Transgenic techniques in chickens have been developed much more slowly than in mammals due to chickens’ unique reproduction mechanism. Retroviral methods have been the most successful. piggyBac (PB) is a transposon that has a 13 bp perfect terminal invert repeat sequence. PB can be inserted into TTAA sites and can also be precisely excised in mammals. Therefore, we have selected PB as a candidate to establish a new method to produce transgenic chickens. We constructed three donor vectors (ZGl-neo, ZGm-neo and ZGs-neo) expressing a GFP marker-gene and a neomycin resistant gene based on PB. We co-transfected each donor vector with a helper vector (CAG-PBase). We found that ZGl-neo was the most efficient PB vector. This vector could insert into TTAA sites in DF-1 cells. PB vectors were microinjected into sub-germinal cavity of newly laid eggs, and electroporation was then performed with a 20-V pulse for 5 cycles of 50 ms on and 100 ms off. GFP was expressed in different tissues of the embryos, including the gonads. Twenty-two chickens hatched after microinjection with compounds ZGl-neo and CAG-PBase (3:1). When we screened the blood DNA, 73 % (16/22) of the individuals were positive. Thirteen of the chickens grew to adulthood, 11 of which were males. 40 % (4/10) of the individuals were semen positive, and their copy numbers ranged from 0.05 to 0.21 (0.11, 0.21, 0.05, 0.06). No G1 offspring containing the integrated transposon were produced. We conclude that the PB transposon system is a novel useful tool for the efficient production of transgenic chickens.  相似文献

基因组选择是指在全基因组范围内通过基因组中大量的标记信息估计出个体全基因组范围的育种值,可进一步提升育种效率和准确性,目前在猪纯繁育种中得到广泛应用。但有研究表明,现有的基因组选择方法在猪杂交育种上的应用效果并不理想,在跨群体条件下预测准确性极低。杂交作为养猪业中最为广泛的育种手段之一,通过结合基因组选择理论进一步提升猪的生产性能,具有重要的经济和研究价值。本文综述了基因组选择的发展及其在猪育种中的应用现状,并结合国内外猪杂交育种的方式,分析了目前基因组选择方法在猪杂交育种应用方面的不足,旨在为未来基因组选择在猪杂交育种中的合理应用提供参考。  相似文献

Liver cancer, one of the most common malignancies, is the second leading cause of cancer death in the world. The citrus reticulate peel and black tea have been studied for their beneficial health effects. In spite of the many studies have been reported, the underlying molecular mechanisms underlying its health benefits are still not fully understood. In present study, we developed a unique citrus reticulate peel black tea (CRPBT) by combined citrus reticulate peel and black tea and assessed its active ingredients, anti-oxidant and anti-liver cancer effects in vitro. The results suggested that CRPBT exhibited antioxidant capacity and effectively inhibited proliferation and migration of liver cancer cells in a dose- and time- dependent manner. Mechanistically, CRPBT significantly down-regulated phosphorylation of PI3K and AKT, and up-regulated the ratio of Bax/Bcl-2, and suppressed the expression of MMP2/9, N-cadherin and Vimetin proteins in liver cancer cells. Taken together, CRPBT has good effect on inhibiting migration, invasion, proliferation, and inducing apoptosis in liver cancer cells.

目的:探讨徒手淋巴引流对全膝关节置换术(TKA)后肢体肿胀程度及血液炎性指标的影响。方法:选取2018年12月~2019年12月广东省中医院珠海医院收治的拟行初次TKA手术的膝骨关节炎(KOA)患者70例,随机分为观察组和对照组各35例,对照组按TKA术后常规方案治疗,观察组在对照组基础上增加徒手淋巴引流术,比较两组术前,术后1 d、4 d、7 d、2周、4周、8周肤温、肿胀程度及血液炎性指标,评估患者术后康复情况及不良事件发生情况。结果:两组术后2周肤温低于术后7 d,术后4周低于术后2周,术后8周低于术后4周,差异均有统计学意义(P0.05)。观察组术后2周、4周、8周肤温均低于对照组,差异均有统计学意义(P0.05)。两组术后1 d肢体肿胀度低于术后4 d,术后2周低于术后7 d,术后4周低于术后2周,术后8周低于术后4周,差异均有统计学意义(P0.05)。观察组术后7d、2周、4周、8周肢体肿胀度均低于对照组,差异均有统计学意义(P0.05)。两组术后4 d白细胞计数(WBC)水平低于术后1 d,而超敏C反应蛋白(hs-CRP)水平高于术后1 d;两组术后7 d WBC、hs-CRP及血沉(ESR)水平均低于术后4 d,且术后2周低于术后7 d,术后4周低于术后2周,术后8周低于术后4周,差异均有统计学意义(P0.05)。观察组术后7 d hs-CRP水平均低于对照组,术后2周、4周、8周hs-CRP、ESR水平均低于对照组,差异均有统计学意义(P0.05)。结论:TKA术后应用徒手淋巴引流手法能有效减轻患者术后肢体肿胀程度,缩短炎症周期,降低炎症反应,在临床上值得推广应用。  相似文献

To enhance the production of isoprene, a volatile 5-carbon hydrocarbon, in the Gram-positive spore-forming rod-shaped bacterium Bacillus subtilis, 1-deoxy-d-xylulose-5-phosphate synthase (Dxs) and 1-deoxy-d-xylulose-5-phosphate reductoisomerase (Dxr) were overexpressed in B. subtilis DSM 10. For the strain that overexpresses Dxs, the yield of isoprene was increased 40% over that by the wild-type strain. In the Dxr overexpression strain, the level of isoprene production was unchanged. Overexpression of Dxr together with Dxs showed an isoprene production level similar to that of the Dxs overproduction strain. The effects of external factors, such as stress factors including heat (48°C), salt (0.3 M NaCl), ethanol (1%), and oxidative (0.005% H(2)O(2)) stress, on isoprene production were further examined. Heat, salt, and H(2)O(2) induced isoprene production; ethanol inhibited isoprene production. In addition, induction and repression effects are independent of SigB, which is the general stress-responsive alternative sigma factor of Gram-positive bacteria.  相似文献
