A thermoreversible fibrillar hydrogel has been formed from an aqueous lysozyme solution in the presence of dithiothreitol (DTT). Its physical properties and potential as a tissue engineering scaffold have been explored. Hydrogels were prepared by dissolving 3 mM protein in a 20 mM DTT/water mixture, heating to 85 degrees C and cooling at room temperature. No gel was observed for the equivalent sample without DTT. The elastic nature of the gel formed was confirmed by rheology, and the storage modulus of our gel was found to be of the same order of magnitude as for other cross-linked biopolymers. Micro differential scanning calorimetry (microDSC) experiments confirmed that the hydrogel was thermally reversible and that gelation and melting occurs through a solid-liquid-like first-order transition. Infrared spectroscopy of the hydrogel and transmission electron microscopy studies of very dilute samples revealed the presence of beta-sheet-rich fibrils that were approximately 4-6 nm in diameter and 1 mum in length. These fibrils are thought to self-assemble along their long axes to form larger fibers that become physically entangled to form the three-dimensional network observed in both cryo-scanning electron microscopy (cryo-SEM) and small-angle neutron scattering (SANS) studies. The hydrogel was subsequently cultured with 3T3 fibroblasts and cells spread extensively after 7 days and stretched actin filaments formed that were roughly parallel to each other, indicating the development of organized actin filaments in the form of stress fibers in cells.  相似文献

doi: 10.1111/j.1741‐2358.2011.00510.x Edentulism and dental caries in Victorian nursing homes Objectives: The aim of this project was to investigate edentulism and dental caries in nursing home residents in Victoria, Australia. Background: The Australian population is ageing with a growing number of people living in nursing homes. These residents are at increased risk for dental caries, have more teeth present now than at any time in the past 50 years and often have difficulty maintaining adequate oral hygiene. Materials and methods: Clinical dental examinations were conducted at 31 nursing homes in Melbourne and regional Victoria between May 2005 and June 2006. A total of 510 residents were examined out of 1345 eligible participants. Socio‐demographic and medical history was collected via questionnaire. Results: Just over half of the residents were dentate (53.9%), and dentate residents had a mean of 14.4 teeth present and 2.66 untreated decayed teeth. Residents who required total assistance with oral hygiene had more decayed teeth and fewer filled teeth than residents who did not require assistance. Conclusions: Nursing home residents in Victoria are retaining an increasing number of natural teeth and have more tooth surfaces at risk for dental caries. Untreated dental caries was a significant problem for residents, particularly for those who are dependent on others for their daily oral hygiene care.  相似文献

Entanglements of large whales in commercial fisheries in Newfoundland and Labrador, Canada, have been consistently recorded since 1979, as part of a program aimed at releasing captured animals and reducing costs to fishermen. This data set represented an opportunity to identify fisheries posing particular entanglement risks to local whale populations. Data were assessed over the periods 1979–1992 and 1993–2008, corresponding to distinct phases in fisheries distribution and intensity. Between 1979 and 2008, 1,209 large whale entanglements were recorded in Newfoundland and Labrador. These were mostly humpback whales (Megaptera novaeangliae; 80%) and minke whales (Balaenoptera acutorostrata; 15%). Dramatic declines in reported inshore whale entanglement rates were observed following the 1992 moratorium on Atlantic cod (Gadus morhua) fisheries. Recently, more entanglements have been reported further offshore, largely due to expansion of fisheries targeting snow crab (Chionoecetes opilio). For all whale species, entanglement rates and associated mortality rates varied considerably in different fishing gear. Fractions of humpback and minke whales found dead in different fishing gear differed substantially, with minke whales far more likely to be found dead than humpback whales.  相似文献

Biopsy techniques have been developed to collect skin and blubber samples through non‐lethal methods. One sample can provide data on genetics, prey preferences, foraging ecology, contaminant loads, and physiological processes. The limited data available suggest that biopsy wounds heal quickly and that there are usually no discernable adverse health effects. Published accounts on factors contributing to the success of collecting biopsy samples and the behavioral impacts to cetaceans following biopsy sampling were standardized to permit statistical analysis. Several factors contribute to the success of acquiring samples; however, sampling rates do not differ significantly between delivery devices. Behavioral responses to biopsy sampling vary by species and other factors. The most predominant response for odontocetes is low, while low and moderate responses are equally prevalent for mysticetes. The use of retrieval lines may increase the occurrence of moderate and strong responses by mysticetes. These findings suggest that biopsy sampling is relatively benign, causing only minor and short‐lived responses. However, most researchers do not report sufficient data to assess short‐ and long‐term physiological and behavioral impacts. Finally, limited data suggest that biopsy sampling does not impact cetacean habitat use or distribution patterns. Yet these impacts are rarely investigated, so additional data are needed.  相似文献

Compensatory mutations in RNA are generally regarded as those that maintain base pairing, and their identification forms the basis of phylogenetic predictions of RNA secondary structure. However, other types of compensatory mutations can provide higher-order structural and evolutionary information. Here, we present a helix-length compensation study for investigating structure-function relationships in RNA. The approach is demonstrated for stem-loop I and stem-loop V of the Neurospora VS ribozyme, which form a kissing-loop interaction important for substrate recognition. To rapidly characterize the substrate specificity (k(cat)/K(M)) of several substrate/ribozyme pairs, a procedure was established for simultaneous kinetic characterization of multiple substrates. Several active substrate/ribozyme pairs were identified, indicating the presence of limited substrate promiscuity for stem Ib variants and helix-length compensation between stems Ib and V. 3D models of the I/V interaction were generated that are compatible with the kinetic data. These models further illustrate the adaptability of the VS ribozyme architecture for substrate cleavage and provide global structural information on the I/V kissing-loop interaction. By exploring higher-order compensatory mutations in RNA our approach brings a deeper understanding of the adaptability of RNA structure, while opening new avenues for RNA research.  相似文献

Typical assays used to discover and analyze small molecules that inhibit the hepatitis C virus (HCV) NS3 helicase yield few hits and are often confounded by compound interference. Oligonucleotide binding assays are examined here as an alternative. After comparing fluorescence polarization (FP), homogeneous time-resolved fluorescence (HTRF®; Cisbio) and AlphaScreen® (Perkin Elmer) assays, an FP-based assay was chosen to screen Sigma’s Library of Pharmacologically Active Compounds (LOPAC) for compounds that inhibit NS3-DNA complex formation. Four LOPAC compounds inhibited the FP-based assay: aurintricarboxylic acid (ATA) (IC50 = 1.4 μM), suramin sodium salt (IC50 = 3.6 μM), NF 023 hydrate (IC50 = 6.2 μM) and tyrphostin AG 538 (IC50 = 3.6 μM). All but AG 538 inhibited helicase-catalyzed strand separation, and all but NF 023 inhibited replication of subgenomic HCV replicons. A counterscreen using Escherichia coli single-stranded DNA binding protein (SSB) revealed that none of the new HCV helicase inhibitors were specific for NS3h. However, when the SSB-based assay was used to analyze derivatives of another non-specific helicase inhibitor, the main component of the dye primuline, it revealed that some primuline derivatives (e.g. PubChem CID50930730) are up to 30-fold more specific for HCV NS3h than similarly potent HCV helicase inhibitors.  相似文献
