Two experiments were designed to test possible effects of photoperiod and temperature during microsporogensis to anthesis on early autumn frost-hardiness of Picea abies progenies. Pollen lots were produced in phytotron rooms and used in crosses in a seed orchard. No biologically important differences in progeny performance were evident either between high and low temperature or between long and short-day treatments, and no significant interaction between photoperiod and temperature was found. In a third experiment, however, an effect of the environment during female flowering was obtained. Crosses performed in early spring (March) inside a heated greenhouse (short day, high temperature) produced progenies which were less hardy than their full-sibs reproduced from crosses indoors (long day, high temperature) and outdoors (long day, low temperature) in May. The most hardy siblings originated from the late-spring outdoor crosses. These results indicate that some stages in reproduction during female flowering, such as female meiosis, pollen tube growth, syngamy, early embryogenesis and embryo competition, may be sensitive to temperature and/or photoperiodic signals which can be transmitted to the progeny. We suspect that the altered performance of the progenies could be due to an activation of a regulatory mechanism affecting the expression of genes controlling adaptive traits. Both the present and earlier results have implications for the genetic interpretation of provenance differences in Norway spruce.  相似文献

The role of gibberellins (GAs) in photoperiodic control of leaf elongation in Poa pratensis was studied by both application of exogenous GAs and analysis of endogenous GAs. Leaf elongation was strongly increased under long day (LD, 24 h) conditions at both 9 and 21°C, leaf length at 9°C LD being similar to that in plants grown in short days (SD, 8 h) at 21°C. However, even at 21°C leaf elongation was enhanced by LD. Exogenous GA1 could completely compensate for LD at both 9 and 21°C. Gibberellins A20, A19 and A44 could also partly replace LD, but they were significantly less active than GA1, GA53 was inactive when applied to plants grown at 9°C in SD and exhibited only marginal activity at 9°C LD and 21°C SD. The total level of GAs of the early 13-hydroxylation pathway (A53, A44, A19, A20 and A1) increased rapidly when plants were transferred from SD to LD at 9°C. After transfer from 9 to 21°C, there was an increase in GA levels at both LD and SD, followed by a decrease under LD conditions. In all cases, GA19 was the predominant GA, accounting for 60 to 80% of the analysed GAs. Levels of the bioactive GA1 were low and increased transiently by LD four days after transfer from SD to LD. At both temperatures, the ratio GA19 to GA20 and GA20 to GA1 at 9°C was enhanced by LD compared with SD. Taken together, these results support the hypothesis that photoperiodic regulation of leaf elongation in Poa pratensis is GA-mediated, and they indicate a photoperiodic control of oxidation of GA53 to GA44 and GA19 to GA20, and perhaps also of 3β-hydroxylation of GA20 to GA1.  相似文献

Successful winter survival of perennial plants, like white clover, is dependent on proper timing of both hardening and dehardening. The purpose of this study was to investigate the regulation of these processes in two cultivars (AberCrest and AberHerald) and two Norwegian ecotypes (Særheim collected at 58°46′N lat. and Bodø at 67°20′N lat.) of white clover (Trifolium repens L.). For hardening and dehardening, plants were exposed to controlled temperature conditions and frost hardiness of stolons was tested by programmed freezing at the rate of 3°C per hour. In addition, stolons were analysed for starch, soluble sugars and soluble amino acids. Cultivars AberCrest and AberHerald, selected for growth at low temperature and winter hardiness in the United Kingdom, were significantly less hardy than the Norwegian populations. After six weeks of hardening (2 weeks at 6°C and 4 weeks at 0.5°C), estimated LT50 values were ?13.8, ?13.0, ?17.8 and ?20.3°C for AberCrest, AberHerald, Saerheim and Bodø, respectively. The rate of dehardening increased with increasing temperature. At low temperature (6°C), the northern ecotype from Bodø was more resistant to dehardening than AberHerald. However, at 18°C the absolute rate of dehardening (°C day?1) was twice as high in Bodø as in AberHerald plants. Stolon elongation during dehardening was initiated at lower temperatures in AberHerald than in plants of the Bodø ecotype. The content of total soluble sugars, sucrose and the amino acids proline and arginine were significantly higher in hardy plants of Bodø than in those of AberHerald. Sucrose levels decreased during dehardening and correlations between sucrose content and LT50 during this process were statistically highly significant for both Bodø and AberHerald. The least hardy populations of white clover were characterized by thick stolons, long internodes and large leaves.  相似文献

The influence of exogenously applied sucrose on cold hardening of raspberry ( Rubus idaeus L.) in vitro was examined. Raspberry plants (cv. Preussen) were cultured on Murashige-Skoog (MS) media with different levels (1, 3, 5 and 7%) of sucrose and subjected to low-temperature acclimation (3/−3°C day/night temperature, 8-h photoperiod) for 14 days. Cold hardiness (LT50 in controlled freezing), shoot moisture content, osmolality and the amounts of sucrose, glucose and fructose were determined. Exogenously applied sucrose was taken up by plants, but the uptake corresponded to less than 10% of total sugar reserves in the culture. Cold hardiness was primarily affected by acclimation treatment, but sucrose increased cold hardiness of nonacclimated plants and significantly enhanced the effect of acclimation treatment, 5% sucrose in the culture medium being optimal for cold hardening. LT50 values ranged between −4.1 and −7.1°C for nonacclimated, and between −14.2 and −20.7°C for cold-acclimated shoots. Shoot moisture content was inversely related to medium sucrose level and declined only slightly during cold acclimation. After cold acclimation, plant osmolality predicted hardiness better than shoot moisture content. Plant osmolality and sugar content were increased by increasing the medium sucrose level and, to a greater extent, by cold acclimation. Sucrose, glucose and fructose accumulated during hardening. Sucrose was the predominant sugar, and the rate of sucrose accumulation during cold acclimation was independent of the medium sucrose level or the initial plant sucrose content. A close correlation between cold hardiness and total sugars, sucrose, glucose and fructose was established. These results suggest that sugars have more than a purely osmotic effect in protecting acclimated raspberry plants from cold.  相似文献

