The complete life cycle of Amblyospora campbelli (Kellen and Wills, 1962) (Microsporida: Amblyosporidae) requires a two-host system involving the mosquito host, Culiseta incidens (Thomson), and an obligatory intermediate copepod host. The parasite has dimorphic spore development producing meiospores (haploid condition) and binucleated spores (diploid condition), either as an exclusive infection or simultaneously (within females only). This is the 1st known report of concurrent spore development within an adult mosquito host, and, therefore, shows the Amblyospora campbelli system to be uniquely different from other Amblyospora spp. cycles previously described. The significance of dimorphic spore development is discussed. In females, diplokaryotic meronts may invade oenocytes, causing a benign-type of infection. A blood-meal is required to initiate sporulation of the binucleate spore. The binucleate spore contains the sporoplasm involved in transovarial transmission. A 2nd sporulation sequence, primarily in adipose tissue, may involve both males and females. In this sequence, repeated merogonic division greatly increased the density of diplokaryotic meronts and generally involved most of the body of the host. Production of meiospores, unlike that for the binucleate spore, appeared to be spontaneous (i.e. no obligatory blood meal). Survivorship of male and female larval mosquitoes was nearly equal. Adult females spread the parasite in three ways: transovarial, transovum, and by meiospore deposition.  相似文献

The dual subcellular fate of fibroblast growth factor 3 (FGF3) is determined by the competing effects of amino-terminal signals for nuclear localization and secretion (P. Kiefer, P. Acland, D. Pappin, G. Peters, and C. Dickson, EMBO J. 13:4126-4136, 1994). Mutation analysis has implicated additional basic domains in the carboxy-terminal region of the protein as necessary for nuclear uptake and the association of FGF3 with the nucleoli. Immunogold electron microscopy shows that FGF3 is predominantly within the dense fibrillar component of the nucleolus. A form of FGF3 that localizes exclusively in the nucleus and nucleolus was generated by removing signals for secretion, and expression of this nonsecreted FGF3 in a mammary epithelial cell line resulted in slowly growing colonies of enlarged cells. Thus, nuclear import and nucleolar association of FGF3 are determined by the concerted interaction of several distinct motifs, and the exclusive production of the nuclear isoform can inhibit DNA synthesis and cell proliferation.  相似文献

The effects of hyper- and hypo-saline stresses on the levels of various inorganic and organic solutes inUlva lactuca have been recorded. Hypoosmotic stress decreased the tissue concentration of K+, Na+ and Cl- while hyper-osmotic stress caused a transient increase in Na+ and a stable accumulation of K+ and Cl-. The tissue content of -dimethylsulphoniopropionate (-dimethylpropiothetin) responded to changes in salinity. The time course of hypersaline stress showed the -dimethylsulophoniopropionate concentration rose as the Na+ level fell. The levels of free sugars and amino acids, including proline, were relatively low in this alga and did not appear to be important in osmotic adjustment. Summary Post-meiotic segregation (PMS) results in the formation of mixed genotypes from single meiotic products. A method is described in which single members of tetrads are selected, and these are then tested for their genetic homogeneity. The method is applied to Ustilago maydis using crosses which are heteroallelic for nar 1, the structural gene for nitrate reductase. In the absence of PMS, meiotic products containing a nar + recombinant are genetically pure (the equivalent of a 6 mutant: 2 wild-type octad). With PMS, a nar + recombinant clone arises in association with a nar - clone and these are otherwise genetically identical (the equivalent of a 7 mutant: 1 wild type octad). The procedure will make it possible to search for mutant strains which are defective in the correction of mismatched bases in hybrid DNA formed during recombination. Among 26 nar + recombinants from a control cross, PMS was detected on 3 occasions. In an equivalent cross, both parents were uvs 3, a mutant defective in the excision of pyrimidine dimers from DNA. Among 43 nar + recombinants, 7 arose from PMS. Thus the frequency of PMS for the nar alleles is about 15% and the excision of pyrimidine dimers is probably unrelated to the repair of mismatched bases in hybrid DNA.  相似文献

