The existence of Salmonella enterica serovar Typhimurium viable-but-nonculturable (VBNC) cells is a public health concern since they could constitute unrecognized sources of infection if they retain their pathogenicity. To date, many studies have addressed the ability of S. Typhimurium VBNC cells to remain infectious, but their conclusions are conflicting. An assumption could explain these conflicting results. It has been proposed that infectivity could be retained only temporarily after entry into the VBNC state and that most VBNC cells generated under intense stress could exceed the stage where they are still infectious. Using a Radioselectan density gradient centrifugation technique makes it possible to increase the VBNC-cell/culturable-cell ratio without increasing the exposure to stress and, consequently, to work with a larger proportion of newly VBNC cells. Here, we observed that (i) in the stationary phase, the S. Typhimurium population comprised three distinct subpopulations at 10, 24, or 48 h of culture; (ii) the VBNC cells were detected at 24 and 48 h; (iii) measurement of invasion gene (hilA, invF, and orgA) expression demonstrated that cells are highly heterogeneous within a culturable population; and (iv) invasion assays of HeLa cells showed that culturable cells from the different subpopulations do not display the same invasiveness. The results also suggest that newly formed VBNC cells are either weakly able or not able to successfully initiate epithelial cell invasion. Finally, we propose that at entry into the stationary phase, invasiveness may be one way for populations of S. Typhimurium to escape stochastic alteration leading to cell death.Like several readily culturable pathogenic bacterial species, Salmonella enterica has been shown to enter into a viable-but-nonculturable (VBNC) state in response to environmental stresses (25, 33). In this state, cells display integrity and activities but escape detection by conventional culture-based monitoring (24). The physiological significance of this phenotype is unclear: some authors have proposed that it is part of an adaptive response aimed at long-term survival under adverse conditions (22, 32); others argue that it is a consequence of stochastic cellular deterioration and that VBNC cells are on their way to death (4, 10, 12, 23). In any case, the existence of VBNC pathogens is a public health concern since they may constitute unrecognized sources of infection if they retain their pathogenicity.To date, many studies have addressed the ability of VBNC pathogens to remain infectious, but the conclusions of some investigators are conflicting (15, 36). In vitro experiments have shown that VBNC cells of Salmonella enterica serovar Typhimurium and Salmonella enterica serovar Oranienburg can recover their culturability (13, 27, 30, 31). This phenomenon, called resuscitation, confirms that at least some VBNC cells ultimately remain able to multiply and are therefore potentially infectious. On the other hand, most in vivo studies ruled out the ability of S. Typhimurium VBNC cells to initiate infection in mice and chicken or to resuscitate during their passage in the animal gut (6, 17, 34, 35). However, one study reported evidence of the maintenance of pathogenicity by VBNC cells of S. Oranienburg in a model of morphine-immunosuppressed mice (1). An assumption could explain these apparently opposite results. It has been proposed that infectivity could be retained only temporarily after entry into the VBNC state (8, 19, 26). Experiments intended for testing the ability of VBNC cells to retain their pathogenicity cannot be fully conclusive if the inocula still contain culturable cells. Therefore, all previously published animal experiments with S. Typhimurium were conducted on populations with VBNC-cell/culturable-cell ratios around 10,000:1. Such populations were obtained after strong exposure to stress, either under intense stressing factors for a short period (e.g., germicidal UV-C for 2 min [6]) or under mild stressing factors for a long period (e.g., starvation for a minimum of 1 week [35]). In such populations, most VBNC cells could exceed the stage where they are still infectious, and the negative outcomes of infection studies could actually reflect their inability to specifically address the fraction of recent VBNC cells.A Radioselectan density gradient centrifugation technique was shown to fractionate stationary-phase populations of Escherichia coli into two subpopulations (10, 12, 18). Interestingly, the VBNC cells formed during a 48-h E. coli culture were specifically recovered in the high-density (HD) subpopulation (12). This technique thus gives the opportunity to increase the VBNC-cell/culturable-cell ratio without increasing exposure to stress and, consequently, to work with a larger proportion of cells having recently entered the VBNC state.Here, this technique was used to discriminate different stationary-phase S. Typhimurium subpopulations. We further investigated the invasiveness of these cell subpopulations by using both gene expression assays of invasion genes and in vitro invasion tests. Human arylamine N-acetyltransferase 1 (NAT1) is a xenobiotic-metabolizing enzyme that biotransforms aromatic amine chemicals. We show here that biologically-relevant concentrations of inorganic (Hg2+) and organic (CH3Hg+) mercury inhibit the biotransformation functions of NAT1. Both compounds react irreversibly with the active-site cysteine of NAT1 (half-maximal inhibitory concentration (IC50) = 250 nM and kinact = 1.4 × 104 M−1 s−1 for Hg2+ and IC50 = 1.4 μM and kinact = 2 × 102 M−1 s−1 for CH3Hg+). Exposure of lung epithelial cells led to the inhibition of cellular NAT1 (IC50 = 3 and 20 μM for Hg2+ and CH3Hg+, respectively). Our data suggest that exposure to mercury may affect the biotransformation of aromatic amines by NAT1. 相似文献

