Many plasmid-harbouring strains of Lactococcus lactis have been isolated from milk and other sources. Plasmids of Lactococcus have been shown to harbour antibiotic resistance genes and those that express some important proteins. The generally regarded
as safe (GRAS) status of L. lactis also makes it an attractive host for the production of proteins that are beneficial in numerous applications such as the

The Cenomanian–Turonian boundary (CTB) in the ?i?arija Mountain region (northern Istria, Croatia) is characterized by calcisphere limestone successions with a firmground and glauconite horizon, bioturbated intervals, tempestites, and slumped structures as well as microbially laminated and organic-rich interbeds deposited in the northwestern part of the intra-Tethyan Adriatic Carbonate Platform (AdCP). Compilation of the results from three studied sections (Vodice–Jelovica, Martinjak and Planik) of litho-, bio-, and microfacies analyses, X-ray diffraction, SEM, EDS, and stable isotope analyses allowed reconstruction of marine paleoenvironmental conditions during this time period. Shallow-marine carbonate deposits of the Milna Formation underlie a drowned-platform succession of the Sveti (Sv.) Duh Formation. The contact between these two formations is sharp and commonly marked by slumped deposits. The Sv. Duh Formation consists of about 100 m of calcisphere wackestone enriched in organic matter. The results of preliminary δ13C and δ18O stable isotope analyses indicate the influence of the global Oceanic Anoxic Event (OAE2) on the deposition of this carbonate succession. Anoxic and hypoxic conditions in the water column lead to major changes in the shallow-marine carbonate system of the AdCP. Numerous benthic foraminifera declined during that time, but planktonic foraminifera and calcareous dinoflagellates diversified and expanded greatly. The results of this research provide new insights into the character of the CTB interval in this part of the Tethyan realm. Local and regional synsedimentary tectonics combined with global upper Cretaceous sea-level dynamics allows the correlation of the investigated deeper-marine lithostratigraphic units with OAE2. 相似文献
Rat sinusoidal liver cells possess the surface receptor for high density lipoprotein (HDL) (Murakami, M., S. Horiuchi, K. Takata, and Y. Morino. 1987. J. Biochem. (Tokyo) 101: 729-741). The present study was undertaken to determine whether cell surface-bound HDL underwent subsequent endocytic internalization by using 125I-labeled HDL and fluorescein isothiocyanate-labeled HDL (FITC-HDL). The cell-associated radioactivity obtained by a 40-min incubation with 125I-labeled HDL at 37 degrees C was released into the medium as acid-precipitable forms upon further incubation at 37 degrees C. When further incubated at 0 degree C instead of 37 degrees C, however, this release was significantly reduced. A similar phenomenon was observed after the cell-associated ligands had been treated with trypsin. The cell-associated ligands obtained after a 1-hr incubation with 125I-labeled HDL at 0 degree C were largely counted for by those bound to the outer surface of the cells, thus suggesting that HDL is internalized into cells at 37 degrees C but not at 0 degree C. Moreover, when cells were incubated with FITC-HDL at 0 degree C, the cell-associated ligands were found in a pH 7.2 +/- 0.1 compartment, whereas when incubated at 37 degrees C, its microenvironmental pH became much more acidic, exhibiting pH 6.2 +/- 0.1. Furthermore, this value returned to 7.1 +/- 0.1 upon treatment with carbonylcyanide m-chlorophenylhydrazone known to dissipate the total protonomotive force. These results suggest, therefore, that the internalization process does follow receptor-mediated binding of HDL in rat sinusoidal liver cells. Developing fragrant rice through marker‐assisted/aided selection (MAS) is an economical and profitable approach worldwide for the enrichment of an elite genetic background with a pleasant aroma. The PCR‐based DNA markers that distinguish the alleles of major fragrance genes in rice have been synthesised to develop rice scent biofortification through MAS. Thus, the present study examined the aroma biofortification potential of these co‐dominant markers in a germplasm panel of 189 F2 progeny developed from crosses between a non‐aromatic variety (MR84) and a highly aromatic but low‐yielding variety (MRQ74) to determine the most influential diagnostic markers for fragrance biofortification. The SSRs and functional DNA markers RM5633 (on chromosome 4), RM515, RM223, L06, NKSbad2, FMbadh2‐E7, BADEX7‐5, Aro7 and SCU015RM (on chromosome 8) were highly associated with the 2AP (2‐acetyl‐1‐pyrroline) content across the population. The alleles traced via these markers were also in high linkage disequilibrium (R2 > 0.70) and explained approximately 12.1, 27.05, 27.05, 27.05, 25.42, 25.42, 20.53, 20.43 and 20.18% of the total phenotypic variation observed for these biomarkers, respectively. F2 plants harbouring the favourable alleles of these effective markers produced higher levels of fragrance. Hence, these rice plants can be used as donor parents to increase the development of fragrance‐biofortified tropical rice varieties adapted to growing conditions and consumer preferences, thus contributing to the global rice market. 相似文献