The molecular weights of a number of 125I-labeled plasma proteins have been determined from an analysis of their sedimentation equilibrium behavior in an air-driven ultracentrifuge. The values obtained agree well with results obtained by other methods. Molecular weights obtained for 125I-labeled bovine serum albumin and the rat serum proteins albumin, α1-acid glycoprotein, and major acute-phase α1-protein were unaffected by the addition of 7% rat plasma. Direct evidence for protein-protein interactions was obtained for mixtures of 125I-labeled rat α1-acid glycoprotein and the plant lectin concanavalin A and for mixtures of 125I-labeled protein A from Staphylococcus aureus and 7% rat plasma. Interactions of a different type were observed when the sedimentation equilibrium profiles of 125I-labeled proteins were determined in concentrated solutions of other proteins. Under these conditions the effects of molecular exclusion or nonideality became significant and low estimates were obtained for the molecular weights of the labeled proteins. Analysis of the data obtained for 125I-labeled bovine serum albumin in concentrated solutions of bovine serum albumin (20–80 mg/ ml) yielded nonideality coefficients in good agreement with literature values. Analysis of the behavior of 125I-labeled rat serum albumin, transferrin, and α1-acid glycoprotein yielded nonideality coefficients and hence activities of these proteins in undiluted rat plasma.  相似文献

Two populations of Trophonema okamotoi parasitized by Pasteuria sp. were found on Liquidambar styraciflua (sweetgum) and on an unidentified tropical grass in north-central Florida. Endospores of this Pasteuria sp. attached to motile vermiform second-stage juveniles (J2) and males of T. okamotoi, but not to other developmental stages. Sporangia and new endospores were produced only inside the bodies of swollen and sedentary third- and fourth-stage juveniles and females that developed in the host roots. No egg masses were produced by infected T. okamotoi females. The endospore diameter from the tropical grass population was 4.93 μm and the central core diameter was 1.97 μm; measurements of endospores from the sweetgum populations were similar. Endospores that were collected from T. okamotoi and added to uninfected T. okamotoi and other plant-parasitic nematodes attached/to J2 of T. okamotoi but did not attach to juveniles and adults of Helicotylenchus pseudorotrustus, Pratylenchus brachyurus, or to J2 of either Meloidogyne arenaria race 1, M. incognita race 1, M. javanica, or Tylenchulus semipenetrans. Pasteuria sp. from T. okamotoi differed from the described Pasteuria species in endospore size, host preference, and rate of attachment.  相似文献

Perturbation of the T cell antigen-specific receptor leads to a series of signaling events that includes a rapid increase in phosphoinositide hydrolysis, intracellular Ca2+, and tyrosine phosphorylation. We have examined the function of tyrosine phosphorylation in isolation by introducing the v-src tyrosine kinase into a T cell hybridoma. T cell receptor-mediated increases in phosphoinositide hydrolysis and, in particular the generation of inositol 1,4,5-trisphosphate, were comparable between v-src+ and v-src- cells. Unexpectedly, the v-src+ cells exhibited spontaneously elevated intracellular Ca2+ and exaggerated Ca2+ increases when stimulated via the T cell receptor. The enhanced Ca2+ response was not due to tyrosine phosphorylation of the T cell receptor itself, since the phenotype was evident in T cell receptor zeta chain-/v-src+ cells as well. These results demonstrate that an active protein tyrosine kinase can markedly affect intracellular Ca2+ handling by a process independent of inositol 1,4,5-trisphosphate production and T cell receptor tyrosine phosphorylation and raise the possibility that tyrosine kinases may directly regulate T cell receptor-mediated changes in intracellular Ca2+.  相似文献

It is difficult to retain and localize radioactive, water-soluble compounds within plant cells. Existing techniques retain water-soluble compounds with varying rates of efficiency and are limited to processing only a few samples at one time. We developed a modified pressure infiltration technique for the preparation of microautoradiographs of 14C-labeled, water-soluble compounds in plant tissue. Samples from cottonwood (Populus deltoides Bartr. ex Marsh.) labeled with 14C were excised, quick frozen in liquid N2, freeze-dried at −50°C, and pressure-infiltrated with epoxy resin without intermediate solvents or prolonged incubation times. The technique facilitates the mass processing of samples for microautoradiography, gives good cellular retention of labeled water-soluble compounds, and is highly reproducible.  相似文献
