Mesenchymal stem cells (MSCs) are multipotent cells that are able to differentiate into mesodermal lineages (osteogenic, adipogenic, chondrogenic), but also towards non-mesodermal derivatives (e.g. neural cells). Recent in vitro studies revealed that, in the absence of any kind of differentiation stimuli, undifferentiated MSCs express neural differentiation markers, but the literature data do not all concur. Considering their promising therapeutic potential for neurodegenerative diseases, it is very important to expand our knowledge about this particular biological property of MSCs. In this study, we confirmed the spontaneous expression of neural markers (neuronal, glial and progenitor markers) by undifferentiated human MSCs (hMSCs) and in particular, we demonstrated that the neuronal markers βIII-tubulin and NeuN are expressed by a very high percentage of hMSCs, regardless of the number of culture passages and the culture conditions. Moreover, the neuronal markers βIII-tubulin and NeuN are still expressed by hMSCs after in vitro osteogenic and adipogenic differentiation. On the other hand, chondrogenically differentiated hMSCs are negative for these markers. Our findings suggest that the expression of neuronal markers could be common to a wide range of cellular types and not exclusive for neuronal lineages. Therefore, the expression of neuronal markers alone is not sufficient to demonstrate the differentiation of MSCs towards the neuronal phenotype. Functional properties analysis is also required. 相似文献
Among 1,236 colony-forming units (CFU) associated with 11 species of marine sponges collected from a Brazilian coast, a total of 100 morphologically different bacterial strains were analyzed. The phylogenetic diversity of the bacterial isolates was assessed by 16S rRNA gene amplification—restriction fragment length polymorphism (RFLP) analysis, using AluI restriction endonuclease. The RFLP fingerprinting resulted in 21 different patterns with good resolution for the identification of the bacterial isolates at the genus level. The genus Bacillus was the most commonly encountered genus, followed by Kocuria. Regarding the relationship between the morphotypes and species of marine sponges, Mycale microsigmatosa presented major diversity, followed by Dragmacidon reticulatum and Polymastia janeirensis. An antibiotic susceptibility profile of the 100 sponge-associated bacterial strains was determined by the disk diffusion method, and we observed a variable resistance profile, with 15 % of the bacteria being multiresistant. In addition, 71 of 100 strains were able to produce biofilm. These 71 strains were divided into 20 strong biofilm producers, 10 moderate biofilm producers, and 41 weak biofilm producers. The plasmid profile of the 100 bacterial strains was analyzed and 38 (38 %) of these samples possessed one or more plasmids. Studies like this are important to increase the information on these associated bacteria found off the coastline of Brazil, a place which has rich biodiversity that is still unknown. 相似文献
The aim of this study was standardization and application of polymerase chain reaction (PCR) for the detection of contaminants in cell cultures, sera and trypsin. Five PCR protocols were standardized to assess the presence of genetic material from mycoplasma, porcine circovirus 1 (PCV1), bovine leukemia virus (BLV) or bovine viral diarrhea virus (BVDV) in cell culture samples. PCR reactions for the genes GAPDH and beta-actin were used to evaluate the efficiency of nucleic acid extraction. The PCR protocols were applied to 88 cell culture samples from eight laboratories. The tests were also used to assess potential contamination in 10 trypsin samples and 13 fetal calf serum samples from different lots from five of the laboratories. The results showed the occurrence of the following as DNA cell culture contaminants: 34.1% for mycoplasma, 35.2% for PCV1, 23.9% for BVDV RNA and 2.3% for BLV. In fetal calf sera and trypsin samples BVDV RNA and PCV1 DNA was detected. The results demonstrated that cell culture, sera and trypsin used by different laboratories show a high rate of contaminants. The results highlight the need for monitoring cell cultures and controlling for biological contaminants in laboratories and cell banks working with these materials. 相似文献
Legionnaires' disease is an emerging, severe, pneumonia‐like illness caused by the Gram‐negative intracellular bacteria Legionella pneumophila, which are able to infect and replicate intracellularly in macrophages. Little is known regarding the mechanisms used by intracellular L. pneumophila for the acquisition of specific nutrients that are essential for bacterial replication. Here, we investigate three L. pneumophila genes with high similarity to the Escherichia coli K+ transporters. These three genes were expressed by L. pneumophila and have been designated kupA, kupB and kupC. Investigation using the L. pneumophilakup mutants revealed that kupA is involved in K+ acquisition during axenic growth. The kupA mutants replicated efficiently in rich axenic media, but poorly in a chemically defined medium. The kupA mutants were defective in the recruitment of polyubiquitinated proteins to the Legionella‐containing vacuole that is formed in macrophages and displayed an intracellular multiplication defect during the replication in Acanthamoeba castellanii and in mouse macrophages. We found that bafilomycin treatment of macrophages was able to rescue the growth defects of kupA mutants, but itdid not influence the replication of wild‐type bacteria. The defects identified in kupA mutants of L. pneumophila were complemented by the expression E. colitrkD/Kup gene in trans, a bona fide K+ transporter encoded by E. coli. Collectively, our data indicate that KupA is a functional K+ transporter expressed by L. pneumophila that facilitates the bacterial replication intracellularly and in nutrient‐limited conditions. 相似文献
Since the 1980s, cetacean morbillivirus (CeMV) has caused mass mortality events worldwide. However, no epizootics had been recorded in the South Atlantic, until an unusual mortality event (UME) linked to Guiana dolphin cetacean morbillivirus (GD-CeMV) began in Ilha Grande Bay, southeastern Brazil, in November 2017. In a five-month period, the UME spread to neighboring Sepetiba Bay and accounted for the death of at least 277 Guiana dolphins (Sotalia guianensis). Prevalence of morbillivirus positive dolphins, as estimated from RT-PCR diagnostics, was 92.3% (24/26) in Ilha Grande Bay and 91.9% (57/62) in Sepetiba Bay. Females had higher mortality rates during the UME (1.5:1), in contrast with historical mortality data from both bays that showed a 2:1 male to female death ratio. Calf mortality rates also increased in both bays. These results suggest that females and calves were more vulnerable to morbilliviral infection. Herein, we discuss possible explanations for such sex-biased death pattern during the UME and their implication for the conservation of endangered Guiana dolphins. Among the skin disorders of dogs, scabies is notable for its high occurrence rate and the need for veterinary interventions. There are two obstacles to making this diagnosis through direct investigation under a microscope (DIM): the continual need to make new slides when the results are negative and the long time needed for reading these slides. Thus, the objective of the present study was to compare efficacy between DIM and the technique of centrifugation-flotation in sucrose solution (CFSS) in samples from dogs in the semiarid region of the state of Paraíba, Brazil. Samples from 136 dogs were used, and three slides were made for each examination (DIM and CFSS). The readings were halted in cases of positivity. Positive samples were obtained from 56.6% of the dogs (77/136), of which 76.6% (59/77) were positive through both techniques, 13% (10/77) only through DIM and 10.4% (8/77) only through CFSS. The positivity rate did not differ statistically between the techniques. CFSS showed higher quality of readings, due to the considerably fewer artifacts on the slides, thereby optimizing the reading time. Sensitivity (85.6%), specificity (88.1%), accuracy (86.8%), positive predictive value (88.1%) and negative predictive value (85.1%) were obtained and the kappa coefficient (0.73) was considered substantial. It was concluded that CFSS showed high diagnostic capacity for scabies, similar to that of DIM, with optimized reading time, fewer artifacts and better display of mites.
