Nonfat dried milk is routinely used as a blocking agent and diluent in immunoassays. However, the data presented in this paper demonstrate that milk contains an inhibitor of the biotin-streptavidin interaction. It is recommended that milk be dialyzed or used at lower concentrations when it is employed as a diluent of streptavidin.  相似文献

Resource allocation patterns, as quantified by residual food intake (RFI), and the consequences for offspring development were investigated during lactation in 96 females of a mouse line selected for 104 generations for high litter size at birth (S-line) and in 87 females of a non-selected control line (C-line). Litters of 45 C-line dams (Cs) and 48 S-line dams (Ss) were standardised (s) at birth; other dams (ns) supported total number of pups born (Cns and Sns, respectively). RFI during lactation was significantly lower in Sns-dams than in C-line dams and Sns-dams. After weaning Sns-dams seemed to be able to restore the negative resource situation. Sns-pups were about 25% less mature than Cns-pups at all times. Maturity was similar for Cs- and Ss-pups from 2 d in lactation on, and about 18% and 53% higher than Cns- and Sns-pups. The pre-weaning mortality rate was significantly higher in Sns-litters (35.6 ± 2.76) than in Cns-litters (4.95 ± 2.23). The results suggest that S-line dams allocated considerably more resources to maintenance of offspring than C-line dams. This was insufficient to provide the offspring with an adequate amount of resources, resulting in reduced pup development and increased pre-weaning mortality rates.  相似文献

Increasing temperature and drought intensity is inducing the phenomenon of the so-called “hotter drought”, which is expected to increase in frequency over the coming decades across many areas of the globe, and is expected to have major implications for forest systems. Consequences of hotter drought could be especially relevant for closely related species overlapping their distributions, since differences in response can translate into range shifts. We assessed the effect of future climatic conditions on the performance of five ecologically distinct pine species common in Europe: Pinus halepensis, P. pinaster, P. nigra, P. sylvestris and P. uncinata. We hypothesised that Mediterranean species inhabiting dry, low-elevation sites will be less affected by the expected warming and drought increase than species inhabiting cold-wet sites. We performed a controlled conditions experiment simulating current and projected temperature and precipitation, and analysed seedling responses in terms of survival, growth, biomass allocation, maximum photochemical efficiency (F v/F m) and plant water potential (Ψ). Either an increase in temperature or a reduction in water input alone reduced seedling performance, but the highest impact occurred when these two factors acted in combination. Warming and water limitation reduced Ψ, whereas warming alone reduced biomass allocation to roots and F v/F m. However, species responded differentially to warmer and drier conditions, with lowland Mediterranean pines (P. halepensis and P. pinaster) showing higher survival and performance than mountain species. Interspecific differences in response to warmer, drier conditions could contribute to changes in the relative dominance of these pine species in Mediterranean regions where they co-occur and a hotter, drier climate is anticipated.  相似文献

A histochemical assay for myofibrillar adenosine triphosphatase (mATPase) activity is routinely utilized in the delineation of fiber types in healthy human skeletal muscle. Each fiber type has a specific pH range of mATPase stability (activation). Outside of this pH range, mATPase activity is labile (inactivated), no reaction product is formed, and the fibers remain unstained. The aim of the present study was to carefully investigate the pH stability/lability of mATPase in postmortem muscles. To this end, vastus lateralis muscle samples were obtained approximately 0.5, 1, 2, 3, and 4 days after death, as well as control samples from a healthy young man and woman. Serial cross sections of the muscle samples were assayed for mATPase activity throughout preincubation pH ranges of 4.15-4.7 and 10.2-10.5 in increments of 0.05 pH units. Myosin heavy chain analysis (as well as a regression analysis comparing fiber type area and relative myosin heavy chain content) verified the mATPase-based fiber types. The pH ranges of mATPase stability/lability for the control samples were as previously reported, and support the use of preincubation pH values of 4.3, 4.6, and 10.4 for the delineation of fiber types in normal human muscle. For the postmortem samples, both quantitative and qualitative changes altered the pH ranges of mATPase activation/inactivation. Quantitative changes consisted of a time-dependent loss of mATPase activity that was inhibited in all fibers outside the pH range of 4.15-10.50. In addition, qualitative changes caused "shifts to the left" in mATPase stability within the fast fiber types (IIA and IIB). As such, complete inhibition of mATPase activity did not occur until preincubation at pH 4.45 and pH 4.30 for fiber types IIA and IIB, respectively. For the postmortem vastus lateralis muscle samples, optimal preincubation pH values for mATPase-based fiber type delineation were pH 4.30, 4.45, and 10.35. The reason for these qualitative changes in mATPase stability is not known. However, postmortem changes such as increased lactate production and marked acidification may play a role.  相似文献

Grass was field-dried to 3 different dry matter (DM) levels (200, 430 and 540 g/kg) and inoculated with 106–107 cfu/g of a Listeria monocytogenes strain sharing a phagovar occasionally involved in food-borne outbreaks of listeriosis. Formic acid (3 ml/kg) or lactic acid bacteria (8·105/g) with cellulolytic enzymes were applied only to forages with low and intermediate DM levels. Forages were ensiled in laboratory silos (1700 ml) and were stored at 25°C for 30 or 90 days. After 90 days of storage, L. monocytogenes could not be detected in any silo, except one with the high dry matter grass without additive. After 30 days of storage, between 102 and 106 cfu L. monocytogenes/g silage were isolated from the untreated silages. Increasing the DM content from 200 to 540 g/kg did not reduce listeria counts possibly because of the lower production of fermentation acids (higher pH). In silages treated with additives, counts of L. monocytogenes were always lower than in silages without additive. In wet silages (DM 200 g/kg) both additives were effective, but in the wilted silages (DM 430 g/kg) only the bacterial additive reduced listeria counts below detection level. Listeria counts were highly correlated to silage pH (r = 0.92), the concentration of lactic acid (r = -0.80) and the pooled amount of undissociated acids (r = -0.83).  相似文献

Epidemiological studies on Greenland Inuits in the 1970s and subsequent human studies have established an inverse relationship between the ingestion of omega-3 fatty acids [C20–22 ω 3 polyunsaturated fatty acids (PUFA)], blood levels of C20–22 ω 3 PUFA, and mortality associated with cardiovascular disease (CVD). C20–22 ω 3 PUFA have pleiotropic effects on cell function and regulate multiple pathways controlling blood lipids, inflammatory factors, and cellular events in cardiomyocytes and vascular endothelial cells. The hypolipemic, anti-inflammatory, anti-arrhythmic properties of these fatty acids confer cardioprotection. Accordingly, national heart associations and government agencies have recommended increased consumption of fatty fish or ω 3 PUFA supplements to prevent CVD. In addition to fatty fish, sources of ω 3 PUFA are available from plants, algae, and yeast. A key question examined in this review is whether nonfish sources of ω 3 PUFA are as effective as fatty fish-derived C20–22 ω 3 PUFA at managing risk factors linked to CVD. We focused on ω 3 PUFA metabolism and the capacity of ω 3 PUFA supplements to regulate key cellular events linked to CVD. The outcome of our analysis reveals that nonfish sources of ω 3 PUFA vary in their capacity to regulate blood levels of C20–22 ω 3 PUFA and CVD risk factors.  相似文献
