Summary The interaction of pH (4 or 6), aluminium (0 or 16 ppm at pH 4) and N source (symbiotic or combined) on the growth and nutrient status of cowpea (Vigna unguiculata) was studied in a glasshouse experiment.Low pH significantly decreased the growth of the plants dependent on symbiotic nitrogen fixation but at pH 4 the addition of 16 ppm Al further depressed growth in both nitrogen regimes. Al-ions appear to exert their effect primarily on the root system, as shown by the reduction in total length and fresh weight. The symbiotic development of the plants was affected by low pH but more markedly by the Al treatment.Shoot nitrogen concentrations were reduced from ca. 2.6% at pH 6 to 1.8% and 0.9% at pH 4 without and with aluminium respectively. Calcium concentration was decreased by low pH and further by Al in both nitrogen regimes.In all Al-treated plants, the aluminium was mainly accumulated in the roots and was associated with an increase in their phosphorus concentration. 相似文献

Structure of inflorescences and flowers and flowering behaviour are reported for the woody liana Anchietea pyrifolia (Violaceae) from Brazil. The specimen studied is grown for some decades now in the greenhouses of Halle Botanical Garden and turned out unisexually male, which adds a further example of dioecism to the family Violaceae, in which this type of sex distribution is rarely encountered. The flowers are exceptional also for the strongly asymmetric anterior petal, which represents a rare case of a species with enantiomorphic flowers pollinated by Lepidoptera. They have a fully developed gynoecium with a complicated architecture comparable to the pistil of bisexual Violaceae flowers, though without ovules. The style head is capable to release viscose liquid on tactile stimulation or pressure, which is known to act as pollen-gathering mechanism in bisexual Violaceae species with usually dry pollen and buzz-pollination. This function has switched in male A. pyrifolia to a mechanism for efficient pollen release mediated by insect pollinators from its short-lived flowers. 相似文献

In 1996, Gottlieb et al. [Gottlieb GL, Song Q, Hong D, Almeida GL, Corcos DM. Coordinating movement at two joints: A principle of linear covariance. J Neurophysiol 1996;75(4):1760–4] identified a linear co-variance between the joint muscle torques generated at two connected joints. The joint muscle torques changed directions and magnitudes in a synchronized and linear fashion and called it the principle of linear co-variance. Here we showed that this principle cannot hold for some class of movements. Neurologically normal subjects performed multijoint movements involving elbow and shoulder with reversal towards three targets in the sagittal plane without any constraints. The movement kinematics was calculated using the X and Y coordinates of the markers positioned over the joints. Inverse dynamics was used to calculate the joint muscle, interaction and net torques. We found that for the class of voluntary movements analyzed, the joint muscle torques of the elbow and the shoulder were not linearly correlated. The same was observed for the interaction torques. But, the net torques at both joints, i.e., the sum of the interaction and the joint muscle torques were linearly correlated. We showed that by decoupling the joint muscle torques, but keeping the net torques linearly correlated, the CNS was able to generate fast and accurate movements with straight fingertip paths. We studied the expression level and cell-specific expression patterns of 5alpha-reductase (5alpha-R) types 1 and 2 iso-enzymes in human hyperplastic and malignant prostate tissue by semi-quantitative RT-PCR and in situ hybridisation analyses. In situ hybridisation established that 5alpha-R1 mRNA is preferentially expressed by epithelial cells and little expressed by stromal cells whereas 5alpha-R2 mRNA is expressed by both epithelium and stroma. Semi-quantitative RT-PCR has been performed on total RNA from different zones of normal prostate, BPH tissues and liver. We found that 5alpha-R1 and 5alpha-R2 mRNAs expression was near the same in all zones of normal prostate. In BPH tissue, 5alpha-R1 and 5alpha-R2 mRNAs expression was slightly but significantly increased, when it was compared to the levels recorded for normal prostate. In cancer samples, 5alpha-R1 mRNA expression was higher than in normal and hyperplastic prostate but the level of 5alpha-R2 mRNA was not statistically different from that observed in the different zones of normal prostate. In liver, 5alpha-R2 mRNA level was similar to that measured in BPH but 5alpha-R1 mRNA expression was ten times higher. The increase observed in 5alpha-R isoenzymes expression in BPH tissue could play an important role in the pathogenesis and/or maintenance of the disease. 相似文献

