Abstract

Helicobacter pylori (H. pylori) is one of the most extensively studied Gram-negative bacteria due to its implication in gastric cancer. The oncogenicity of H. pylori is associated with cytotoxin-associated gene A (CagA), which is injected into epithelial cells lining the stomach. Both the C- and N-termini of CagA are involved in the interaction with several host proteins, thereby disrupting vital cellular functions, such as cell adhesion, cell cycle, intracellular signal transduction, and cytoskeletal structure. The N-terminus of CagA interacts with the tumor-suppressing protein, apoptosis-stimulating protein of p53 (ASPP2), subsequently disrupting the apoptotic function of tumor suppressor gene p53. Here, we present the in-depth molecular dynamic mechanism of the CagA–ASPP2 interaction and highlight hot-spot residues through in silico mutagenesis. Our findings are in agreement with previous studies and further suggest other residues that are crucial for the CagA–ASPP2 interaction. Furthermore, the ASPP2-binding pocket possesses potential druggability and could be engaged by decoy peptides, identified through a machine-learning system and suggested in this study. The binding affinities of these peptides with CagA were monitored through extensive computational procedures and reported herein. While CagA is crucial for the oncogenicity of H. pylori, our designed peptides possess the potential to inhibit CagA and restore the tumor suppressor function of ASPP2.  相似文献

In the present study, the regeneration pathway, especially the different events of somatic embryogenesis (SE) have been studied morphologically and biochemically in Catharanthus roseus. Firstly, the calluses were induced from different explant sources (hypocotyl, epicotyl and root) by using various auxins. Embryogenic and non-embryogenic calluses were identified based on their morphology, colour and dry weight. Embryogenic callus was later cultivated on MS added with 0.45 μM 2,4-D, 6.62 μM BAP and 1.44 μM GA3 for obtaining various developmental stages of embryos. Different stages of embryos have been assayed for the establishment of marker based embryogenesis, particularly on embryo specific proteins whose presence or absence will ensure a rapid and efficient production of embryos that has a special application to clonal biotechnology. Two embryo specific proteins (38 and 33 kD) have been identified for the first time in C. roseus during torpedo stage of embryogenesis. Besides, multiple shoot formation from in vitro raised emblings was also attempted to examine the role of BAP and kinetin for shoot proliferation. The shoots were rooted with 5.37 μM NAA and 5.71 μM IAA before transplantation.  相似文献

High energy density, fast recharging ability, and sustained cycle life are the primary requisite of supercapacitors (SCs); these necessities can be fulfilled by engineering a smart current collector with hierarchical combination of different active materials. This study reports a multicomponent design of hierarchical zinc cobalt sulfide (ZCS) hollow nanotube arrays wrapped with interlaced ultrathin Ni(OH)2 nanoflakes for high‐performance electrodes. The ZCS exhibits a unique pentagonal cross‐section and a rough surface that facilitates the deposition of Ni(OH)2 nanoflakes with a thickness of 7.5 nm. The ZCS/Ni(OH)2 hierarchical electrode exhibits a high specific capacitance of 2156 F g?1 and excellent cyclic stability with 94% retention over 3000 cycles. This is attributed to enhanced redox reactions, the direct growth of arrays on 3D porous foam acting as a “superhighway” for electron transport, and the increased availability of electrochemical active sites provided by the ultrathin Ni(OH)2 flakes that also sustain the stability of the electrode by sacrificing themselves during long charge/discharge cycles. Symmetric SCs are assembled to achieve high energy density of 74.93 W h kg?1 and exhibit superior cyclic stability of 78% retention with 81% coulombic efficiency over 10 000 cycles.  相似文献

During the preoviposition and oviposition periods of ixodid ticks, the salivary glands degenerate. It is unclear whether this is a necrotic or a programmed cell death event. We used an in situ TUNEL technique to determine if salivary gland degeneration involves apoptosis. Salivary glands were dissected from replete females at days 3, 5, 8, 11, 13, and 33 post-detachment. There were no differences in tick weight at detachment, suggesting that changes were not due to engorgement abnormalities. The onset of apoptosis began at day 5 and continued through oviposition at day 33. The greatest amount of nuclei containing fragmented DNA was observed on day 8 post-detachment, suggesting this was the peak occurrence of programmed cell death. Further, the temporal organization of programmed cell death suggests that the granule-secreting acini undergo apoptosis first, and that during the first week of oviposition the type I acini do not exhibit programmed cell death. These data suggest that the type I acini may still function in maintaining off-host hydration state of ovipositing females. These data provide morphological evidence that salivary gland degeneration involves a temporal programmed cell death event.  相似文献

Neuropathic pain is a debilitating form of treatment-resistant chronic pain caused by damage to the nervous system. Cannabinoids have been known for suppressing neuropathic pain by modulating the endo cannabinoid system. Since the canonical Wnt/β-catenin signaling has recently been implicated in pain sensation, we investigated the impact of major cannabinoids (16) from the leaves of Cannabis sativa and an epoxy derivative of compound 2, here upon referred to as 2a, on modulating Wnt/β-catenin signaling pathway. The results presented in this study show that compound 1, 2 and 2a exhibited potent inhibitory activity against Wnt/β-catenin pathway in a dose-dependent manner. Compound 2a was seen to inhibit this pathway at slightly lower concentrations than its parent molecule 2, under similar conditions. Taken together, compound 1, 2 and 2a, by virtue of their inhibition of Wnt-catenin signaling pathway, could be developed as effective neuroprotective agents for the management of neuropathic pain.  相似文献

An efficient somatic embryogenesis system has been established in Catharanthus roseus (L.) G. Don in which primary and secondary embryogenic calluses were developed from hypocotyls and primary cotyledonary somatic embryos (PCSEs), respectively. Two types of calluses were different in morphology and growth behaviour. Hypocotyl-derived embryogenic callus (HEC) was friable and fast-growing, while secondary callus derived from PCSE was compact and slow-growing. HEC differentiated into somatic embryos which proliferated quickly on medium supplemented with NAA (1.0 mg l−1) and BA (1.5 mg l−1). Although differentiation and proliferation of somatic embryos were faster in primary HEC, maturation and germination efficiency were better in somatic embryos developed from primary cotyledonary somatic embryo-derived secondary embryogenic callus (PCSEC). At the biochemical level, two somatic embryogenesis systems were different. Both primary and secondary/adventive somatic embryogenesis and the role of plant growth regulators in two modes of somatic embryo formation have been discussed.  相似文献

Taxonomic identification and classifications of insect pest genera Chrysodeixis and Ctenoplusia of the subfamily Plusiinae is very compulsory due to their phytophagous nature and potential to damage the cash as well as cereal crops. Taxonomy plays a key role in proper not only in identification and classification of the pest but also in designing a successful managing strategy. In current study, specimens of Chrysodeixis and Ctenoplusia genera were collected from different geographical areas of south Punjab, Pakistan and their diagnostic features were examined following taxanomic keys. The data of temperature, relative humidity and rainfall were also recorded during the study period. Genitalia was extracted by dissecting of the abdomen and inspected under Stereo microscope. The results revealed two new species, Chryodeixis maxus and Ctenoplusia oleraceaus, from south Punjab region in addition to previously reported species: Chrysodeixis furihatai. Suitable management of the voracious insect pest at appropriate time may help in sustaining the agriculture in Pakistan.  相似文献
