Soluble ADP-glucose: α-1,4-glucan-4-glucosyltransferase with primed activity was extracted from rice leaves and purified by (NH4)2SO4 fractionation, gradient elution on DEAE-cellulose and finally by Sephadex G200 gel filtration or amylopectin-cellulose chromatography. The purified enzyme was essentially homogeneous electrophoretically, but exhibited two peaks corresponding to MW of 22 000 and 67 000 on Sephadex G200 chromatography and five distinct bands on sodium dodecyl sulfate gel electrophoresis with MW of 11·5, 20, 35, 50 and 68 × 103. 相似文献

Stability of antisense DNA oligodeoxynucleotide analogs in cellular extracts and sera 总被引:14,自引:0,他引:14
Antisense DNA oligodeoxynucleotides can selectively inhibit the expression of individual (undesirable) genes and thus, have potential in the treatment of cancer and viral diseases. A prerequisite to their use as therapeutic agents is information on the stability of oligodeoxynucleotides, and their structurally modified analogs, in the biological milieu. To this end, degradation of 5' end and internally [32P] labelled unmodified DNA oligodeoxynucleotide (D-oligo) and analogs containing phosphorothioate (S-oligo), methylphosphonate (MP-oligo), and novel alternating methylphosphonate and phosphodiester (Alt-MP-oligo) internucleoside linkages was studied in Hela cell nuclear extract, S100 cytoplasmic extract, normal human serum and calf serum at 37 degrees C. Both 5' end and internally labelled D-oligos showed complete degradation within 30 min incubation in human serum at 37 degrees C. In any given medium, the D-oligo was the least stable oligodeoxynucleotide to nuclease degradation whereas the Alt-MP, MP and S-oligos were generally of comparable stability and all relatively more stable than D-oligo. Interestingly, MP and Alt-MP-oligos also exhibited greater resistance to phosphatases in cellular extracts compared to D and S-oligos. Under the conditions of the experiments, increasing degradation for any given oligonucleotide was observed in the order: S100 cytoplasmic extract less than nuclear extract less than normal human serum less than calf serum. In a study involving alpha-MEM cell culture medium containing 10% heat inactivated fetal calf serum (heated to 56 degrees C for 1 hour), the D-oligo was found to be rapidly degraded (degradation evident within 10 mins) whereas degradation products for the S-oligo were observed within 1 hour. In contrast, the Alt-MP oligo remained stable throughout the 3 hour experiment. These results indicated that in cell culture medium containing heat inactivated serum Alt-MP oligo was more stable than D- and S-oligos. 相似文献

Amphotericin B is an efficacious but extremely toxic anti fungal drug. Recently it has been shown that the incorporation of Amphotericin B in multilamellar liposomes results in a marked reduction in drug toxicity in mice with no loss of anti fungal potency. Until now, the mechanistic basis of the enhanced therapeutic index of liposomal Amphotericin B has been unclear. In this report, however, we show that the in vivo effects can be mimicked in vitro where free but not liposomal Amphotericin B causes lysis of erythrocytes while both free and liposomal drug kill fungal cells. These results suggest that the markedly improved therapeutic index of liposomal Amphotericin B is largely due to a fundamental alteration in the ability of the drug to interact with mammalian cell membranes rather than to alterations in pharmacokinetics or drug distribution. 相似文献

(Arg-Gly-Asp)n-albumin conjugates as a model substratum for integrin-mediated cell adhesion 总被引:2,自引:0,他引:2
We have prepared protein-peptide conjugates composed of bovine serum albumin (BSA) derivatized with short peptides containing the Arg-Gly-Asp (RGD) sequence derived from the adhesion site of fibronectin. The RGD-BSA conjugates were used to coat tissue culture plastic surfaces which then served as substrata in cell adhesion experiments. Our results indicate that the efficiency of adhesion to RGD-BSA-coated surfaces is highly dependent on the valency of the (RGD)n-BSA conjugates. For example, on surfaces with approximately equal amounts of RGD ligand, CHO cells adhered virtually 100% to the (RGD)n-BSA (n = 20.8) conjugate and not at all to the (RGD)n-BSA (n = 3.5) conjugate. Adhesion on (RGD)n-BSA-coated substrata and on fibronectin- or vitronectin-coated substrata was also examined in terms of the relationship between cell adhesion and the intermolecular distances of adsorbed proteins. It was observed that for substrata coated with relatively compact, symmetric molecules, such as RGD-BSA or vitronectin, adhesion dropped off sharply as intermolecular distances increased; by contrast, for fibronectin, a large asymmetric molecule, adhesion declined more gradually as intermolecular distances increased. Finally, we have examined the role of different cell-surface receptors in the process of adhesion to RGD-BSA substrata. Interestingly, competition and blocking experiments with antibodies and with soluble competing proteins suggest that it is the vitronectin receptor rather than the fibronectin receptor which mediates adhesion to RGD-BSA. 相似文献

