling protocols, or in studies that are conducted over small spatial scales. In our case, the abundance–distribution relationship becomes negative when approximately 70 per cent of the species are included. This observation suggests that the abundance–distribution relationship may in fact not be linear over the entire range of distributions. This intriguing possibility combined with some taxonomic biases in the literature may undermine the generalization that for a given taxonomic assemblage there is a positive interspecific relationship between local abundance and regional distribution. 相似文献

Microcystins (MCs) are a group of closely related cyclic heptapeptides produced by a variety of common cyanobacteria. These are potent and highly specific hepatotoxins, the toxicity of which is based upon their inhibition of type-1 (PP1) and type-2A (PP2A) protein phosphatases. Apart from protein phosphatases, it is not known whether these phosphatase-inhibiting peptides could bind any other cellular proteins. We wanted to determine whether any possible unknown MC-adducts could explain the apoptotic effects observed at high concentrations of MCs. The question of other possible cellular proteins binding to MCs is also relevant when these compounds are employed for affinity purification of protein phosphatases. In MC-treated cell lysates, antibodies to MC recognized three protein adducts of 35-37 and 55 kD. By immunochemical and proteomics approaches, these proteins were identified as the catalytic subunits of type-1 and type-2A protein phosphatases and the ATP-synthase beta-subunit. A group-specific bioluminescent Escherichia coli strain for studying the action of beta-lactam antibiotics is described. The strain contains a plasmid, pBlaLux1, in which the luciferase genes from Photorhabdus luminescens are inserted under the control of the beta-lactam-responsive element ampR/ampC from Citrobacter freundii. In the presence of beta-lactams, the bacterial cells are induced to express the luciferase enzyme and three additional enzymes generating the substrate for the luciferase reaction. This biosensor for beta-lactams does not need any substrate or cofactor additions, and the bioluminescence can be measured very sensitively in real time by using a luminometer. Basic parameters affecting the light production and induction in the gram-negative model organism E. coli SNO301/pBlaLux1 by various beta-lactams were studied. The dose-response curves were bell shaped, indicating toxic effects for the sensor strain at high concentrations of beta-lactams. Various beta-lactams had fairly different assay ranges: ampicillin, 0.05-1.0 microg/ml; piperacillin, 0.0025-25 microg/ml; imipenem, 0.0025-0.25 microg/ml; cephapirin, 0.025-2.5 microg/ml; cefoxitin, 0.0025-1.5 microg/ml; and oxacillin, 25-500 microg/ml. Also, the induction coefficients (signal over background noninduced control) varied considerably from 3 to 158 in a 2-hour assay. Different non-beta-lactam antibiotics did not cause induction. Because the assay can be automated using microplate technologies, the approach may be suitable for higher throughput analysis of beta-lactam action. 相似文献

Worm density of a deposit feeding oligochaete (Lumbriculus variegatus) did not affect egestion whereas both temperature and
sediment type had a significant influence. The worms egested less actively at the lowest temperature (6 °C). The egestion
rate, expressed as mg dry feces produced, was highest in the sandy sediment and lowest in the sediment derived almost exclusively
from decaying plant material. The amount of dry material and the volume of wet sediment passing through the worms varied between
the test sediments; the results are probably dependent on the chosen unit of measure. Reproduction was significantly decreased
in sandy sediment with low organic carbon content. Reproduction was also dependent on the worm size, larger worms reproducing
more frequently. Most worms lost weight during the tests, but the loss was lowest in the sediment with the highest organic
carbon content.

The Streptomyces strain Streptomyces anulatus JH801 showed submerged sporulation and production of a genotoxic activity. The genotoxin production of this strain was found to be correlated to morphology and is likely to be related to sporulation. The influence of the medium on genotoxin production was investigated. Genotoxin production and sporulation were not inhibited by glucose, but were negatively affected by low ammonia concentrations. No correlation was found between genotoxin production and other morphological parameters such as total hyphal length and total average tip number. Bioassay-directed fractionation of the chloroform extract of S. anulatus JH801 resulted in the isolation of dihydroabikoviromycin. This compound shows selective activity towards DNA repair-deficient Escherichia coli . The isolation and structure of this genotoxin are reported. 相似文献

The nucleus of interphase eukaryotic cell is a highly compartmentalized structure containing the three-dimensional network of chromatin and numerous proteinaceous subcompartments. DNA viruses induce profound changes in the intranuclear structures of their host cells. We are applying a combination of confocal imaging including photobleaching microscopy and computational methods to analyze the modifications of nuclear architecture and dynamics in parvovirus infected cells. Upon canine parvovirus infection, expansion of the viral replication compartment is accompanied by chromatin marginalization to the vicinity of the nuclear membrane. Dextran microinjection and fluorescence recovery after photobleaching (FRAP) studies revealed the homogeneity of this compartment. Markedly, in spite of increase in viral DNA content of the nucleus, a significant increase in the protein mobility was observed in infected compared to non-infected cells. Moreover, analyzis of the dynamics of photoactivable capsid protein demonstrated rapid intranuclear dynamics of viral capsids. Finally, quantitative FRAP and cellular modelling were used to determine the duration of viral genome replication. Altogether, our findings indicate that parvoviruses modify the nuclear structure and dynamics extensively. Intranuclear crowding of viral components leads to enlargement of the interchromosomal domain and to chromatin marginalization via depletion attraction. In conclusion, parvoviruses provide a useful model system for understanding the mechanisms of virus-induced intranuclear modifications. 相似文献

Hereditary factors are presumed to play a role in one third of colorectal cancer (CRC) cases. However, in the majority of familial CRC cases the genetic basis of predisposition remains unexplained. This is particularly true for families with few affected individuals. To identify susceptibility genes for this common phenotype, we examined familial cases derived from a consecutive series of 1514 Finnish CRC patients. Ninety-six familial CRC patients with no previous diagnosis of a hereditary CRC syndrome were included in the analysis. Eighty-six patients had one affected first-degree relative, and ten patients had two or more. Exome sequencing was utilized to search for genes harboring putative loss-of-function variants, because such alterations are likely candidates for disease-causing mutations. Eleven genes with rare truncating variants in two or three familial CRC cases were identified: UACA, SFXN4, TWSG1, PSPH, NUDT7, ZNF490, PRSS37, CCDC18, PRADC1, MRPL3, and AKR1C4. Loss of heterozygosity was examined in all respective cancer samples, and was detected in seven occasions involving four of the candidate genes. In all seven occasions the wild-type allele was lost (P = 0.0078) providing additional evidence that these eleven genes are likely to include true culprits. The study provides a set of candidate predisposition genes which may explain a subset of common familial CRC. Additional genetic validation in other populations is required to provide firm evidence for causality, as well as to characterize the natural history of the respective phenotypes. 相似文献
