Glutenin is a major determinant of baking performance and viscoelasticity, which are responsible for high-quality bread with a light porous crumb structure of a well-leavened loaf. We analyzed the diversity of glutenin genes from six wheat cultivars (Korean cvs. Keumgang and Jinpum, Chinese cvs. China-108 and Yeonnon-78, and Japanese cvs. Norin-61 and Kantou-107). Glutenins contain two types of isoforms such as high molecular weight glutenin subunit (HMW-GS) and low molecular weight glutenin subunit (LMW-GS). Glutenin fractions were extracted from wheat endosperm using Osborne solubility method. A total of 217 protein spots were separated on two-dimensional gel electrophoresis with isoelectric focusing (wide range of pH 3–10). The proteins spots were subjected to tryptic digestion and identified by matrix assisted laser desorption/ionization–time of flight mass spectrometry. HMW-GS (43 isoforms) and LMW-GS (seven isoforms) are directly responsible for producing high-quality bread and noodles. Likewise, all the seed storage proteins are digested to provide nutrients for the embryo during seed germination and seedling growth. We identified the diverse glutenin subunits in wheat cultivars and compared the gluten isoforms among different wheat cultivars according to quality. This work gives an insight on the quality improvement in wheat crop.  相似文献

Acer. ITS 1 sequences in twenty-eight species of Acer and a species of Dipteronia in the family Aceraceae ranged from 220 to 242 bp and ITS 2 sequences from 215 to 251 bp. The size of the 5.8S coding region was 164 bp for all species examined in the family. Phylogenetic analysis of ITS sequences placed a very robust clade of section Palmata at the base of the tree. Three species of section Parviflora sensu de Jong (1994), A. spicatum, A. distylum and A. nipponicum, did not form a monophyletic clade. Acer spicatum was separated from the robust clade of A. distylum and A. nipponicum. Molecular tree strongly supports the close relationship among section Platanoidea, Glabra series Arguta, and section Macrantha. The close relationship between sections Pentaphylla and Trifoliata was also strongly suggested in ITS tree. Sections Rubra and Hyptiocarpa appeared to be closely allied with each other. The average rate of nucleotide substitution was estimated as (8.0±1.9)×10−11 substitutions per site per year for ITS 1 and (9.0±1.6) ×10−11 for ITS 2. Li metal, which has a high theoretical specific capacity and low redox potential, is considered to the most promising anode material for next‐generation Li ion‐based batteries. However, it also exhibits a disadvantageous solid electrolyte interphase (SEI) layer problem that needs to be resolved. Herein, an advanced separator composed of reduced graphene oxide fiber attached to aramid paper (rGOF‐A) is introduced. When rGOF‐A is applied, F? anions, generated from the decomposition of the LiPF6 electrolyte during the SEI layer formation process form semi‐ionic C? F bonds along the surface of rGOF. As Li+ ions are plated, the “F‐doped” rGO surface induces the formation of LiF, which is known as a component of a chemically stable SEI, therefore it helps the Li metal anode to operate stably at a high current of 20 mA cm?2 with a high capacity of 20 mAh cm?2. The proposed rGOF‐A separator successfully achieves a stable SEI layer that could resolve the interfacial issues of the Li metal anode.  相似文献

Several mycobacterial strains, such as Mycobacterium flavescens, Mycobacterium gastri, Mycobacterium neoaurum, Mycobacterium parafortuitum, Mycobacterium peregrinum, Mycobacterium phlei, Mycobacterium smegmatis, Mycobacterium tuberculosis, and Mycobacterium vaccae, were found to grow on carbon monoxide (CO) as the sole source of carbon and energy. These bacteria, except for M. tuberculosis, also utilized methanol as the sole carbon and energy source. A CO dehydrogenase (CO-DH) assay, staining by activity of CO-DH, and Western blot analysis using an antibody raised against CO-DH of Mycobacterium sp. strain JC1 (formerly Acinetobacter sp. strain JC1 [J. W. Cho, H. S. Yim, and Y. M. Kim, Kor. J. Microbiol. 23:1-8, 1985]) revealed that CO-DH is present in extracts of the bacteria prepared from cells grown on CO. Ribulose bisphosphate carboxylase/oxygenase (RubisCO) activity was also detected in extracts prepared from all cells, except M. tuberculosis, grown on CO. The mycobacteria grown on methanol, except for M. gastri, which showed hexulose phosphate synthase activity, did not exhibit activities of classic methanol dehydrogenase, hydroxypyruvate reductase, or hexulose phosphate synthase but exhibited N,N-dimethyl-4-nitrosoaniline-dependent methanol dehydrogenase and RuBisCO activities. Cells grown on methanol were also found to have dihydroxyacetone synthase. Double immunodiffusion revealed that the antigenic sites of CO-DHs, RuBisCOs, and dihydroxyacetone synthases in all mycobacteria tested are identical with those of the Mycobacterium sp. strain JC1 enzymes.  相似文献

