Manchego cheese is a high-fat pressed ewe's-milk cheese made in Castilla-La Mancha (Spain) and produced by enzymatic coagulation. The minimum ripening time before marketing required by the Regulatory Board of the Manchego Cheese Appellation of Origin is 60 days.
This paper describes the physicochemical, proteolysis, sensory and texture characteristics of Manchego cheese, and the degree of homogeneity of cheeses made under the Manchego Appellation of Origin. The data gathered in this study indicate that sensory and instrumental analysis are useful tools for detecting changes in Manchego cheese during ripening. These changes were first detected by the instrumental analysis (2 months). The panelists detected differences after 4 months' ripening in all the factories. With physicochemical analysis, on the other hand, longer ripening times (6–8 months) are required before such changes become appreciated. 相似文献

Morphologically similar microbial communities that often form on the walls of geographically distinct limestone caves have not yet been comparatively studied. Here, we analysed phylotype distribution in yellow microbial community samples obtained from the walls of distinct caves located in Spain, Czech Republic and Slovenia. To infer the level of similarity in microbial community membership, we analysed inserts of 474 16S rRNA gene clones and compared those using statistical tools. The results show that the microbial communities under investigation are composed solely of Bacteria. The obtained phylotypes formed three distinct groups of operational taxonomic units (OTUs). About 60% of obtained sequences formed three core OTUs common to all three sampling sites. These were affiliated with actinobacterial Pseudonocardinae (30-50% of sequences in individual sampling site libraries), but also with gammaproteobacterial Chromatiales (6-25%) and Xanthomonadales (0.5-2.0%). Another 7% of sequences were common to two sampling sites and formed eight OTUs, while the remaining 35% were site specific and corresponded mostly to OTUs containing single sequences. The same pattern was observed when these data were compared with sequence data available from similar studies. This comparison showed that distinct limestone caves support microbial communities composed mostly of phylotypes common to all sampling sites. 相似文献

A pilot experiment designed to test the effect of cattle, small mammals, and elevation on the success of reforestation of an endemic dwarf pine species in northeastern Mexico was implemented. Pinus culminicola (Andresen et Beaman) grows only in four high peaks in the Sierra Madre Oriental and is under pressure from grazing, wildfires, and human activities such as mining, road development for timber extraction, and telecommunication and aerial navigation devices. We planted and monitored 2‐year‐old seedlings at three elevations within the natural distribution range of this species at Cerro El Potosí in Nuevo León, Mexico. At each elevation three treatments were established: (1) seedlings protected from cattle plus small mammals, (2) seedlings protected from cattle, and (3) seedlings with free access to cattle and small mammals. Seedling survival was approximately 50% in (1) after 4 years, but there were no surviving seedlings with free access to cattle. Elevation in general did not account for variation in survival. Seedling growth was poor during the 4 years, which implies that seedlings remain susceptible to grazing and trampling by cattle and small mammals. The implications for a large‐scale restoration program are discussed. 相似文献

The reproductive cycle of the blue jack mackerel, Trachurus picturatus, had not been described for the Canary Islands. Between March 2005 and March 2006 monthly samples of T. picturatus were collected randomly at the central fishery wharf from the commercial catches of purse‐seiners in Tenerife Island waters (Canary Islands). Some 2472 specimens were analysed, with total lengths from 10.4 to 31.9 cm. Although females outnumbered males in summer, males were more abundant in the sex ratio (1.36 : 1). Based on the monthly evolution of the gonado‐somatic index and the proportion of mature individuals, the spawning season occurred between January and April, peaking in February. Lengths at first maturity (LFM) were calculated from the maturity ogives by the Gompertz model for all specimens (22.79 cm), for males (21.20 cm) and for females (23.05 cm). In this area the minimum legal size for T. picturatus is actually smaller than the length at first maturity and should be revised to avoid depletion of the stock. 相似文献

Cationic liposomes have been proposed as biocompatible gene delivery vectors, able to overcome the barriers imposed by cell membranes. Besides lipids, other surfactant molecules have been successfully used in the composition of gene carriers. In the present work, we used a Gemini surfactant, represented by the general structure [C(14)H(29)(CH(3))(2)N(+)(CH(2))(2)N(+)(CH(3))(2)C(14)H(29)]2Br(-) and herein designated 14-2-14, to prepare cationic gene carriers, both as the sole component and in combination with neutral helper lipids, cholesterol and DOPE. The effectiveness of three Gemini-based formulations, namely neat 14-2-14, 14-2-14:Chol (1:1 molar ratio) and 14-2-14:Chol:DOPE (2:1:1 molar ratio), to mediate gene delivery was evaluated in DNA mixtures of +/- charge ratios ranging from 1/1 to 12/1. After ruling out cytotoxicity as responsible for the differences observed in the transfection competence, structural and physical properties of the vector were investigated, using several techniques. The size and surface charge density (zeta potential) of surfactant-based structures were determined by conventional techniques and the thermotropic behaviour of aqueous dispersions of surfactant/lipid/DNA formulations was monitored by fluorescence polarization of DPH and DPH-PA probes. The capacity of lipoplexes to interact with membrane-mimicking lipid bilayers was evaluated, using the PicoGreen assay and a FRET technique. Our data indicate inefficiency of the neat 14-2-14 formulation for gene delivery, which could result from the large dimensions of the particles and/or from its relative incompetence to release DNA upon interaction with anionic lipids. The addition of cholesterol or cholesterol and DOPE conferred to Gemini-based gene carrier transfection activity at specific ranges of +/- charge ratios. Fluorescence polarization data suggest that an order parameter within a specific range was apparently needed for complexes to display maximal transfection efficiency. The transfection-competent formulations showed to be efficiently destabilized by interaction with different anionic and zwitterionic bilayers, including those containing PS and cardiolipin. ? It has long been believed that plant species from the tropics have higher levels of traits associated with resistance to herbivores than do species from higher latitudes. A meta-analysis recently showed that the published literature does not support this theory. However, the idea has never been tested using data gathered with consistent methods from a wide range of latitudes. ? We quantified the relationship between latitude and a broad range of chemical and physical traits across 301 species from 75 sites world-wide. ? Six putative resistance traits, including tannins, the concentration of lipids (an indicator of oils, waxes and resins), and leaf toughness were greater in high-latitude species. Six traits, including cyanide production and the presence of spines, were unrelated to latitude. Only ash content (an indicator of inorganic substances such as calcium oxalates and phytoliths) and the properties of species with delayed greening were higher in the tropics. ? Our results do not support the hypothesis that tropical plants have higher levels of resistance traits than do plants from higher latitudes. If anything, plants have higher resistance toward the poles. The greater resistance traits of high-latitude species might be explained by the greater cost of losing a given amount of leaf tissue in low-productivity environments. 相似文献
