Carbonic anhydrase isoenzyme VI (CA VI), which is transported in high concentrations in saliva and milk into the alimentary tract, is an important element of mucosal protection in the upper alimentary tract. Like alimentary tract mucosa, the respiratory tract mucosa is also exposed to heavy microbial, physical, and chemical stress. The protective and renewal-promoting factors present in the surface mucus of the respiratory tract are mainly produced by the seromucous tracheobronchial glands. Here we studied the secretion of CA VI by these glands in adult and developing rats using immunohistochemical techniques. The serous acinar and duct cells of the tracheobronchial glands stained for CA VI. The presence of the enzyme also in the duct content indicates its active secretion into the surface mucus. CA VI was also visible in the secretory cells and at the base of the ciliated cells of the tracheobronchial surface epithelium. Moreover, the Clara cells of the bronchiolar surface epithelium stained for CA VI. These findings are consistent with the hypothesis that CA VI has a mucosa-protective role not only in the gastrointestinal tract but also in the respiratory tract, where CA VI may act as a pivotal pH neutralizer and growth factor.  相似文献

Wetland buffers may play an important role in the retention of nitrogen (N) and phosphorus (P) that can be released in large quantities from forestry operations. In this study, we investigated the retention capacity of N and P of wetland vegetation comparing the control area with two experimental areas within one site before and after N and P pulse (45 kg N and 15 kg P) lasting one growing season (ca. 150 d). N and P pulse caused a significant increase in the plant biomass and N and P content in the upper experimental area, which received most of the added nutrients. Added N and P was mainly retained in the above and below ground parts of E. vaginatum, especially in storage organs and roots which form a long-term sink for nutrients. Total N retention in the plant biomass during the first year after N and P treatment ranged from 25.3 kg (equals to 126.7 kg N ha–1) in the upper experimental area to 6.1 kg (20.4 kg N ha–1) in the lower experimental area and 4.7 kg (15.7 kg N ha–1) in the control area. P retention ranged from 2.6 kg (13.1 kg P ha–1) in the upper experimental area to 1.0 kg (3.4 kg P ha–1) in the lower experimental area and 0.5 kg (1.8 kg P ha–1) in the control area. The retained proportions of N and P in the plant biomass in the two experimental areas were approximately 70% of the added N (45 kg N y–1) and approximately 25% of the added P (15 kg P y–1) during the first year after N and P addition in 1999. Our study shows that vigorously colonising and growing vegetation is the main factor in the retention of N, a significant factor in the retention of P in a constructed wetland buffer, and thus an important contributor to the prevention of detrimental effects of N and P leaching on watercourses.  相似文献

To investigate the effects of extremely low frequency magnetic fields on ultraviolet radiation (UV) exposed budding yeast, haploid yeast (Saccharomyces cerevisiae) cells of the strain SEy2101a were exposed to 50 Hz sine wave magnetic field (MF) of 120 microT with simultaneous exposure to UV radiation. Most of the UV energy was in the UVB range (280-320 nm). The biologically weighted (CIE action spectrum) dose level for the UV radiation was 175 J/m2. We examined whether 50 Hz MF affected the ability of UV irradiated yeast cells to form colonies (Colony Forming Units, CFUs). In addition, the effect of coexposure on cell cycle kinetics was investigated. Although the significant effect of MF on the cell cycle phases of UV exposed yeast cells was seen only at one time point, the overall results showed that MF exposure may influence the cell cycle kinetics at the first cycle after UV irradiation. The effect of our particular MF exposure on the colony forming ability of the UV irradiated yeast cells was statistically significant 420 min after UV irradiation. Moreover, at 240, 360, and 420 min after UV irradiation, there were fewer CFUs in every experiment in (UV+MF) exposed populations than in only UV exposed yeast populations. These results could indicate that MF exposure in conjunction with UV may have some effects on yeast cell survival or growth.  相似文献

We used logistic and Poisson regression models to determine factors of forest and landscape structure that influence the presence and abundance of rodent species in the rain forest of Ranomafana National Park in southeastern Madagascar. Rodents were collected using live-traps along a gradient of human disturbance. All five endemic rodent species (Nesomys rufus, N. audeberti, Eliurus tanala, E. minor and E. webbi) and the introduced rat Rattus rattus were captured in both secondary and primary forests, but the introduced Mus musculus was only trapped in secondary forest. The abundance of R. rattus increased with the level of habitat disturbance, and it was most common in the heavily logged secondary forest. Furthermore, the probability of the presence of R. rattus increased with decreasing distance from forest edge and decreasing canopy cover, while the probability of presence increased with increasing herbaceous cover, altitude and overstory tree height. The species was never observed farther than 500 m away from human habitation or camp-site. N. rufus prefered selectively-logged forest at altitudes above 900 m a.s.l. Its probability of presence increased with increasing canopy cover, herbaceous cover and distance from forest edge, and with decreasing density of fallen logs, overstory tree height and distance from human habitation. N. audeberti prefered heavily-logged areas, while E. tanala was the only species occurring along the entire range of forest disturbance. We suggest that in the Ranomafana National Park the spread of R. rattus is associated with deforestation.  相似文献

