Theiler's murine encephalomyelitis virus is divided into two subgroups, TO and GDVII, inducing subgroup‐specific diseases. In order to investigate the role(s) of nonstructural proteins of TMEV, L and L*, leaders of two subgroups, were separately expressed with or without L* in BHK‐21 cells. Expression of L increased the number of apoptotic cells. L*/BHK‐21 cells constitutively expressing L* showed the decrease in cell death induced by L. These results suggest that L and L* regulate apoptosis during viral infection and contribute to TMEV subgroup‐specific biological activities. 相似文献

Interleukin-8 (IL-8) has been reported to promote tumor cell growth in colon cancer cells after binding to its receptors, which are members of the G-protein coupled receptor (GPCR) family. Recent studies demonstrated that stimulation of GPCR can induce shedding of epidermal growth factor (EGF) ligands via activation of a disintegrin and metalloprotease (ADAM), with subsequent transactivation of the EGF receptor (EGFR). In this study, we investigated mechanisms of cell proliferation and migration stimulated by IL-8 in a human colon carcinoma cell line (Caco2). IL-8 increased DNA synthesis of Caco2 in a dose dependent manner and this was inhibited by ADAM, EGFR kinase, and MEK inhibitors. IL-8 transiently induced EGFR tyrosine phosphorylation after 5-90 min and this was completely inhibited by ADAM inhibitor. Neutralizing antibody against HB-EGF as a key ligand for EGFR also blocked transactivation of EGFR and cell proliferation by IL-8. Since IL-8-induced cell migration was further suppressed by the ADAM inhibitor and the HB-EGF neutralizing antibody, our data indicate that IL-8 induces cell proliferation and migration by an ADAM-dependent pathway, and that HB-EGF plays an important role as the major ligand for this pathway. 相似文献

The LET-RBE spectra for cell killing for cultured mammalian cells exposed to accelerated heavy ions were investigated to design a spread-out Bragg peak beam for cancer therapy at HIMAC, National Institute of Radiological Sciences, Chiba, prior to clinical trials. Cells that originated from a human salivary gland tumor (HSG cells) as well as V79 and T1 cells were exposed to (3)He-, (12)C- and (20)Ne-ion beams with an LET ranging from approximately 20-600 keV/micrometer under both aerobic and hypoxic conditions. Cell survival curves were fitted by equations from the linear-quadratic model and the target model to obtain survival parameters. RBE, OER, alpha and D(0) were analyzed as a function of LET. The RBE increased with LET, reaching a maximum at around 200 keV/micrometer, then decreased with a further increase in LET. Clear splits of the LET-RBE or -OER spectra were found among ion species and/or cell lines. At a given LET, the RBE value for (3)He ions was higher than that for the other ions. The position of the maximum RBE shifts to higher LET values for heavier ions. The OER value was 3 for X rays but started to decrease at an LET of around 50 keV/micrometer, passed below 2 at around 100 keV/micrometer, and then reached a minimum above 300 keV/micrometer, but the values remained greater than 1. A simple and sensitive procedure for the quantitative estimation of Triton X-100 is described. The method is based on the formation of turbidity from Triton X-100 with phenol. The turbidity is proportional to Triton X-100 in a range of 20–80 μg/ml. Protein, mucopolysaccharide, and nucleic acid do not interfere in this turbidity formation. The method is especially useful for detection of the residue after the removal of Triton X-100 from solubilized samples. 相似文献

Recent rapid development of mechanization and automatization in many industries in Japan have increased simple and repetitive tasks that have resulted in some new types of occupational health hazards, in particular among young female workers. One of them is occupational cervicobrachial disorder, the concept of which was established by the committee organized by the Japan Association of Industrial Health in 1972. The committee demonstrated that the disorder resulted from repetitive upper limb manipulation in punching and operating various kinds of business machines, and concluded that the disorder was closely related to chronic fatigue in the shoulders, neck, lower back and other parts of the body. In 1975 a study team was organized with a grant from the Ministry of Education. The team was divided into four working groups. Topics of these groups were 1) etiology of the disorder, 2) natural history of the disorder in various kinds of jobs, 3) mass examination techniques for the disorder, and 4) a health care system to be provided for the workers. The persent report summarizes the results of group discussions with a view to indicating the current problems of occupational cervicobrachial disorder and the necessary future steps for its care treatment and prevention. 相似文献
