A cell has been isolated from explanted rabbit liver which contains, during all phases of its growth in culture, hundreds of lipid-rich particles with a distinct limiting membrane. The cell grows logarithmically with a generation time of 19 to 20 hours and during mitosis the particles are distributed between the daughter cells. Associated with the particles is the high total lipid content of the rabbit liver cell as compared with a rat liver cell, which contains few, if any, lipid-rich particles. This difference in lipid content between the two cells is due primarily to an increase in the triglyceride fraction, in contradistinction to small differences in the polar lipid and sterol ester fractions. The lipid-rich particles have been isolated and found to contain 90 per cent triglyceride on a dry weight basis. The "genetic" factors responsible for the high concentration of lipid-rich particles and triglycerides in the rabbit liver cell require for their full expression one or more factors which are present in much higher effective concentrations in rabbit serum than in horse serum. The hypothesis is advanced that the lipid-rich particles represent a normal state of the non-structural cell lipid. A procedure is described for the quantitative isolation of the lipid of cultured cells.  相似文献

Twenty equine microsatellites were isolated from a genomic phage library, and their genetical and physical localization was sought by linkage mapping and fluorescent in situ hybridization (FISH). Nineteen of the markers were found to be polymorphic with, in most cases, heterozygosities exceeding 50%. The markers were mapped in a Swedish reference family for gene mapping, comprising eight half-sib families from Standardbred and Icelandic horse sires. Segregation was analyzed against a set of 35 other markers typed in the pedigree. Thirteen of the microsatellites showed linkage to at least one other marker, with a total of 21 markers being involved in these linkages. In parallel, 18 of the microsatellites could be assigned to their chromosomal region by FISH. These assignments involved eight equine autosomes: ECA1, 2, 4, 6, 9, 10, 15, and 16. The genetical and physical mappings revealed by this study represent a significant extension of the current knowledge of the equine genome map. The effects of a short-day treatment on pool-size, synthesis, degradation, and transport of 3-indole-acetic acid (IAA) in Scots pine ( Pinus sylvestris L.) seedlings were studied. The short-day treatment decreased the pool of free IAA in the needles but did not significantly affect the pool-sizes of free IAA in the shoot-axis, hypocotyls, cotyledons and roots. The bound alkali-hydrolysable IAA-pools were not affected by the short-day treatment. The synthesis and degradation of radioactive IAA were also unaffected by the short-day treatment. The transport of radioactive IAA was greatest from needles to roots, and this transport was significantly decreased by the short-day treatment.  相似文献

Nuñas (Thaseolus vulgaris, Fabaceae), commonly called popping beans, are traditionally grown in the Andean highlands of South America, and are consumed as a snack food after a quick toasting process. Proximate analysis of their nutritive value revealed that nunas have a higher content of starch, amylose, and copper than four dry bean varieties and a lower mean content of protein, phosphorous, iron, and boron. The unique texture and taste of nuñas may be related to their high starch content. Antinutritional factors such as lectins were higher in raw and boiled nuña samples than in toasted nuñas, while tannin levels did not change from raw to toasted treatments. Overall in-vitro digestibility was slightly lower for toasted nunas than boiled dry bean.  相似文献

Studies of sleep influences on human pharyngeal and other respiratory muscles suggest that the activity of these muscles may be affected by non-rapid-eye-movement (NREM) sleep in a nonuniform manner. This variable sleep response may relate to the pattern of activation of the muscle (inspiratory phasic vs. tonic) and peripheral events occurring in the airway. Furthermore, the ability of these muscles to respond to respiratory stimuli during NREM sleep may also differ. To systematically investigate the effect of NREM sleep on respiratory muscle activity, we studied two tonic muscles [tensor palatini (TP), masseter (M)] and two inspiratory phasic ones [genioglossus (GG), diaphragm (D)], also measuring the response of these muscles to inspiratory resistive loading (12 cmH2O.l-1.s) during wakefulness and NREM sleep. Seven normal male subjects were studied on a single night with intramuscular electrodes placed in the TP and GG and surface electrodes placed over the D and M. Sleep stage, inspiratory airflow, and moving time average electromyograph (EMG) of the above four muscles were continuously recorded. The EMG of both tonic muscles fell significantly (P less than 0.05) during NREM sleep [TP awake, 4.3 +/- 0.05 (SE) arbitrary units, stage 2, 1.1 +/- 0.2; stage 3/4, 1.0 +/- 0.2. Masseter awake, 4.8 +/- 0.6; stage 2, 3.3 +/- 0.5; stage 3/4, 3.1 +/- 0.5]. Summary DBA/2 mice inoculated with either cells from the syngeneic P815 tumor or tumor cell membrane extracts develop T suppressor cells which suppress the in vitro generation of cytotoxic T lymphocytes with specificity for the tumor. A soluble suppressor factor with similar properties can be isolated from suppressor cell-enriched populations. It can be highly purified by appropriate immunoadsorption. Antisera to this suppressor factor raised in either DBA/2 or C57BL/6 mice can specifically absorb out suppressor factor and eliminate suppressor cells in the presence of complement. The in vivo effects of these antisera were tested for their ability to modulate the growth of P815 tumors in DBA/2 mice. It was found that the antiserum raised in syngeneic (DBA/2) but not allogeneic (C57BL/6) mice was able to significantly slow the rate of tumor growth and to prolong survival in treated mice. The antiserum was effective in this way only if it was administered early in the course of tumor growth. It was shown that this effect was not attributable to the presence in the serum of antibodies directed to antigens present on P815 cells, and it therefore appears to be due to interference with the function of T suppressor cells arising early in the immune response to the tumor cells.  相似文献

A cell-associated lectin activity that mediates lactose-inhibitable adherence of Actinomyces viscosus T14V has been localized to a specific population of fimbriae by the use of monoclonal antibodies. Nine monoclonal antibodies were produced that reacted with only 1 of 2 immunoelectrophoretically distinct fimbrial components on T14V. The fibrillar morphology of this component was revealed by the immunoelectronmicroscopic examination of bacteria incubated with the monoclonal antibodies. The lectin activity associated with these structures was detected when isolated fimbriae were cross-linked with monoclonal antibodies to form immune complexes with agglutination activity for neuraminidase-treated human erythrocytes, a reaction that was inhibited by lactose. Although the 9 monoclonal antibodies differed in their fine specificities, they reacted only with strains of A. viscosus and A. naeslundii that exhibited lactose-inhibitable adherence. These findings indicate that the lectin activity common to these bacteria resides on fimbriae that are antigenically related to those of T14V.  相似文献
