Members of the ZFY and ZNF6 gene families have been cloned from species representing different taxa and different modes of sex determination. Comparisons of these genes show the ZFY-like and ZNF6 sequences to be strongly conserved across marsupials, birds, and lepidosaurians. Sequence analyzed by neighbor-joining indicated that both gene families are monophyletic with a high bootstrap value. Pairing of sequences from males and females of nonmammalian species showed there to be no significant difference between male and female sequences from a single species, consistent with autosomal locations. The molecular distances between murine Zfy-1, Zfy-2, and other ZFY-like sequences suggested that Zfy genes have undergone a period of rapid evolutionary change not seen in human ZFY.   相似文献

The synthesis and X-ray crystal structures of u-fac-[Ni(dien)2](NO3)2, s-fac-[Ni(dien)2](tos)2, fac-fac-[(H2O)(dien)Ni(μ-Cl)2(dien)(H2O)]Cl2, s-fac-[Ni(dien)2][ZnCl4], mer-[Ni(dien)2][CdCl4] · H2O, fac-[Ni(dien)(H2O)3](tos)2 · (H2O), mer-[Cu(dien)(H2O)](tos)2, fac-[Zn(dien)(H2O)2](tos)2 (dien = bis(2-aminoethyl)amine = diethylenetriamine; tos = p-toluenesulfonate) are described. Abstract: Várzeas are species-rich forest communities of the Central Amazon floodplains, inhabited by highly adapted tree species that can withstand long flooding periods. The leaf shedding behaviour and morphological traits that may contribute to adaptation to low oxygen levels were studied at the Ilha de Marchantaria on the lower Solimoes-Amazonas river, Brazil, and in greenhouse experiments with cuttings of six tree species typical of the Amazon floodplain. Comparison of deciduousness in situ revealed that four of the species under investigation, Salix martiana , Tabernaemontana juruana , Laetia corymbulosa and Pouteria glomerata , are able to maintain their leaf system during the aquatic period. Adventitious roots were formed by S. martiana and T. juruana , but anatomical characteristics differed between the species. Whereas S. martiana developed lysigenous aerenchyma in its roots during aerobic and anaerobic growth, only small intercellular spaces of schizogenous origin were formed in the root cortex of T. juruana . Similar to the latter species, such spaces were constitutively formed in the deciduous species Crateva benthami and Vitex cymosa . Suberin deposits were observed in tangential and radial cell walls of the hypodermis of roots from T. juruana , L. corymbulosa and P. glomerata . Suberin deposits were less pronounced in roots of S. martiana and absent in V. cymosa and C. benthami . The data show that different, almost contrasting, survival mechanisms have evolved in roots of plants with similar life forms in the same habitat. The results further suggest that the morphological traits of the investigated trees are causally linked with the in situ leaf shedding behaviour.  相似文献

Optical tweezers (infrared laser-based optical traps) have emerged as a powerful tool in molecular and cell biology. However, their usefulness has been limited, particularly in vivo, by the potential for damage to specimens resulting from the trapping laser. Relatively little is known about the origin of this phenomenon. Here we employed a wavelength-tunable optical trap in which the microscope objective transmission was fully characterized throughout the near infrared, in conjunction with a sensitive, rotating bacterial cell assay. Single cells of Escherichia coli were tethered to a glass coverslip by means of a single flagellum: such cells rotate at rates proportional to their transmembrane proton potential (. J. Mol. Biol. 138:541-561). Monitoring the rotation rates of cells subjected to laser illumination permits a rapid and quantitative measure of their metabolic state. Employing this assay, we characterized photodamage throughout the near-infrared region favored for optical trapping (790-1064 nm). The action spectrum for photodamage exhibits minima at 830 and 970 nm, and maxima at 870 and 930 nm. Damage was reduced to background levels under anaerobic conditions, implicating oxygen in the photodamage pathway. The intensity dependence for photodamage was linear, supporting a single-photon process. These findings may help guide the selection of lasers and experimental protocols best suited for optical trapping work.  相似文献

Salinity intrusion in the coastal freshwater rivers due to climate change and construction of the dam in the upstream rivers are alarming in aquaculture. Hence, an experiment was conducted to know the effects of salinity on growth performance, hemato‐biochemical parameters and erythrocytes structure in a commercially cultivable catfish species, striped catfish (Pangasianodon hypophthalmus). Firstly, median lethal concentration (LC50) of salinity for striped catfish was determined and then the fish were exposed to three salinity conditions (4, 8 and 12‰) and a control (0‰). Fish were sacrificed at day 7, 14, 28 and 56 after the start of salinity exposure. The 96 hr LC50 value was found to be 14.87‰. Salinity levels from freshwater to 8‰ showed optimal conditions with high survival rate and good growth performances of fish in terms of weight gain and specific growth rate (SGR). Interestingly, the lowest food conversion ratio (FCR) was found in 4‰ group. The hemoglobin (Hb) level and number of red blood cells (RBCs) were found to be decreased significantly in 8 and 12‰ compared to 0 and 4‰ at the initial days of exposure, while number of white blood cells (WBCs) and glucose level showed opposite scenario. Frequencies of ENA (erythrocytic nuclear abnormalities) and ECA (erythrocytic cellular abnormalities) were significantly increased with increasing salinities in the initial days of exposure. Overall, findings of the present study revealed that striped catfish might be suitable fish species for culture in the brackish water containing salinity up to 10‰.  相似文献

The structure of human serum albumin loaded with a metal porphyrin and fatty acids in solution is characterized by orientation-selective double electron-electron resonance (DEER) spectroscopy. Human serum albumin, spin-labeled fatty acids, and Cu(II) protoporphyrin IX—a hemin analog—form a fully self-assembled system that allows obtaining distances and mutual orientations between the paramagnetic guest molecules. We report a simplified analysis for the orientation-selective DEER data which can be applied when the orientation selection of one spin in the spin pair dominates the orientation selection of the other spin. The dipolar spectra reveal a dominant distance of 3.85 nm and a dominant orientation of the spin-spin vectors between Cu(II) protoporphyrin IX and 16-doxyl stearic acid, the electron paramagnetic resonance reporter group of the latter being located near the entry points to the fatty acid binding sites. This observation is in contrast to crystallographic data that suggest an asymmetric distribution of the entry points in the protein and hence the occurrence of various distances. In conjunction with the findings of a recent DEER study, the obtained data are indicative of a symmetric distribution of the binding site entries on the protein's surface. The overall anisotropic shape of the protein is reflected by one spin-spin vector orientation dominating the DEER data.  相似文献