resistance of resident communities to invasive species. We tested the hypothesis that coexistence or replacement of a resident
North American mosquito Aedes aegypti with the invasive Aedes albopictus is affected by interpopulation variation in the inherent competitive ability of A. aegypti and variation in the fecundity–size relationship. We postulated that such variation creates differential population-level
outcomes of competition with A. albopictus. We compared competitive abilities of eight North American populations of A. aegypti, four populations sympatric to A. albopictus, and four populations allopatric to A. albopictus. Competition among larvae from each A. aegypti population and a single A. albopictus population was tested in laboratory microcosms in a response-surface design. We found origin of A. aegypti influences its competitive response to competition from A. albopictus and competitive effect on A. albopictus. A. aegypti from allopatric sites preformed better in competition with A. albopictus than did A. aegypti from sympatric sites because they had a stronger average effect on A. albopictus. This average was strongly influenced by the allopatric population from Miami. Competitive effect and response were uncorrelated
among populations, indicating inconsistent ranking of A. aegypti in competitive effect and response. Although A. albopictus is generally a superior competitor to A. aegypti, a stronger competitive effect of particular A. aegypti populations on invading A. albopictus may contribute to competition-mediated biotic resistance to the invader. These results suggest that interpopulation variation
in competitive ability of A. aegypti may contribute to failure of A. albopictus to invade parts of the southeastern United States and offer evidence of a contribution to biotic resistance by an inferior
competitor. Geographic variation in competitive ability may be common and one general cause of variation in invasion success

We examined the substrate specificity of the carboxydipeptidase activity of neprilysin (NEP) using fluorescence resonance energy transfer (FRET) peptides containing ortho-aminobenzoyl (Abz) and 2,4-dinitrophenyl (Dnp) as a donor/acceptor pair. Two peptide series with general sequences Abz-RXFK(Dnp)-OH and Abz-XRFK(Dnp)-OH (X denotes the position of the altered amino acid) were synthesized to study P1 (cleavage at the X-F bond) and P2 (cleavage at R-F bond) specificity, respectively. In these peptides a Phe residue was fixed in P1' to fulfill the well-known NEP S1' site requirement for a hydrophobic amino acid. In addition, we explored NEP capability to hydrolyze bradykinin (RPPGFSPFR) and its fluorescent derivative Abz-RPPGFSPFRQ-EDDnp (EDDnp=2,4-dinitrophenyl ethylenediamine). The enzyme acts upon bradykinin mainly as a carboxydipeptidase, preferentially cleaving Pro-Phe over the Gly-Phe bond in a 9:1 ratio, whereas Abz-RPPGFSPFRQ-EDDnp was hydrolyzed at the same bonds but at an inverted proportion of 1:9. The results show very efficient interaction of the substrates' C-terminal free carboxyl group with site S2' of NEP, confirming the enzyme's preference to act as carboxydipeptidase at substrates with a free carboxyl-terminus. Using data gathered from our study, we developed sensitive and selective NEP substrates that permit continuous measurement of the enzyme activity, even in crude tissue extracts. 相似文献

Episodic resource inputs (i.e., pulses) can affect food web properties and community dynamics, but detailed mechanistic understanding of such effects remain elusive. Natural aquatic microsystems (e.g., tree holes, human-made containers) are colonized by invertebrates that form complex food webs dependent on episodic and sometimes sizeable inputs of allochthonous detritus from adjacent terrestrial environments. We investigated how variation in pulse frequency, amount, and resource type interacted to affect richness, abundance, composition, and population sizes of colonizing invertebrates in water-filled tires and tree hole analogs in a forest habitat. Different container types were used to assess the generality of effects across two environmental contexts. Containers received large infrequent or small frequent pulses of animal or leaf detritus of different cumulative amounts distributed over the same period. Invertebrates were sampled in June and September when cumulative detritus input was equal for the two pulse frequencies. Pulse frequency and detritus type interacted to affect the responses of richness and abundance in both months; pulse frequency alone in June affected the relationship between richness and abundance. Richness and abundance were also greater with more detritus regardless of detritus type. One group, the filter feeders, were most important in driving the response of abundance and richness to pulses, especially in June. This work highlights the potential complex nature of responses of communities and populations to resource pulses and implicates the ability of certain groups to exploit pulses of detrital resources as a key to understanding community-level responses to pulses. 相似文献