Peptide mass fingerprinting (PMF) has become one of the most widely used methods for rapid identification of proteins in proteomics research. Many peaks, however, remain unassigned after PMF analysis, partly because of post-translational modification and the limited scope of protein sequences. Almost all PMF tools employ only known or predicted protein sequences and do not include open reading frames (ORFs) in the genome, which eliminates the chance of finding novel functional peptides. Unlike most tools that search protein sequences from known coding sequences, the tool we developed uses a database for theoretical small ORFs (tsORFs) and a PMF application using a tsORFs database (tsORFdb). The tsORFdb is a database for ORFeome that encompasses all potential tsORFs derived from whole genome sequences as well as the predicted ones. The massProphet system tries to extend the search scope to include the ORFeome using the tsORFdb. The tsORFdb and massProphet should be useful for proteomics research to give information about unknown small ORFs as well as predicted and registered proteins.  相似文献

Cholesterol 24S-hydroxylase (CYP46A1) is of key importance for cholesterol homeostasis in the brain. This enzyme seems to be resistant toward most regulatory factors and at present no drug effects on its activity have been described. The crystal structures of the substrate-free and substrate-bound CYP46A1 were recently determined (Mast et al., Crystal structures of substrate-bound and substrate-free cytochrome P450 46A1, the principal cholesterol hydroxylase in the brain. Proc. Natl. Acad. Sci. USA. 2008. 105: 9546–9551). These structural studies suggested that ligands other than sterols can bind to CYP46A1. We show here that the antifungal drug voriconazole binds to the enzyme in vitro and inhibits CYP46A1-mediated cholesterol 24-hydroxylation with a Ki of 11 nM. Mice treated with daily intraperitoneal injections of voriconazole for 5 days had high levels of voriconazole in the brain and significantly reduced brain levels of 24S-hydroxycholesterol. The levels of squalene, lathosterol, and HMG-CoA reductase mRNA were reduced in the brain of the voriconazole-treated animals as well, indicating a reduced cholesterol synthesis. Most of this effect may be due to a reduced utilization of cholesterol by CYP46A1. One of the side-effects of voriconazole is visual disturbances. Because CYP46A1 is also expressed in the neural retina, we discuss the possibility that the inhibition of CYP46A1 by voriconazole contributes to these visual disturbances.  相似文献

We examined 15 species of ornamental tropical fishes originating from Southeast Asia to determine the cause of losses among 8 fish farms in Korea. A total of 351 individuals belonging to 5 different families (1 species of Characidae, 6 of Cichlidae, 3 of Cyprinidae, 1 of Heleostomatidae, and 4 of Poecilidae) were collected for the purpose of detecting metazoan and protozoan parasites. Parasites were fixed and stained using routine methods, and identified. We found 3 ciliates, 2 monogeneans, 1 nematode, and 1 copepod from 7 host species. Of these, Ichthyophthirius multifiliis was the most common parasite in our study, and together with Trichodina sp., caused mass mortality of Sumatra barb Puntius tetrazona at 1 farm. We also found Camallanus cotti and Tetrahymena corlissi from guppies Poecilia reticulata, both for the first time in Korea. Farmers consider these 2 pathogens to be the most serious ones in Korea. Gussevia asota from oscar Astronotus ocellatus, and Gyrodactylus bullatarudis from platy Xiphophorus maculatus were also found in Korea for the first time. We believe that appropriate quarantine practices for tropical ornamental fishes should be introduced because the failure to require and implement quarantines has already resulted in the accidental introduction of exotic parasites to fish farms, and because these parasites can cause further economic losses if they become established in the wild.  相似文献