The ability of Interleukin-15 (IL-15) to activate many immune antitumor mechanisms renders the cytokine a good candidate for the therapy of solid tumors, particularly renal cell carcinoma. Although IL-15 is being currently used in clinical trials, the function of the cytokine on kidney's components has not been extensively studied; we thus investigated the role of IL-15 on normal and tumor renal epithelial cells. Herein, we analyzed the expression and the biological functions of IL-15 in normal renal proximal tubuli (RPTEC) and in their neoplastic counterparts, the renal clear cell carcinomas (RCC). This study shows that RPTEC express a functional heterotrimeric IL-15Rαβγc complex whose stimulation with physiologic concentrations of rhIL-15 is sufficient to inhibit epithelial mesenchymal transition (EMT) commitment preserving E-cadherin expression. Indeed, IL-15 is not only a survival factor for epithelial cells, but it can also preserve the renal epithelial phenotype through the γc-signaling pathway, demonstrating that the cytokine possess a wide range of action in epithelial homeostasis. In contrast, in RCC in vitro and in vivo studies reveal a defect in the expression of γc-receptor and JAK3 associated kinase, which strongly impacts IL-15 signaling. Indeed, in the absence of the γc/JAK3 couple we demonstrate the assembly of an unprecedented functional high affinity IL-15Rαβ heterodimer, that in response to physiologic concentrations of IL-15, triggers an unbalanced signal causing the down-regulation of the tumor suppressor gene E-cadherin, favoring RCC EMT process. Remarkably, the rescue of IL-15/γc-dependent signaling (STAT5), by co-transfecting γc and JAK3 in RCC, inhibits EMT reversion. In conclusion, these data highlight the central role of IL-15 and γc-receptor signaling in renal homeostasis through the control of E-cadherin expression and preservation of epithelial phenotype both in RPTEC (up-regulation) and RCC (down-regulation). 相似文献

