Warnericin RK is the first antimicrobial peptide known to be active against Legionella pneumophila, a pathogen bacterium that is responsible for severe pneumonia. Strikingly, this peptide displays a very narrow range of antimicrobial activity, almost limited to the Legionella genus, and a hemolytic activity. A similar activity has been described for δ-lysin, a well-known hemolytic peptide of Staphylococci that has not been described as antimicrobial. In this study we aimed to understand the mode of action of warnericin RK and to explain its particular target specificity. We found that warnericin RK permeabilizes artificial membranes in a voltage-independent manner. Osmotic protection experiments on erythrocytes showed that warnericin RK does not form well-defined pores, suggesting a detergent-like mode of action, as previously described for δ-lysin at high concentrations. Warnericin RK also permeabilized Legionella cells, and these cells displayed a high sensitivity to detergents. Depending on the detergent used, Legionella was from 10- to 1000-fold more sensitive than the other bacteria tested. Finally, the structure of warnericin RK was investigated by means of circular dichroism and NMR spectroscopy. The peptide adopted an amphiphilic α-helical structure, consistent with the proposed mode of action. We conclude that the specificity of warnericin RK toward Legionella results from both the detergent-like mode of action of the peptide and the high sensitivity of these bacteria to detergents.  相似文献

Oxidation of unsaturated lipids is a fundamental process involved in cell bioenergetics as well as in cell death. Using giant unilamellar vesicles and a chlorin photosensitizer, we asymmetrically oxidized the outer or inner monolayers of lipid membranes. We observed different shape transitions such as oblate to prolate and budding, which are typical of membrane curvature modifications. The asymmetry of the shape transitions is in accordance with a lowered effective spontaneous curvature of the leaflet being targeted. We interpret this effect as a decrease in the preferred area of the targeted leaflet compared to the other, due to the secondary products of oxidation (cleaved-lipids). Permeabilization of giant vesicles by light-induced oxidation is observed after a lag and is characterized in relation with the photosensitizer concentration. We interpret permeabilization as the opening of a pore above a critical membrane tension, resulting from the budding of vesicles. The evolution of photosensitized giant vesicle lysis tension was measured and yields an estimation of the effective spontaneous curvature at lysis. Additionally photo-oxidation was shown to be fusogenic.  相似文献

In polygynous ungulates, the rut imposes constraints on male time budgets that generate a trade‐off between maintenance and reproduction, leading to a reduction in time spent foraging. As mating activities can incur substantial somatic costs, males are expected to spend their ‘non‐rutting’ time recovering during the breeding season. If the diminution in time allocated to foraging by males is only a consequence of time budget constraints, males should keep a similar ratio of time spent foraging to lying to that observed in the pre‐rut, leading to an overall reduction of these two activities (the ‘foraging constraint’ hypothesis). Alternatively, if males adopt an energy‐saving strategy, they should limit energy expenditures by reducing foraging but not lying time, as the energy gains of forage intake may not meet the basal energetic requirements, especially in northern and temperate regions (the ‘energy‐saving’ hypothesis). Here, we contrast these two hypotheses by comparing individual daily time budgets of marked adult bighorn sheep rams (Ovis canadensis) and male mountain goats (Oreamnos americanus) during the pre‐rut and the rut. Concordant results for both species support the ‘foraging constraint’ hypothesis, as sexually‐active males reduced time spent foraging and lying from the pre‐rut to the rut because of an increase in time spent in mating‐related activities. Bighorn sheep rams also increased time spent foraging when not engaged in mating tactics, providing further support for a ‘maximisation’ of energy intake in the absence of reproductive opportunities. Because there are also known physiological changes that occur during the rut which may cause appetite suppression, for example to produce metabolic compounds linked with olfactory communication (the ‘scent‐urination’ hypothesis) or to cope with increased burden of parasites (the ‘parasite‐induced anorexia’ hypothesis), further research should aim at simultaneously testing these current hypotheses to better understand rut‐induced hypophagia and its effects on the life histories of male ungulates.  相似文献

Cistus creticus L., an aromatic species from the Mediterranean area, contains various diterpenes bearing the labdane skeleton. The production of essential oil from this species has potential economic value, but so far, it has not been optimized. In order to contribute to a better knowledge of this species and to its differentiation, the morphological characters, volatile chemical composition and genetic data of two subspecies (C. creticus subsp. eriocephalus and C. creticus subsp. corsicus) were investigated. The leaf trichomes were studied using scanning electron microscopy. The chemical composition of Corsican essential oil (C. creticus subsp. corsicus) has been reported using GC, GC/MS and 13C NMR; the main constituents were oxygenated labdane diterpenes (33.9%) such as 13-epi-manoyl oxide (18.5%). Using plant material (54 samples) collected from 18 geographically distinct areas of the islands of Corsica and Sardinia, the basis of variation in the headspace solid-phase microextraction volatile fraction and an inter-simple sequence repeat genetic analysis were also examined. It was shown that the two subspecies of C. creticus differed in morphology, essential oil production, volatile fraction composition and genetic data.  相似文献

