Alix [ALG-2 (apoptosis-linked gene 2)-interacting protein X], a component of the endosomal sorting machinery, contains a three-dimensional docking site for HIV-1 p6(Gag) or EIAV (equine infectious anaemia virus) p9(Gag), and binding of the viral protein to this docking site allows the virus to hijack the host endosomal sorting machinery for budding from the plasma membrane. In the present study, we identified a monoclonal antibody that specifically recognizes the docking site for p6(Gag)/p9(Gag) and we used this antibody to probe the accessibility of the docking site in Alix. Our results show that the docking site is not available in cytosolic or recombinant Alix under native conditions and becomes available upon addition of the detergent Nonidet P40 or SDS. In HEK (human embryonic kidney)-293 cell lysates, an active p6(Gag)/p9(Gag) docking site is specifically available in Alix from the membrane fraction. The findings of the present study demonstrate that formation or exposure of the p6(Gag)/p9(Gag) docking site in Alix is a regulated event and that Alix association with the membrane may play a positive role in this process.  相似文献

Low-power laser therapy in medicine is widespread but the mechanisms are not fully understood. It has been suggested that low-power laser irradiation (LPLI) could induce photochemical reaction and activate several intracellular signaling pathways. Reactive oxygen species (ROS) are considered to be the key secondary messengers produced by LPLI. Here, we studied the signaling pathway mediated by ROS upon the stimulation of LPLI. Src tyrosine kinases are well-known targets of ROS and can be activated by oxidative events. Using a Src reporter based on fluorescence resonance energy transfer (FRET) and confocal laser scanning microscope, we visualized the dynamic Src activation in Hela cells immediately after LPLI. Moreover, Src activation by LPLI was in a dose-dependent manner. The increase of Src phosphorylation at Tyr416 was detected by Western blotting. In the presence of vitamin C, catalase alone, or the combination of catalase and superoxide dismutase (SOD), the activation of Src by LPLI is significantly abolished. In contrast, G?6983 loading, a PKC inhibitor, did not affect this response. Treatment of Hela cells with exogenous H(2)O(2) also resulted in a concentration-dependent activation of Src. Structural genomics initiatives aim to elucidate representative 3D structures for the majority of protein families over the next decade, but many obstacles must be overcome. The correct design of constructs is extremely important since many proteins will be too large or contain unstructured regions and will not be amenable to crystallization. It is therefore essential to identify regions in protein sequences that are likely to be suitable for structural study. Scooby-Domain is a fast and simple method to identify globular domains in protein sequences. Domains are compact units of protein structure and their correct delineation will aid structural elucidation through a divide-and-conquer approach. Scooby-Domain predictions are based on the observed lengths and hydrophobicities of domains from proteins with known tertiary structure. The prediction method employs an A*-search to identify sequence regions that form a globular structure and those that are unstructured. On a test set of 173 proteins with consensus CATH and SCOP domain definitions, Scooby-Domain has a sensitivity of 50% and an accuracy of 29%, which is better than current state-of-the-art methods. The relationships between cover and AGB for the dominant and widely distributed alpine grasslands on the northern Tibetan Plateau is still not fully examined. The objectives of this study are to answer the following question: (1) How does aboveground biomass (AGB) of alpine grassland relate to plant cover at different spatial scales? (2) What are the major biotic and abiotic factors influencing on AGB–cover relationship? A community survey (species, cover, height, and abundance) was conducted within 1 m × 1 m plots in 70 sites along a precipitation gradient of 50–600 m. Ordinary linear regression was employed to examine AGB–cover relationships of both community and species levels at regional scale of entire grassland and landscape scale of alpine meadow, alpine steppe, and desert steppe. Hierarchical partitioning was employed to estimate independent contributions of biotic and abiotic factors to AGB and cover at both scales. Partial correlation analyses were used to discriminate the effects of biotic and abiotic factors on AGB–cover relationships at two spatial scales. AGB and community cover both exponentially increased along the precipitation gradient. At community level, AGB was positively and linearly correlated with cover for all grasslands except for alpine meadow. AGB was also linearly correlated with cover of species level at both regional and landscape scales. Contributions of biotic and abiotic factors to the relationship between AGB and cover significantly depended on spatial scales. Cover of cushions, forbs, legumes and sedges, species richness, MAP, and soil bulk density were important factors that influenced the AGB–cover relationship at either regional or landscape scale. This study indicated generally positive and linear relationships between AGB and cover are at both regional and landscape scales. Spatial scale may affect ranges of cover and modify the contribution of cover to AGB. AGB–cover relationships were influenced mainly by species composition of different functional groups. Therefore, in deriving AGB patterns at different spatial scales, community composition should be considered to obtain acceptable accuracy.  相似文献