Plants are an invaluable source of potential new anti-cancer drugs. Here, we investigated the cytotoxic activity of the acetonic extract of Buxus sempervirens on five breast cancer cell lines, MCF7, MCF10CA1a and T47D, three aggressive triple positive breast cancer cell lines, and BT-20 and MDA-MB-435, which are triple negative breast cancer cell lines. As a control, MCF10A, a spontaneously immortalized but non-tumoral cell line has been used. The acetonic extract of Buxus sempervirens showed cytotoxic activity towards all the five studied breast cancer cell lines with an IC(50) ranging from 7.74 μg/ml to 12.5 μg/ml. Most importantly, the plant extract was less toxic towards MCF10A with an IC(50) of 19.24 μg/ml. Fluorescence-activated cell sorting (FACS) analysis showed that the plant extract induced cell death and cell cycle arrest in G0/G1 phase in MCF7, T47D, MCF10CA1a and BT-20 cell lines, concomitant to cyclin D1 downregulation. Application of MCF7 and MCF10CA1a respective IC(50) did not show such effects on the control cell line MCF10A. Propidium iodide/Annexin V double staining revealed a pre-apoptotic cell population with extract-treated MCF10CA1a, T47D and BT-20 cells. Transmission electron microscopy analyses indicated the occurrence of autophagy in MCF7 and MCF10CA1a cell lines. Immunofluorescence and Western blot assays confirmed the processing of microtubule-associated protein LC3 in the treated cancer cells. Moreover, we have demonstrated the upregulation of Beclin-1 in these cell lines and downregulation of Survivin and p21. Also, Caspase-3 detection in treated BT-20 and T47D confirmed the occurrence of apoptosis in these cells. Our findings indicate that Buxus sempervirens extract exhibit promising anti-cancer activity by triggering both autophagic cell death and apoptosis, suggesting that this plant may contain potential anti-cancer agents for single or combinatory cancer therapy against breast cancer. 相似文献

We examined if naturally recruited herbaceous vegetation in abandoned Belgian limestone quarries tend towards plant communities analogous to semi-natural habitats of conservation interest. We studied taxon-based assemblages (using two-dimensional non-metric multidimensional scaling ordination) and functional patterns (relative to Grime’s competitor, stress tolerator and ruderal plant strategies (CSR) classification) of plant communities (n = 360 plots) among three different time periods after quarry abandonment (< 3 y, 3–20 y, >?20 y). We compared those successional assemblages with those of habitat of conservation interest plant communities (n = 53 plots): lowland hay meadows and rupicolous, xerophilous and mesophilous calcareous grasslands. Our results indicate that naturally recruited herbaceous vegetation compositionally resembled mesophilous grassland, even though initial substrate conditions were more similar to rupicolous or xerophilous grasslands. The specific successional pathway we found in CSR state-space differs from Grime's predictions because there was a functional shift in plant assemblages from dominance by ruderals to dominance by stress-tolerant species. The differences in successional trajectories we found on different types of rock substrate suggest that conservation management should adopt a site-specific approach, recognizing that the highest probabilities of success on hard limestone will be restoration to calcareous grassland analogues. 相似文献

In this study multipotent adipose-derived stem cells isolated from human adipose tissue (hMADS cells) were shown to differentiate into adipose cells in serum-free, chemically defined medium. During the differentiation process, hMADS cells exhibited a gene expression pattern similar to that described for rodent clonal preadipocytes and human primary preadipocytes. Differentiated cells displayed the key features of human adipocytes, i.e., expression of specific molecular markers, lipolytic response to agonists of beta-adrenoreceptors (beta2-AR agonist > beta1-AR agonist > beta3-AR agonist) and to the atrial natriuretic peptide, insulin-stimulated glucose transport, and secretion of leptin and adiponectin. hMADS cells were able to respond to drugs as inhibition of adipocyte differentiation was observed in the presence of prostaglandin F2alpha, tumour necrosis factor-alpha, and nordihydroguaiaretic acid, a natural polyhydroxyphenolic antioxidant. Thus, for the first time, human adipose cells with normal karyotype and indefinite life span have been established. The identification of molecular motors that modulate the neuronal cytoskeleton has been elusive. Here, we show that a molecular motor protein, myosin Va, is present in high proportions in the cytoskeleton of mouse CNS and peripheral nerves. Immunoelectron microscopy, coimmunoprecipitation, and blot overlay analyses demonstrate that myosin Va in axons associates with neurofilaments, and that the NF-L subunit is its major ligand. A physiological association is indicated by observations that the level of myosin Va is reduced in axons of NF-L-null mice lacking neurofilaments and increased in mice overexpressing NF-L, but unchanged in NF-H-null mice. In vivo pulse-labeled myosin Va advances along axons at slow transport rates overlapping with those of neurofilament proteins and actin, both of which coimmunoprecipitate with myosin Va. Eliminating neurofilaments from mice selectively accelerates myosin Va translocation and redistributes myosin Va to the actin-rich subaxolemma and membranous organelles. Finally, peripheral axons of dilute-lethal mice, lacking functional myosin Va, display selectively increased neurofilament number and levels of neurofilament proteins without altering axon caliber. These results identify myosin Va as a neurofilament-associated protein, and show that this association is essential to establish the normal distribution, axonal transport, and content of myosin Va, and the proper numbers of neurofilaments in axons. 相似文献

