Steady state sensitivity analysis of a model of carbohydrate metabolism and anaplerotic synthesis of oxalacetate were, in Aspergillus niger under conditions of citric acid accumulation, carried out. The flux and metabolite concentration control structure of the system obtained shows that the hexokinase/substrate transport step is the main controlling step of the pathway. The quantitative contribution of the other enzyme catalyzed or transport steps are also discussed. Lantana, or red sage (lantana camara, Verbenaceae) is an aromatic shrub with rough, opposite or whorled leaves and four-angled, prickly or smooth stems, showy heads of small flowers (usually pink-and-yellow changing to red-and-orange) and dark-blue or blackish fruits. Native from tropical America north to lower Texas and southern Georgia, the bird-distributed plant has become naturalized in the Caribbean area, the Pacific islands, Australia, New Zealand, South Africa and southern Asia. Hundreds of cultivars are grown as ornamentals in temperate climates. In warm regions, L. camara is a pest in farms, pastures and forests, providing breeding places for diseases, detrimental animals and insects, including the tsetse fly. Of 29 named races in Australia, 19 are toxic or gravely pestiferous. Active principles are the triterpenes lantadene A and B. Ingestion of 340-453 g of leaves causes liver and kidney damage, photosensitization, intestinal hemorrhage, paralysis of the gall bladder, and death in 1-4 days in horses, cattle and sheep (not goats). Ripe fruits are edible, but green, unripe fruits have caused illness and one known fatality in children in Florida. Leaves and roots are popular folk remedies. Stems are used as toothbrushes, and the leaves for polishing wood. Prickly races are grown as protective hedges around forest dwellings.  相似文献

TheRhizobium tropici strain CFN 299 was maintained on PY medium and was grown in minimal medium (MM) with sucrose, glucose, fructose and glutamate (or their combination) as carbon sources. Bacteria were able to simultaneously use different carbon sources and, with a combination sucrose and glutamate, the growth rate was faster than with either carbon source alone. Sucrose transport was induced by sucrose and partially repressed by glucose and glutamate if they were included in MM as additional carbon sources. The transport of sucrose was active because both an uncoupler (dinitrophenol, DNP) and inhibitors of terminal oxidation (KCN, NaN3) severely reduced sucrose uptake. Sucrose transport was also sensitive to a functional sulfhydryl reagent but was much less sensitive to EDTA and arsenate. We obtained nonlinear Lineweaver-Burk plots for the uptake of sucrose (by sucrose-grown bacteria), and this implied the existence of at least two uptake mechanisms. Invertase (EC 3.2.1.26) is the main enzyme for sucrose hydrolysis in this organism. This enzyme was induced by sucrose and had high activity in mid-log phase cells when sucrose was the sole carbon source (0.2%). Invertase activity was not detected in growth medium. In general, the results obtained support the idea, thatR. tropici is adapted to sucrose utilization and to multicarbon nutrition during its interaction with plants.  相似文献

Water level, ash content, proximate (protein, lipid, carbohydrate and chitin) and elemental (carbon and nitrogen) composition were analyzed in twentythree species of Antarctic Zooplankton collected during the austral fall (1986) and winter (1988) from the Scotia/Weddell Sea region. Extremes in water level, ash content and organic components were typified by copepods and gelatinous forms. Ostracods and polychaetes were generally similar in composition to copepods, being only slightly higher in water level and ash content. Chaetognaths exhibited a composition intermediate in character with some components similar in value to that shown by crustaceans (i.e. protein) while other components were more in the range of values seen in gelatinous forms (i.e. water level and ash content). Protein was the major proximate component and measured values (as % Afdw) were fairly uniform among non-gelatinous species (x=33.9±6.9). Lipid levels were variable, with high values (>30% AFDW) only found for the copepods Calanoides acutus, Calanus propinquus and Euchaeta antarctica. Carbohydrate values were low in all species examined. Chitin was measured in crustacean species only. With the exception of C. acutus (x=2.5% AFDW chitin), values were similar among species with mean values being slightly higher in fall (x=11.8±2.5) than in winter (x=6.7±1.8). Among non-gelatinous species, the ratio of carbon to nitrogen was positively correlated with the lipid to protein ratio, underscoring the compositional association between elemental and proximate components in these groups. In gelatinous species, the relationship between carbon:nitrogen and lipid:protein was inconsistent and less pronounced. Caloric content was estimated from recovered organic matter for nongelatinous species. As a function of wet weight and dry weight, values reflected differences in water level and ash content among individual species. As a function of ashfree dry weight, values were similar among all species (x=3.6±0.9 kcal/g).Seasonal comparisons were possible for 12 of the 23 species. Among crustaceans, changes in water level and organic components were variable reflecting dissimilar trophic, reproductive or ecological habits among different species. Essentially no change in composition between fall and winter was observed for diapause species (e.g. Calanoides acutus and Rhincalanus gigas) as well as for omnivorous/ carnivorous species (e.g. Gaetanus tenuispinus). Conversely, large compositional changes were evident for Calanus propinquus, a small-particle grazer that relies heavily on lipid reserves. Chaetognaths and some gelatinous species exhibited a considerable decrease in ash content from fall to winter which, for most cases, was mirrored by some degree of increase in lipid level. At present, however, scant data are available to help explain the observed patterns of compositional change within non-crustacean species.  相似文献

