Abstract Aspergillus oryzae IFO4202 produces at least two extracellular lipolytic enzymes L1 and L2 (cutinase, and mono- and diacylglycerol lipase, respectively). Southern hybridization of restriction enzyme-digested genomic DNA fragments with 23mer oligonucleotides synthesized according to the amino acid sequence of the L2 as probe suggested the presence of the L2 gene (tentatively designated as mdlB ) and an additional weakly hybridizing region. A fragment containing the genomic mdlB gene was cloned in Escherichia coli . Nucleotide sequencing of the fragment revealed an open reading frame, comprising 1021 nucleotides, which contains two introns (51 and 52 nucleotides). Putative polyadenylation signals were found 182 and 287 bp downstream of the stop codon. The deduced amino acid sequence of the mdlB gene corresponds to 306 amino acid residues including a leader sequence of 28 amino acids and is highly similar to that of the mdlA gene of Penicillium camembertii . Three residues presumed to form the catalytic triad (serine, aspartic acid and histidine) of lipases were also conserved. 相似文献

Indican (indoxyl beta-D-glucoside) was found to accumulate only in green leaves of the indigo plant, and not in any other tissues. Comparisons of the indican content of protoplasts and vacuoles showed that indican was stored only in the vacuole of the cell. Indican content appeared and increased with the appearance and growth of leaves. In mature plants, the younger leaves contained larger amounts of indican than the older ones. Cell extracts of young leaves of indigo plant catalyzed the synthesis of indican from UDP-glucose and indoxyl. Indican synthase was extracted and purified from young leaves. The enzyme was separated into two fractions by anion-exchange chromatography. The enzyme in the fraction which was eluted by 0.1 M NaCl had a molecular weight of 53,000 by SDS-PAGE. Optimum pH of the enzyme was at about 10.0, indicating that the enzyme is likely localized in a different intracellular compartment from that of indican storage. The enzyme showed normal Michaelis-Menten kinetics and a K(m) value of 0.13 mM for UDP-glucose. 相似文献

To elucidate compositional changes of the common bile and main pancreatic ducts with aging, the authors investigated age-related
changes of element contents in the common bile and pancreatic ducts by inductively coupled plasma-atomic emission spectrometry.
After ordinary dissection by medical students was finished, the common bile ducts and main pancreatic ducts (pancreatic ducts)
were resected and the element contents were determined. The Mg content increased significantly only in the pancreatic duct
with aging, but the other element contents did not change significantly in both the common bile and pancreatic ducts with
aging. Regarding the relationship among the elements, significant direct correlations were found among the contents of Ca,
P, S, and Mg in the common bile ducts, with some exceptions between P and either S or Mg contents. In the pancreatic ducts,
significant direct correlations were found between S and Mg contents and between P and Na contents. The relationships in the
elements between the common bile and pancreatic ducts were examined. It was found that there were significant direct correlations
in the Ca, Mg, and Fe contents between the common bile and pancreatic ducts; that is, as Ca, Mg, and Fe increased in the common

Chemistry text mining tools should be interoperable and adaptable regardless of system-level implementation, installation or even programming issues. We aim to abstract the functionality of these tools from the underlying implementation via reconfigurable workflows for automatically identifying chemical names. To achieve this, we refactored an established named entity recogniser (in the chemistry domain), OSCAR and studied the impact of each component on the net performance. We developed two reconfigurable workflows from OSCAR using an interoperable text mining framework, U-Compare. These workflows can be altered using the drag-&-drop mechanism of the graphical user interface of U-Compare. These workflows also provide a platform to study the relationship between text mining components such as tokenisation and named entity recognition (using maximum entropy Markov model (MEMM) and pattern recognition based classifiers). Results indicate that, for chemistry in particular, eliminating noise generated by tokenisation techniques lead to a slightly better performance than others, in terms of named entity recognition (NER) accuracy. Poor tokenisation translates into poorer input to the classifier components which in turn leads to an increase in Type I or Type II errors, thus, lowering the overall performance. On the Sciborg corpus, the workflow based system, which uses a new tokeniser whilst retaining the same MEMM component, increases the F-score from 82.35% to 84.44%. On the PubMed corpus, it recorded an F-score of 84.84% as against 84.23% by OSCAR. 相似文献

Photoinhibition of the electron transport activity from tyrosine Z (YZ) in PS II to NADP+in Tris-treated thylakoids was suppressed by electron donation with either diphenylcarbazide or ascorbate (AsA) during the photoinhibition treatment. This suggests that AsA prevents donor side-induced photoinhibition in vivo as an endogenous donor. AsA in the lumen is photooxidized to monodehydroascorbate (MDA) in Tris-treated thylakoids, as detected by electron spin resonance spectrometry, but not in oxygenic thylakoids. Redox analysis of pyridine nucleotide in the presence of either MDA reductase or dehydroascorbate (DHA) reductase showed that the MDA photoproduced in the lumen is disproportionated to AsA and DHA, and the DHA leaking into the stroma is reduced to AsA by DHA reductase. No leakage of MDA through the thylakoid membrane was observed. Thus, the DHA-reducing enzyme system is indispensable in maintaining AsA concentrations in chloroplasts. 相似文献
