为鉴定香蕉枯萎病菌(尖孢镰刀菌古巴专化型4号生理小种,Fusarium oxysporum f.sp.cubense race 4,Foc4)中的2个假想谷胱甘肽S转移酶(GSTs),采用RT-PCR方法克隆了这2个GSTs基因cDNA编码序列,随后分别将2个基因定名为Fogst1和Fogst2。其中,Fogst1的开放阅读框长609 bp,编码202个氨基酸残基,Fogst2的开放阅读框长693 bp,编码230个氨基酸残基。进化树分析表明:Fogst1属于GSTs超家族的sigma(σ)亚型成员,Fogst2属于GSTs超家族中目前未知的亚家族成员。为了验证Fogst1和Fogst2的表达,分别构建了Fogst1和Fogst2的原核表达重组载体pET28a-Fogst1和pET28a-Fogst2,并将pET28a-Fogst1和pET28a-Fogst2转化到大肠杆菌表达菌株BL21,经IPTG诱导后获得以可溶形式表达的重组蛋白Fogst1和Fogst2。GSTs活性分析表明,以CDNB为底物检测,2个重组蛋白均具有GSTs酶活性。分别取外源氧化胁迫处理后1、5、12、24 h菌丝样品进行相对荧光定量PCR分析,结果表明:Fogst1和Fogst2在前5 h表达量均大幅上调,表达量随后下调并恢复正常水平。这些结果均暗示Fogst1和Fogst2可能参与了Foc4抗外源氧化胁迫过程。  相似文献

构建了8个PLZF-RARα融合基因突变体.用“滞后”胶实验证实PLZF-RARα与PML-RARα一样,亦能以同二聚体的形式结合到维甲酸反应元件(RARE)上,且PLZF的POZ结构域介导PLZF-RARα同二聚体的形成和稳定.但两者与RARE的结合类型存在差异,在DR5G,PML-RARα的结合强度大于PLZF-RARα;而在DR5T,则是PLZF-RARα强于PML-RARα.进一步工作证实PLZF-RARα能与RXR形成异二聚体,并产生4种复合物.用免疫沉淀法发现PLZF-RARα亦能与PLZF形成异二聚体,而且也是通过POZ结构域介导PLZF-RARα和PLZF异二聚体的形成.同PML-RARα一样,PLZF-RARα对RARE的结合反应亦受维甲酸调控.  相似文献

利用鱼类远缘杂交和雌核发育相结合的方法培育出改良四倍体鲫鲤和改良二倍体红鲫,两者交配制备出一种改良三倍体鲫鱼(湘云鲫2号).对改良三倍体鲫鱼的染色体数目和组型、性腺和垂体结构、外形特征、生长速度等方面进行了系统的研究.结果表明,改良三倍体鲫鱼的染色体数目为3n=150,核型公式为33m+51sm+33st+33t;其精巢和卵巢在繁殖季节不能产生成熟配子,垂体超微结构显示,GTH细胞内的分泌颗粒和分泌小球在繁殖季节没有大量排出,该特征从内分泌的角度证明了它们的不育性;改良三倍体鲫鱼具有体背高、尾柄短和头部小的优良外形特征.与三倍体湘云鲫相比,改良三倍体鲫鱼不但保留了三倍体鱼生长速度快、不育等特点,而且在体形方面表现出明显的改良特征,是一种新型改良三倍体鲫鱼.  相似文献

为了提高人血小板因子 4(humanplateletfactor 4,hPF4)的表达 ,在PT7 7 hPF4表达质粒的基础上 ,采用PCR定位突变技术 ,改造人血小板因子 4(hPF4)cDNA基因片段 ,去除cDNA 3′端非翻译区AT富含序列 ,改用大肠杆菌强串联终止密码子TAATAA ,成功构建了高效表达质粒pBV2 2 0 hPF4。摇瓶发酵重组人血小板因子 4的产量达 1 60mg L较原表达质粒PT7 7 hPF4表达量提高了近 80倍。经包涵体的洗涤、变性、复性后 ,采用鸡胚绒毛尿囊膜血管生成抑制实验测定复性后rhPF4的生物学活性 ,结果显示 :rhPF4具有抑制血管生成活性。  相似文献

涡度相关法被认为是测定农田蒸散量的标准方法。然而,能量不闭合现象在涡度相关测量中普遍存在。分析能量不闭合对涡度相关观测的影响,对于提高涡度相关观测精度具有重要意义。以蒸渗仪法为参照,探讨涡度相关观测的能量闭合状况对农田蒸散测定的影响,在导致涡度相关观测能量不闭合的诸多因素中,寻找对蒸散测定有影响的因素。结果表明:涡度相关观测的白天能量平衡比率(EBR)呈秋冬高、春夏低的变化特征,麦季日均EBR范围在0.26—2.84之间,平均1.15;玉米季日均EBR范围在0.19—2.59之间,平均0.78。无论麦季或玉米季,涡度相关法测定的平均蒸散量(ETec)均明显低于蒸渗仪法观测值(ETL),但两者显著相关(P<0.01),并有相似的季节变化。平均蒸散比(ETec/ETL)麦季约为0.61,玉米季约为0.50。在冬小麦田和夏玉米田,ETec/ETL均与EBR显著相关(P<0.01)。麦田种植密度大,下垫面较均匀,蒸散比与EBR成正比(P<0.01),且不受叶面积指数(LAI)大小影响;反之,玉米田种植密度小,只有当LAI>1,下垫面变得较均匀后,蒸散比与EBR的关系才变得显著(P<0.01)。风速小时ETec/ETL与EBR显著相关,风速增加时二者相关性减弱。尤其在玉米田,当摩擦风速(u*)大于0.3 m/s时,ETec/ETL与EBR的相关性不再显著。风速小时,大气湍流微弱,湍流的涡旋较大。在有限的观测时段(0.5h)内,涡度相关仪的传感器难以捕捉足够的湍涡能量,所测湍流能量偏低,导致能量不闭合。以上结果为应用能量平衡比率校正农田蒸散提供了可能途径。  相似文献