The HLA class II sequences included in this compilation are taken from publications listed in the papers: Nomenclature for factors of the HLA system, 1989, Nomenclature for factors of the HLA system, 1990, and Nomenclature for factors of the HLA system, 1991 (WHO Nomenclature Committee 1990, 1991, 1992). Where discrepancies have arisen between reported sequences, the original authors have been contacted where possible, and necessary amendments to published sequences have been incorporated into this alignment. Future sequencing may identify errors in this list, and we would welcome any evidence that helps to maintain the accuracy of this compilation. In the sequence alignments, identity between residues is indicated by a hyphen (-), an unavailable sequence is indicated by an asterisk (*), and gaps in the sequence are inserted to maintain the alignment between different alleles showing variation in amino acid number. The gene that encodes the acyl carrier protein (ACP) of the actinorhodin polyketide synthase (PKS) of Streptomyces coelicolor A3(2) was replaced with homologs from the granaticin, oxytetracycline, tetracenomycin, and putative frenolicin polyketide synthase gene clusters. All of the replacements led to expression of functional synthases, and the recombinants synthesized aromatic polyketides similar in chromatographic properties to actinorhodin or to shunt products produced by mutants defective in the actinorhodin pathway. Some regions within the ACP were also shown to be interchangeable and allow production of a functional hybrid ACP. Structural analysis of the most abundant polyketide product of one of the recombinants by electrospray mass spectrometry suggested that it is identical to mutactin, a previously characterized shunt product of an actVII mutant (deficient in cyclase and dehydrase activities). Quantitative differences in the product profiles of strains that express the various hybrid synthases were observed. These can be explained, at least in part, by differences in ribosome-binding sites upstream of each ACP gene, implying either that the ACP concentration in some strains is rate limiting to overall PKS activity or that the level of ACP expression also influences the expression of another enzyme(s) encoded by a downstream gene(s) in the same operon as the actinorhodin ACP gene. These results reaffirm the idea that construction of hybrid polyketide synthases will be a useful approach for dissecting the molecular basis of the specificity of PKS-catalyzed reactions. However, they also point to the need for reducing the chemical complexity of the approach by minimizing the diversity of polyketide products synthesized in strains that produce recombinant polyketide synthases.  相似文献

Addition of polyamines or their analogs to newly confluent LLC-PK1 cells resulted in down-regulation of Na+-glucose transport (symport) activity. Polyamines prevented the induction of this symporter by the differentiation inducer hexamethylene bisacetamide (HMBA) but did not influence induction by the phosphodiesterase inhibitor 3-isobutyl-1-methylxanthine (IBMX). Partial depletion of endogenous polyamines after addition of α -difluoromethylornithine (DFMO) resulted in a 4 to 5-fold increase in symporter expression. Symporter induction by either HMBA or DFMO was inhibited by the protein kinase inhibitor H-7 but H-7 did not affect symporter induction by IBMX. Changes in symporter activity were accompanied by changes in levels of the 75 kD symporter subunit detected by Western blot. Cultures exposed to HMBA exhibited reduced levels of ornithine decarboxylase activity. Our results suggest that induction of symporter expression by HMBA may be mediated in part by its effects on polymine metabolism, and point to parallel roles of polyamines and cyclic AMP in regulating the expression of this physiologically important renal transport system. © 1993 Wiley-Liss, Inc.  相似文献

Transfer of resistance toHeterodera avenae, the cereal cyst nematode (CCN), by a stepping-stone procedure from the wild grassAegilops ventricosa to hexaploid wheat has been demonstrated. The number of nematodes per plant was lower, and reached a plateau much earlier, in the resistant introgression line H93-8 (1–2 nematodes per plant) than in the recipient H10-15 wheat (14–16 nematodes per plant). Necrosis (hypersensitive reaction) near the nematode, little cell fusion, and few, often degraded syncytia were observed in infested H93-8 roots, while abundant, well-formed syncytia were present in the susceptible H10-15 wheat. Line H93-8 was highly resistant to the two Spanish populations tested, as well as the four French races (Fr1-Fr4), and the British pathotype Hall, but was susceptible to the Swedish pathotypes HgI and HgIII. Resistance was inherited as though determined by a single quasi-dominant factor in the F2 generations resulting from crosses of H93-8 with H10-15 and with Loros, a resistant wheat carrying the geneCre1 (syn.Ccn1). The resistance gene in H93-8 (Cre2 orCcn2) is not allelic with respect to that in Loros. RFLPs and other markers, together with the cytogenetical evidence, indicate that theCre2 gene has been integrated into a wheat chromosome without affecting its meiotic pairing ability. Introduction ofCre2 by backcrossing into a commercial wheat backgroud increases grain yield when under challenge by the nematode and is not detrimental in the absence of infestation.  相似文献

Long-term atmospheric CO2 concentration records have suggested a reduction in the positive effect of warming on high-latitude carbon uptake since the 1990s. A variety of mechanisms have been proposed to explain the reduced net carbon sink of northern ecosystems with increased air temperature, including water stress on vegetation and increased respiration over recent decades. However, the lack of consistent long-term carbon flux and in situ soil moisture data has severely limited our ability to identify the mechanisms responsible for the recent reduced carbon sink strength. In this study, we used a record of nearly 100 site-years of eddy covariance data from 11 continuous permafrost tundra sites distributed across the circumpolar Arctic to test the temperature (expressed as growing degree days, GDD) responses of gross primary production (GPP), net ecosystem exchange (NEE), and ecosystem respiration (ER) at different periods of the summer (early, peak, and late summer) including dominant tundra vegetation classes (graminoids and mosses, and shrubs). We further tested GPP, NEE, and ER relationships with soil moisture and vapor pressure deficit to identify potential moisture limitations on plant productivity and net carbon exchange. Our results show a decrease in GPP with rising GDD during the peak summer (July) for both vegetation classes, and a significant relationship between the peak summer GPP and soil moisture after statistically controlling for GDD in a partial correlation analysis. These results suggest that tundra ecosystems might not benefit from increased temperature as much as suggested by several terrestrial biosphere models, if decreased soil moisture limits the peak summer plant productivity, reducing the ability of these ecosystems to sequester carbon during the summer.  相似文献