βPIX (p21-activated kinase interacting exchange factor) and Shank/ProSAP protein form a complex acting as a protein scaffold that integrates signaling pathways and regulates postsynaptic structure. Complex formation is mediated by the C-terminal PDZ binding motif of βPIX and the Shank PDZ domain. The coiled-coil (CC) domain upstream of the PDZ binding motif allows multimerization of βPIX, which is important for its physiological functions. We have solved the crystal structure of the βPIX CC-Shank PDZ complex and determined the stoichiometry of complex formation. The βPIX CC forms a 76-Å-long parallel CC trimer. Despite the fact that the βPIX CC exposes three PDZ binding motifs in the C-termini, the βPIX trimer associates with a single Shank PDZ. One of the C-terminal ends of the CC forms an extensive β-sheet interaction with the Shank PDZ, while the other two ends are not involved in ligand binding and form random coils. The two C-terminal ends of βPIX have significantly lower affinity than the first PDZ binding motif due to the steric hindrance in the C-terminal tails, which results in binding of a single PDZ domain to the βPIX trimer. The structure shows canonical class I PDZ binding with a β-sheet interaction extending to position − 6 of βPIX. The βB-βC loop of Shank PDZ undergoes a conformational change upon ligand binding to form the β-sheet interaction and to accommodate the bulky side chain of Trp − 5. This structural study provides a clear picture of the molecular recognition of the PDZ ligand and the asymmetric association of βPIX CC and Shank PDZ. 相似文献

Individual specialization can influence important ecological and evolutionary traits and both inter‐ and intra‐individual variation in resource use can drive niche shifts in natural populations. We evaluated the predominance of these two factors for determining seasonal differences in the trophic niche of the didelphid marsupial Gracilinanus agilis (Burmeister, 1854) in the highly seasonal Brazilian savanna. In the three sampled sites, the population of G. agilis increased its dietary niche width in the warm–wet season, when food resources are more abundant, and there were no differences between sexes and no interaction between season and sex. However, the evaluation of intra‐individual variation indicated that females reduce the number of items consumed during the warm–wet season, whereas males show no seasonal differences. Inter‐individual variation nonetheless followed the overall population pattern because both sexes increased their spread with respect to food‐item consumption in the warm–wet season. Additionally, we found positive relationships between body length and diet only in the warm–wet season, when larger animals fed more on invertebrates and less on fruits than the small ones. The cytoplasmic fates of mRNAs are influenced by interactions between RNA-binding proteins and cis regulatory motifs. In the cytoplasm, mRNAs are present as messenger ribonucleoprotein particles, which include not only proteins that bind directly to the mRNA, but also additional proteins that are recruited via protein-protein interactions. Many labs have sought to purify such particles from cells, with limited success. We here describe a simple two-step procedure to purify actively translated mRNAs, with their associated proteins, from polysomes. We use a reporter mRNA that encodes a protein with three streptavidin binding peptides at the N-terminus. The polysomal reporter mRNA, with associated proteins, is purified via binding to a streptavidin matrix. The method takes four days, and can be applied in any cell that can be genetically manipulated. Using Trypanosoma brucei as a model system, we routinely purified 8% of the input reporter mRNA, with roughly 22-fold enrichment relative to un-tagged mRNAs, a final reporter-mRNA:total-mRNA ratio of about 1:10, and a protein purification factor of slightly over 1000-fold. Although the overall reporter mRNP composition is masked by the presence of proteins that are associated with many polysomal mRNAs, our method can be used to detect association of an RNA-binding protein that binds to specifically to a reporter mRNA. 相似文献

The primary structure of apolipoprotein J (apoJ) was deduced by the combined strategies of protein sequencing and cDNA cloning and sequencing. ApoJ, an apolipoprotein associated with discrete subclasses of high-density lipoproteins, is encoded by a single gene in both the human and mouse genomes. ApoJ is synthesized as a 427 amino acid polypeptide that is posttranslationally cleaved at an internal bond between Arg-205 and Ser-206. The subunits of apoJ are designated apoJ alpha, corresponding to residues 1-205, and apoJ beta, corresponding to resides 206-427. The subunits are associated through disulfide bonds. Analysis of the primary structure of apoJ predicts the existence of amphiphilic helices, which may account for the association of apoJ with lipoproteins, and heparin-binding motifs in both subunits. ApoJ appears to be the human analogue of a rat protein present in high concentrations in the testis, sulfated glycoprotein 2. ApoJ mRNA (1.9 kb) is expressed in all but one tissue examined. The mRNA is present in relatively high levels in brain, ovary, testis, and liver, is less abundant in heart, spleen, lung, and breast, and is absent in T-lymphocytes.