Reliable information about soil conservation capacities of different natural ecosystems is an important reference for the design of targeted erosion and sediment control strategies. The objective of this paper is to quantify the soil conservation capacities of different natural ecosystems that can represent different climatic zones. The 137Cs technique has been used to estimate soil redistribution rates in different natural ecosystems over the past 40 years in Wolong Nature Reserve. The reserve, transiting from the Chengdu plain to the Qinghai-Tibet plateau, maintains rich ecosystems from subtropical to frigid.The net soil erosion rates of 5 selected ecosystems that represent a warm coniferous-broadleaf-mixed forest, a cold-resistant deciduous taiga forest, a cold-resistant shrub, an evergreen cold-resistant taiga servation capacities are reversed in order. The reference inventories for 137Cs in different ecosystems velop effective erosion and sediment strategies in areas with similar climates should consider natural ecosystem types.  相似文献

A series of novel nitrofuranyl methyl N-heterocycles based on the structure of IIIM-MCD-211 were designed and synthesized. Compounds 6d, 8b and 12a show excellent activity against MTB H37Rv strain (MIC: 0.031–0.062?μg/mL) roughly comparable to INH and IIIM-MCD-211. In addition, a three-dimensional quantitative structure-activity relationship (3D-QSAR) study was performed on the above mentioned chemical series employing comparative molecular field analysis (CoMFA) and comparative molecular similarity index analysis (CoMSIA) techniques. The developed CoMFA and CoMSIA models display high external predictability (r2pred of 0.954 and 0.935, respectively) and good statistical robustness. More importantly, the newly designed compounds 16a and 16b (MIC: <0.016?μg/mL) based on the two models, as expected, were found to be more active than 12a and IIIM-MCD-21. Design and synthesis of more potent nitrofuranyl methyl N-heterocycles as anti-TB agents are currently in progress.  相似文献

Cellulose samples extracted from cornhusk have been successfully acetylated in an ionic liquid 1-allyl-3-methylimidazolium chloride (AmimCl). Without using any catalyst, cornhusk cellulose acetates (CCAs) with the degree of substitution (DS) in a range from 2.16 to 2.63 were prepared in one-step. Under the homogeneous state, the DS value of CCAs was easily controlled by the acetylation time. The obtained CCAs were characterized by means of FT-IR, 13C NMR, DSC, TGA, and a mechanical test. The NMR results showed that the distribution of the acetyl moiety among the three OH groups of the anhydroglucose unit shows a preference at the C6 position. The CCAs exhibited good solubility in some organic solvents, such as acetone and DMSO. The cast CCA films from their acetone solutions had good mechanical properties. At the end of each acetylation of cornhusk cellulose, the ionic liquid AmimCl could be effectively recovered. Therefore, this study presents a promising approach and “green process” to make use of crop by-products.  相似文献

Entomopathogenic fungi have great potential to control agricultural and horticultural insect pests, however optimizing conidial production systems to demonstrate high productivity and stability still needs additional efforts for successful field application and industrialization. Although many virulent entomopathogenic fungal isolates have been viewed as potential candidates in a laboratory environment, very few of the isolates are being used in practice for application in agricultural fields as commercial products. I. javanicus is an entomopathogenic fungus that is parasitic to various diverse coleopteran and lepidopteran insects and thought good candidate as biopesticdes. In this work, the basic characteristics of two entomopathogenic fungi, I. javanica FG340 and Pf04, were investigated in morphological examinations, genetic identification, and virulence against Thrips palmi, and then the feasibility of various grains substrates for conidial production was assessed, particularly focusing on conidial productivity and thermotolerance. Isaria javanica FG340 and Pf04 conidia were solid-cultured on 12 grains for 14?days in a Petri dish. Of the tested Italian millet, perilla seed, millet and barley-based cultures showed high conidial production. The four-grain media yielded >1?×?109 conidia/g of I. javanica FG340 and Pf04. Pf04 strain had enhanced thermotolerance up to 45?°C when cultured on Italian millet. In application, it was easy to make a conidial suspension using the cultured grains, and several surfactants were tested to release the conidia. This work suggests several possible inexpensive grain substrates by which to promote conidial production combined with enhanced stability against exposure to high temperature.  相似文献