In addition to a good perovskite light absorbing layer, the hole and electron transport layers play a crucial role in achieving high‐efficiency perovskite solar cells. Here, a simple, one‐step, solution‐based method is introduced for fabricating high quality indium‐doped titanium oxide electron transport layers. It is shown that indium‐doping improves both the conductivity of the transport layer and the band alignment at the ETL/perovskite interface compared to pure TiO2, boosting the fill‐factor and voltage of perovskite cells. Using the optimized transport layers, a high steady‐state efficiency of 17.9% for CH3NH3PbI3‐based cells and 19.3% for Cs0.05(MA0.17FA0.83)0.95Pb(I0.83Br0.17)3‐based cells is demonstrated, corresponding to absolute efficiency gains of 4.4% and 1.2% respectively compared to TiO2‐based control cells. In addition, a steady‐state efficiency of 16.6% for a semi‐transparent cell is reported and it is used to achieve a four‐terminal perovskite‐silicon tandem cell with a steady‐state efficiency of 24.5%.  相似文献

【目的】分离好氧氯苯降解菌,并通过研究降解特性为应用提供理论依据。【方法】利用富集培养技术分离菌株,通过形态、生理生化反应特征及16S rRNA基因序列分析鉴定菌株,测定培养液中氯苯、其它氯苯类化合物和氯离子的浓度以及菌体细胞的密度和菌体细胞粗提液中邻苯二酚双加氧酶的活性,研究菌株的降解特性。【结果】16S rRNA基因序列相似性比较表明,分离出的菌株与乙酸钙不动杆菌(Acinetobacter calcoaceticus)的相似性高达98.5%。以初始浓度为50mg/L的氯苯为唯一碳源和能源时,120h内菌株对氯苯的降解率高达98.2%,氯离子净释放量和氯苯降解量的摩尔比范围为1:1.85-1:1.39,菌体细胞粗提液中邻苯二酚1,2-双加氧酶的平均活性为0.538U/mg蛋白质。加入葡萄糖后,菌体细胞数量和氯离子浓度明显增加,但单位细胞的氯苯降解能力明显下降。在二氯苯和三氯苯共存时,菌株对氯苯的降解能力受到明显的抑制作用,但对二氯苯有一定的降解作用,降解能力大小顺序为:1,3-二氯苯1,2-二氯苯1,4-二氯苯。【结论】分离出的好氧氯苯降解菌属于Acinetobacter属菌株,该菌株对氯苯和二氯苯均具有降解作用,可能通过邻位裂环途径降解氯苯,氯苯对菌株的降解能力和邻苯二酚1,2-双加氧酶的活性具有明显的增强作用。  相似文献

Cytochrome b(6)f complex with stoichiometrically bound beta-carotene molecule was purified from spinach chloroplasts. The configuration of this beta-carotene was studied by reversed-phase HPLC and resonance Raman spectroscopy. Both the absorption spectrum of this beta-carotene in dissociated state and the Raman spectrum in native state can be unambiguously assigned to a 9-cis configuration. This finding is in contrast to the predominantly all-trans isomers commonly found in membranes and protein-pigment complexes of chloroplasts, suggesting that the 9-cis-beta-carotene is an authentic component and may have a unique structural and functional role in cytochrome b(6)f complex.  相似文献

Tumorhead (TH) is a novel maternal gene product from Xenopus laevis containing several basic domains and a weak coiled-coil. Overexpression of wild-type TH resulted in increased proliferation of neural plate cells, causing expansion of the neural field followed by neural tube and craniofacial abnormalities. Overexpressed TH protein repressed neural differentiation and neural crest markers, but did not inhibit the neural inducers, pan-neural markers or mesodermal markers. Loss of function by injection of anti-TH antibody inhibited cell proliferation. Our data are consistent with a model in which tumorhead functions in regulating differentiation of the neural tissues but not neural induction or determination through its effect on cell proliferation.  相似文献