Spatial similarity of urban bird communities: a multiscale approach   总被引:14,自引:0,他引:14  
Aim Human land use, especially urbanization, might homogenize the world's biota. The objective of this study is to find out if urbanization homogenizes wintering bird communities, and if habitat type affects the spatial variation of urban bird communities across spatial scales. Location We compared the quantitative similarity of winter bird communities between town centres, apartment block areas and single‐family house areas across regional and local scales in five towns in northern Finland. Methods The wintering birds were surveyed using a single‐visit study plot (30 ha) method in January and February 2001. The validity of single‐visit and single‐year data was confirmed by using data from several‐visit surveys and multi‐year data set. The level of urbanization was measured according to the number of inhabitants and general structure of the habitat. Results Temporal variability in species richness and total number of individuals was low, both between winters and within winter. Bird community similarity between different habitat types within a single town was about the same as that in similar habitats in different towns. At the regional scale, bird community similarity between town centres (30%) was lower than between areas of apartment blocks (54%) or between areas of single‐family houses (54%). We detected a threshold point between towns of 35,000 and 105,000 inhabitants and between town sizes of 5.0–8.5 km in diameter where human impact causes marked changes in bird community structure. At the local scale, bird community similarity level between apartment block areas (49%) and single‐family house areas (62%) were about the same. Passer domesticus, Parus major and Pica pica dominated the bird communities in residential areas. Different habitat factors affected bird species abundances on the local and regional scales. Conclusions Urbanization cannot be seen as a process that monotonically increases the similarity of bird communities. Our results indicate that the similarity between urban bird communities is dependent on the size of the town, location of the study site within the town and especially the local habitat structure. Because different habitat factors affected bird species abundances, it is difficult to extrapolate bird–habitat relationships derived from one scale to other scales. In wintertime, single‐family house areas are important biodiversity hotspots in cities. Therefore, it is especially important to understand the factors affecting the occurrence of birds in the single‐family house area in order to maintain or even increase diversity on winter birds in other urban habitats.  相似文献

Environmental stress has been suggested to increase host susceptibility to infections and reduce host ability to resist parasite growth and reproduction, thus benefiting parasites. This prediction stems from expected costs of immune defence; hosts in poor condition should have less resources to be allocated to immune function. However, the alternative hypothesis for response to environmental stress is that hosts in poor condition provide less resources for parasites and/or suffer higher mortality, leading to reduced parasite growth, reproduction and survival. We contrasted these alternative hypotheses in a trematode–snail ( Diplostomum spathaceum – Lymnaea stagnalis ) system by asking: (1) how host condition affects parasite reproduction (amount and quality of produced transmission stages) and (2) how host condition affects the survival of infected host individuals. We experimentally manipulated host condition by starving the snails, and found that parasites produced fewer and poorer quality transmission stages in stressed hosts. Furthermore, starvation increased snail mortality. These findings indicate that in well-established trematode infections, reduced ability of immune allocation has no effect on host exploitation by parasites. Instead, deteriorating resources for the snail host can directly limit the amount of resources available for the parasite. This, together with increased host mortality, may have negative effects on parasite populations in the wild.  相似文献

SIRT2 inhibitors with a N-(3-phenylpropenoyl)-glycine tryptamide backbone were studied. This backbone has been developed in our group, and it is derived from a compound originally found by virtual screening. In addition, compounds with a smaller 3-phenylpropenoic acid tryptamide backbone were also included in the study. Binding modes for the new compounds and the previously reported compounds were analyzed with molecular modelling methods. The approach, which included a combination of molecular dynamics, molecular docking and cluster analysis, showed that certain docking poses were favourable despite the conformational variation in the target protein. The N-(3-phenylpropenoyl)-glycine tryptamide backbone is also a good backbone for SIRT2 inhibitors, and the series of compounds includes several potent SIRT2 inhibitors.  相似文献

We found that the human intestinal isolate Bifidobacterium bifidum MIMBb75 strongly adhered to Caco-2 cells. Proteinase K and lithium chloride treatments showed that proteins play a key role in MIMBb75 adhesion to Caco-2 cells. By studying the cell wall-associated proteins, we identified a surface protein, which we labeled BopA. We purified the protein chromatographically and found that it functioned as an adhesion promoter on Caco-2 cells. In silico analysis of the gene coding for this protein and globomycin experiments showed that BopA is a cysteine-anchored lipoprotein expressed as a precursor polypeptide. A database search indicated that BopA appears to function biologically as an oligopeptide/tripeptide-solute-binding protein in the ABC transport system. We discovered a protein corresponding to BopA and its gene in eight other highly adherent B. bifidum strains. Finally, we found that B. bifidum MIMBb75 and BopA affected the production of interleukin-8 in Caco-2 epithelial cells. BopA is the first protein described to date to be directly involved in the adhesion of bifidobacteria to Caco-2 cells and to show immunomodulatory activity.  相似文献

Despite the high prevalence of nonalcoholic fatty liver disease (NAFLD), little is known of its pathogenesis based on study of human liver samples. By the use of Affymetrix GeneChips (17,601 genes), we investigated gene expression in the human liver of subjects with extreme steatosis due to NAFLD without histological signs of inflammation (liver fat 66.0 +/- 6.8%) and in subjects with low liver fat content (6.4 +/- 2.7%). The data were analyzed by using sequence-based reannotation of Affymetrix probes and a robust model-based normalization method. We identified genes involved in hepatic glucose and lipid metabolism, insulin signaling, inflammation, coagulation, and cell adhesion to be significantly associated with liver fat content. In addition, genes involved in ceramide signaling (MAP2K4) and metabolism (UGCG) were found to be positively associated with liver fat content. Genes involved in lipid metabolism (PLIN, ACADM), fatty acid transport (FABP4, CD36), amino acid catabolism (BCAT1), and inflammation (CCL2) were validated by real-time PCR and were found to be upregulated in subjects with high liver fat content. The data show that multiple changes in gene expression characterize simple steatosis.  相似文献