Seeds from the African legume shrub Griffonia simplicifolia contain several lectins. Among them the tetrameric lectin GS I-B(4) has strict specificity for terminal alpha Gal residues, whereas the closely related lectin GS I-A(4) can also bind to alpha GalNAc. These two lectins are commonly used as markers in histology or for research in xenotransplantation. To elucidate the basis for the fine difference in specificity, the amino acid sequences of both lectins have been determined and show 89% identity. The crystal structure of GS I-B(4), determined at 2.5-A resolution, reveals a new quaternary structure that has never been observed in other legume lectins. An unexpected loss of both Ca(2+) and Mn(2+) ions, which are necessary for carbohydrate binding in legume lectins, may be related to a particular amino acid sequence Pro-Glu-Pro in the metal binding loop. Comparison with demetallized concanavalin A reveals a different process for the loss of metal ions and for the subsequent loss of carbohydrate binding activity. The GS I-A x alpha GalNAc and GS I-B x alpha Gal complexes were constructed using homology modeling and docking approaches. The unusual presence of an aromatic amino acid at position 47 (Tyr in I-A and Trp in I-B) explains the strong preference for alpha-anomeric sugars in both isolectins. Alteration at one amino acid position, Ala(106) in I-A versus Glu(106) in I-B, is the basis for the observed specificities toward alpha GalNAc and alpha Gal. 相似文献

A new fluorescent acridine orange derivative, 3-amino-6-methoxy-9-(2-hydroxyethylamino) acridine (AMHA), has been applied to Hela cells in order to set up appropriate conditions for the detection of mycoplasma contaminations. Since AMHA staining reveals intensely fluorescent nuclei and slight fluorescent cytoplasm, we can visualize and localize mycoplasma contamination on each cell. In combination with a shortened Chen's staining method (1977), AMHA should allow a better detection of mycoplasma in animal cell cultures than the well established Hoechst dye. 相似文献

To preserve biodiversity and ecosystem functions in a globally changing world it is crucial to understand the effect of land use on ecosystem processes such as pollination. Floral colouration is known to be central in plant-pollinator interactions. To date, it is still unknown whether land use affects the colouration of flowering plant communities. To assess the effect of land use on the diversity and composition of flower colours in temperate grasslands, we collected data on the number of flowering plant species, blossom cover and flower reflectance spectra from 69 plant communities in two German regions, Schwäbische Alb (SA) and Hainich-Dün (HD). We analysed reflectance data of flower colours as they are perceived by honeybees and studied floral colour diversity based upon spectral loci of each flowering plant species in the Maxwell triangle. Before the first mowing, flower colour diversity decreased with increasing land-use intensity in SA, accompanied by a shift of mean flower colours of communities towards an increasing proportion of white blossom cover in both regions. By changing colour characteristics of grasslands, we suggest that increasing land-use intensity can affect the flower visitor fauna in terms of visitor behaviour and diversity. These changes may in turn influence plant reproduction in grassland plant communities. Our results indicate that land use is likely to affect communication processes between plants and flower visitors by altering flower colour traits. 相似文献