Effects of exogenous gibberellins GA53, GA44, GA19, GA20 and GA1 on photoperiodically controlled shoot elongation in seedlings of Salix pentandra L. were studied. Gibberellins GA20 and GA1 induced shoot elongation under short days (SD) and could substitute for a transfer to long day (LD), while gibberellins A53, A44 and A19 were inactive. In seedlings exposed to a prolonged SD-treatment (30 days) there was a significant positive interaction between a transfer to LD and a treatment with GA20 and GA1 on shoot elongation. In addition, GA19 enhanced the growth promotive effect of LD in these seedlings. The results are compatible with the suggestion that conversion of GA19 to GA20 is blocked under SD. This effect is supposed to be an early process leading to the cessation of shoot elongation under SD. Responsiveness of the seedlings to LD and to a GA-treatment gradually decreased with an increasing length of exposure to SD.  相似文献

In the temperate-zone woody species Salix pentandra elongation growth is regulated by the photoperiod. Long days sustain active growth, whereas short days induce cessation of apical growth, which is a prerequisite for winter hardening. It is shown that this is correlated to quantitative changes in levels of endogenous GA19 GA20, and GA1. Within two short days the amount of the active GA1 and its immediate precursor GA20, decreased markedly in young leaves us well as in stem tissue. Also, the amount of GA19, declined, but the decrease was delayed relative to that of GA1 and GA20. The ability of S. pentandra seedlings to respond to exogenous GA19, decreased with increasing numbers of short days. Observations that support the hypothesis that the level of GA1 in S. pentandra is regulated by the photoperiod in a quantitative mode with conversion of GA19, to GA20, being one target for control.
Different distribution of GAs in various plant parts was observed. The level of GA was higher in young leaves than in other plant parts, and the amount of GA19 was 5–10 times higher in stem tissue than in leaves and roots. The ratios of GA8 to GA1 and GA20, were higher in roots as compared with other parts, as rods contained very low levels of GA1 and GA20, but amounts of GA20 comparable with other parts.  相似文献

By application of a recently developed method allowing analysis of gibberellins (GAs) in mg amounts of tissue, the effect of photoperiod on levels of GAs in shoot tips of individual seedlings of the woody species Salix pentandra was studied. In elongating long day-grown seedlings, maximum levels of GA1 were found 5–20 mm below the apex, approximately twice the levels in other segments. After exposure of plants to 5 or 15 short days, the levels of GA1 were about 50% lower within this specific region of the stem, as compared with seedlings grown under long days. Short day-induced cessation of shoot elongation also correlated with overall declines in the levels of GA53, GA19, GA20 and GA8, Within each photoperiodic treatment the levels of these GAs were generally relatively similar throughout the upper 35 mm of stems. No differences in internode lengths or in lengths of pith or epidermal cells were found in plants grown under long days compared with those exposed to 5 short days. In both cases, cells in mitosis were observed in the subapical stem tissues of shoot tips. After 15 short days, stem elongation was completed, and dividing cells were generally not found in the subapical part of the stem. However, short day exposure did not prevent elongation of internodes and cells differentiated before the treatment was started. Thus, the localised decrease in level of GA1 in shoot tips under short days precedes the morphological and anatomical changes connected with the short day-induced cessation of elongation growth. This supports the hypothesised role for GA1 in photoperiodic control of shoot elongation in S. pentandra .  相似文献

Survival of temperate-zone tree species under the normal summer-winter cycle is dependent on proper timing of apical growth cessation and cold acclimatization. This timing is primarily based on the perception of daylength, and through evolution many tree species have developed photoperiodic ecotypes which are closely adapted to the local light conditions. The longest photoperiod inducing growth cessation, the critical photoperiod, is inherited as a quantitative character. The phytochrome pigment family is the probable receptor of daylength, but the exact role of phytochrome and the physiological basis for the different responses between photoperiodic ecotypes are not known. This report shows for the first time that over-expression of the oat phytochrome A gene ( PHYA ) in a tree significantly changes the critical daylength and effectively prevents cold acclimatization. While the critical daylength for elongation growth in the wild-type of hybrid aspen ( Populus tremula × tremuloides ) was approximately 15 h, transgenic lines with a strong expression of the oat PHYA gene did not stop growing even under a photoperiod of 6 h. Quantitative analysis of gibberellins (GA) as well as indole-3-acetic acid (IAA) revealed that levels of these were not down-regulated under short days in the transgenic plants expressing high levels of oat PHYA , as in the wild-type. These results indicate that photoperiodic responses in trees might be regulated by the amount of PHYA gene expressed in the plants, and that the amount of phytochrome A (phyA) affects the metabolism of GAs and IAA.  相似文献