Human embryonic stem cells (hESCs) hold great promise in regenerative medicine. However, before the full potential of these cells is achieved, major basic biological questions need to be addressed. In particular, there are still gaps in our knowledge of the molecular mechanisms underlying the derivation of hESCs from blastocysts, the regulation of the undifferentiated, pluripotent state, and the control of differentiation into specific lineages. Furthermore, we still do not fully understand the tumorigenic potential of hESCs, limiting their use in regenerative medicine. The RB pathway is a key signaling module that controls cellular proliferation, cell survival, chromatin structure, and cellular differentiation in mammalian cells. Members of the RB pathway are important regulators of hESC biology and manipulation of the activity of this pathway may provide novel means to control the fate of hESCs. Here we review what is known about the expression and function of members of the RB pathway in hESCs and discuss areas of interest in this field. J. Cell. Sexual ornaments might indicate better condition, fewer parasites or a greater immune responsiveness. Carotenoid-based ornaments are common sexual signals of birds and often influence mate choice. Skin or beaks pigmented by carotenoids can change colour rapidly, and could be particularly useful as honest indicators of an individual's current condition and/or health. This is because carotenoids must be acquired through diet and/or allocation for ornamental coloration might be to the detriment of self-maintenance needs. Here, we investigated whether the carotenoid-based coloration of eye rings and beak of male red-legged partridges Alectoris rufa predicted condition (mass corrected for size), parasite load (more specifically infection by coccidia, a main avian intestinal parasite) or a greater immune responsiveness (swelling response to a plant lectin, phytohaemagluttinin, or PHA). Redness of beak and eye rings positively correlated with plasma carotenoid levels. Also, males in better condition had fewer coccidia, more circulating carotenoids and a greater swelling response to PHA. Carotenoid-based ornamentation predicted coccidia abundance and immune responsiveness (redder males had fewer coccidia and greater swelling response to PHA), but was only weakly positively related to condition. Thus, the carotenoid pigmentation of beak and eye rings reflected the current health status of individuals. Our results are consistent with the hypothesis that allocation trade-offs (carotenoid use for ornamentation versus parasite defence needs) might ensure reliable carotenoid-based signalling.  相似文献

Although intestinal (I) and liver (L) fatty acid binding proteins (FABP) have been widely studied, the physiological significance of the presence of the two FABP forms (I- and L-FABP) in absorptive cells remains unknown as do the differences related to their distribution along the crypt-villus axis, regional expression, ontogeny and regulation in the human intestine. Our morphological experiments supported the expression of I- and L-FABP as early as 13 weeks of gestation. Whereas cytoplasmic immunofluorescence staining of L-FABP was barely detectable in the lower half of the villus and in the crypt epithelial cells, I-FABP was visualized in epithelial cells of the crypt-villus axis in all intestinal segments until the adult period in which the staining was maximized in the upper part of the villus. Immunoelectron microscopy revealed more intense labeling of L-FABP compared with I-FABP, accompanied with a heterogeneous distribution in the cytoplasm, microvilli and basolateral membranes. By western blot analysis, I- and L-FABP at 15 weeks of gestation appeared predominant in jejunum compared with duodenum, ileum, proximal and distal colon. Exploration of the maturation aspect documented a rise in L-FABP in adult tissues. Permanent transfections of Caco-2 cells with I-FABP cDNA resulted in decreased lipid export, apolipoprotein (apo) biogenesis and chylomicron secretion. Additionally, supplementation of Caco-2 with insulin, hydrocortisone and epidermal growth factor differentially modulated the expression of I- and L-FABP, apo B-48 and microsomal triglyceride transfer protein (MTP), emphasizing that these key proteins do not exhibit a parallel modulation. Overall, our findings indicate that the two FABPs display differences in localization, regulation and developmental pattern.  相似文献

Background, aim and scope  

Fly ash, a by-product of coal-fired power stations, is substituted for Portland cement to improve the properties of concrete and reduce the embodied greenhouse gas (GHG) emissions. Much of the world’s fly ash is currently disposed of as a waste product. While replacing some Portland cement with fly ash can reduce production costs and the embodied emissions of concrete, the relationship between fly ash content and embodied GHG emissions in concrete has not been quantified. The impact of fly ash content on embodied water is also unknown. Furthermore, it is not known whether a global trade in fly ash for use in concrete is feasible from a carbon balance perspective, or if transport over long distances would eliminate any CO2 savings. This paper aims to quantify GHG emissions and water embodied in concrete (fc = 32 MPa) as a function of fly ash content and to determine the critical fly ash transportation distance, beyond which use of fly ash in concrete increases embodied GHG emissions.  相似文献