The frequency of damaged Varroadestructor Anderson and Trueman (Mesostigmata: Varroidae) found on the bottom board of hives of the honey bee, Apis mellifera L. (Hymenoptera: Apidae) has been used as an indicator of the degree of tolerance or resistance of honey bee colonies against
mites. However, it is not clear that this measure is adequate. These injuries should be separated from regular dorsal dimples
that have a developmental origin. To investigate damage to Varroa mites and regular dorsal dimples, 32 honey bee (A. mellifera) colonies were selected from four Iranian provinces: Isfahan, Markazi, Qazvin, and Tehran. These colonies were part of the
National Honey bee Breeding Program that resulted in province-specific races. In April, Varroa mites were collected from heavily infested colonies and used to infest the 32 experimental colonies. In August, 20 of these
colonies were selected (five colonies from each province). Adult bees from these colonies were placed in cages and after introducing
mites, damaged mites were collected from each cage every day. The average percentage of injured mites ranged from 0.6 to 3.0%
in four provinces. The results did not show any statistical differences between the colonies within provinces for injuries
to mites, but there were some differences among province-specific lines. Two kinds of injuries to the mites were observed:
injuries to legs and pedipalps, and injuries to other parts of the body. There were also some regular dorsal dimples on dorsal
idiosoma of the mites that were placed in categories separate from mites damaged by bees. This type of classification helps
identifying damage to mites and comparing them with developmental origin symptoms, and may provide criteria for selecting
bees tolerant or resistant to this mite. 相似文献
Biology, morphology and oviposition behavior of Anagrus atomus (Linnaeus), an egg parasitoid of the grape leafhopper Arboridia kermanshah Dlabola in Isfahan, Iran, were investigated. Adults were smaller than those so far reported from other regions. Females continuously drummed on plant surfaces with their antennae to search for host eggs. Parasitoid eggs hatched 2–3 days after oviposition, and A. atomus had two larval instars. First instar larvae were sacciform and immobile. Second instar larvae appeared 4 days after oviposition and were very active, and doubled their body length. The prepupal and pupal stages lasted for 1 and 5–6 days, respectively. Adult emergence began 16 days after oviposition, and peaked on day 17. 相似文献
A lysosomal pepstatin-insensitive proteinase (CLN2p) deficiency is the underlying defect in the classical late-infantile neuronal ceroid lipofuscinosis (LINCL, CLN2). The natural substrates for CLN2p and the causative factors for the neurodegeneration in this disorder are still not understood. We have now purified the CLN2p from bovine brain to apparent homogeneity. The proteinase has a molecular mass of 46 kDa and an aminoterminal sequence, L-H-L-G-V-T-P-S-V-I-R-K, that is identical to the human enzyme. Peptide: N-glycosidase F and endoglycosidase H treatment of the CLN2p reduced its molecular mass to 39.5 and 40.5 kDa, respectively, suggesting the presence of as many as five N-glycosylated residues. The CLN2p activity was not affected by common protease inhibitors, and thiol reagents, metal chelators, and divalent metal ions had no significant effect on the proteolytic activity of the CLN2p. Among the naturally occurring neuropeptides, angiotensin II, substance P, and beta-amyloid were substrates for the CLN2p, whereas angiotensin I, Leu-enkephalin, and gamma-endorphin were not. Peptide cleavage sites indicated that the CLN2p is a tripeptidyl peptidase that cleaves peptides having free amino-termini. Synthetic amino- and carboxyl-terminal peptides from the subunit c sequence, which is the major storage material in LINCL, are hydrolyzed by the CLN2p, suggesting that the subunit c may be one of the natural substrates for this proteinase and its accumulation in LINCL is the direct result of the proteinase deficiency. 相似文献
Plant-derived smoke plays a key role in plant growth. Proteomic technique was used for underlying mechanisms of plant-derived smoke on the growth of soybean (Glycine max L.) under flooding stress. The length and weight of soybean root increased with 2000 parts per million plant-derived smoke under flooding stress within 4 days. Altered proteins by plant-derived smoke treatment under flooding stress were mainly related to protein metabolism, stress, and redox. Furthermore, proteins related to mitochondrial electron transport chain decreased by flooding stress; however, they increased by addition of plant-derived smoke under flooding stress. Based on the results of proteomic analysis, confirmation experiments were performed. ATPase abundance and ATP content increased with the treatment of plant-derived smoke under flooding stress. Furthermore, the ascorbate/glutathione cycle was activated with the treatment of plant-derived smoke under flooding stress. These results suggest that plant-derived smoke improves the root growth of soybean with energy production and reactive oxygen scavenging even if it is under flooding stress, which might positively regulate soybean tolerance towards flooding stress.
